    Six-Week Endurance Exercise Alters Gut Metagenome That Is not Reflected in Systemic Metabolism in Over-weight Women

    Recent studies suggest that exercise alters the gut microbiome. We determined whether six-weeks endurance exercise, without changing diet, affected the gut metagenome and systemic metabolites of overweight women. Previously sedentary overweight women (n = 19) underwent a six-weeks endurance exercise intervention, but two were excluded due to antibiotic therapy. The gut microbiota composition and functions were analyzed by 16S rRNA gene amplicon sequencing and metagenomics. Body composition was analyzed with DXA X-ray densitometer and serum metabolomics with NMR metabolomics. Total energy and energy-yielding nutrient intakes were analyzed from food records using Micro-Nutrica software. Serum clinical variables were determined with KONELAB instrument. Soluble Vascular Adhesion Protein 1 (VAP-1) was measured with ELISA and its' enzymatic activity as produced hydrogen peroxide. The exercise intervention was effective, as maximal power and maximum rate of oxygen consumption increased while android fat mass decreased. No changes in diet were observed. Metagenomic analysis revealed taxonomic shifts including an increase in Akkermansia and a decrease in Proteobacteria. These changes were independent of age, weight, fat % as well as energy and fiber intake. Training slightly increased Jaccard distance of genus level β-diversity. Training did not alter the enriched metagenomic pathways, which, according to Bray Curtis dissimilarity analysis, may have been due to that only half of the subjects' microbiomes responded considerably to exercise. Nevertheless, tranining decreased the abundance of several genes including those related to fructose and amino acid metabolism. These metagenomic changes, however, were not translated into major systemic metabolic changes as only two metabolites, phospholipids and cholesterol in large VLDL particles, decreased after exercise. Training also decreased the amine oxidase activity of pro-inflammatory VAP-1, whereas no changes in CRP were detected. All clinical blood variables were within normal range, yet exercise slightly increased glucose and decreased LDL and HDL. In conclusion, exercise training modified the gut microbiome without greatly affecting systemic metabolites or body composition. Based on our data and existing literature, we propose that especially Akkermansia and Proteobacteria are exercise-responsive taxa. Our results warrant the need for further studies in larger cohorts to determine whether exercise types other than endurance exercise also modify the gut metagenome

    Damage analysis of masonry structures subjected to rockfalls

    Masonry structures present substantial vulnerability to rockfalls. The methodologies for the damage quantification of masonry structures subjected to rockfalls are scarce. An analytical procedure for the damage assessment of masonry structures is presented. The procedure comprises three stages: (1) determination of the rockfall impact actions which are applied to a masonry structure, in terms of external forces, using the particle finite element method (PFEM), (2) evaluation of the mechanical properties, modelling of the masonry structure, and calculation of the internal stresses, using the finite element method (FEM), (3) assessment of the damage due to the rockfall actions, applying a failure criterion adapted to masonries, and calculation of the damage in terms of the percentage of the damaged wall surface. Three real rockfall events and their impact on buildings are analysed. A sensitivity analysis of the proposed procedure is then used to identify the variables that mostly affect the extent of the wall damage, which are the masonry width, the tensile strength, the block diameter and lastly, velocity.Peer Reviewe

    O Programa de Controle da Doença de Chagas (PCDCh) no Estado de São Paulo encontra-se na fase de vigilância entomológica, submetendo a exames sorológicos os moradores de unidades domiciliares (UDs), em que triatomíneos vetores tenham sido encontrados infectados por Trypanosoma cruzi. No decênio 1990-1999 foram trabalhadas localidades que, segundo graus de infestações no intra ou no peridomicílio, desencadearam trabalhos de rotina de busca desses vetores e de atendimento a notificações por parte de moradores. Em 1.415 UDs examinadas, 5.587 amostras de sangue foram obtidas, 87 (1,56%) das quais reagentes. Dessas, sete correspondiam a pessoas com menos de 29 anos de idade. As espécies mais freqüentemente associadas, em suas formas adultas, às UDs foram: Panstrongylus megistus, Triatoma sordida e T. tibiamaculata. Não foi observada associação entre UDs com sororreagentes e presença de triatomíneos infectados por Tr. cruzi (odds ratio = 1,498; 0,875 < OR < 2,564, 95% de confiança). Propõe-se utilizar inquéritos sorológicos amostrais no PCDCh, para investigar a situação prevalente em áreas identificáveis como de risco de transmissão, complementando-os com estudos isolados de Tr. cruzi aí obtidos.<br>The Chagas Disease Control Program in São Paulo State, Brazil, now in the entomological surveillance phase, includes a serological examination of individuals residing in domiciliary units infested with vector triatomines infected with Trypanosoma cruzi. From 1990 to 1999, this action included area in which triatomine searches were conducted either as a routine procedure, according to their levels of intra- or peridomiciliary infestation, or at the request of local residents. Among residents of the 1,415 UDs inspected, we collected 5,587 blood samples for serological examination, 87 of which (1.56%) tested positive, seven of which from individuals under 29 years of age. The species most frequently captured were Panstrongylus megistus, Triatoma sordida, and Triatoma tibiamaculata in the adult stage. No association was found between presence of seropositive residents and triatomines infected with Tr. cruzi (OR = 1.498; 0.875 < OR < 2.564, 95% C.I.). Our purpose was to use serological testing to investigate the situation of areas identifiable as being at risk of Chagas disease transmission and to compare the results with extant data about Tr. cruzi infection both in humans and vector triatomines
