    The Kast Ground Based UV Spectral Survey of 79 QSOs at Redshift 2 for Lyman Alpha Forest and Metal Absorption

    We present a moderate resolution (~1.15 Angstroms/pixel) survey of 79 quasars obtained using the Kast spectrograph on the Shane 3m telescope at Lick observatory. The spectra span the wavelength range of 3175-5880 Angstroms, and have typical signal to noise of 6-20 in the regions of the spectra showing Lyman alpha forest absorption. The quasars have a mean emission redshift of z=2.17, and nearly all cover the entire Lyman alpha forest between Lyman alpha and Lyman beta. Although the quasars were selected to avoid BAL, two quasars in the survey are BAL, one of which is a new discovery. We list the HI and metal ions observed in a total of 140 absorption systems. We also identify 526 emission lines, and list their observed wavelengths, along with new redshifts of the quasars. We determine the rest wavelengths of 3 emission lines or line blends in the forest to be 1070.95 +/- 1.00, 1123.13 +/- 0.51, and 1175.88 +/- 0.30 Angstroms.Comment: Submitted to the Astronomical Journa

    Twisted Conjugacy Classes in Abelian Extensions of Certain Linear Groups

    Given an automorphism ϕ:Γ→Γ\phi:\Gamma\to \Gamma, one has an action of Γ\Gamma on itself by ϕ\phi-twisted conjugacy, namely, g.x=gxϕ(g−1)g.x=gx\phi(g^{-1}). The orbits of this action are called ϕ\phi-twisted conjugacy classes. One says that Γ\Gamma has the R∞R_\infty-property if there are infinitely many ϕ\phi-twisted conjugacy classes for every automorphism ϕ\phi of Γ\Gamma. In this paper we show that SL(n,Z)(n,\mathbb{Z}) and its congruence subgroups have the R∞R_\infty-property. Further we show that any (countable) abelian extension of Γ\Gamma has the R∞R_\infty-property where Γ\Gamma is a torsion free non-elementary hyperbolic group, or SL(n,Z)(n,\mathbb{Z}), Sp(2n,Z)(2n,\mathbb{Z}) or a principal congruence subgroup of SL(n,Z)(n,\mathbb{Z}) or the fundamental group of a complete Riemannian manifold of constant negative curvature

    peer-reviewedStaphylococcus aureus and other coagulase-positive Staphylococcus spp. bind the Fc region of IgG antibodies through expression of protein A (SpA). These species have consequently been a source of false-positive signals in antibody-based assays designed to detect other target bacteria. Here, flow cytometry was used to study the influence of a number of factors on the SpA-mediated binding of single cells to an anti-human IgG antibody, including strain, heat killing, overnight storage, growth phase, cell physiology, surface adhesion, and growth in model food systems. Through the costaining of antibody-stained cells with the permeability dye pr opidium iodide and calcein violet AM, the cell physiological status was related to SpA-mediated antibody binding. Generally, permeabilized cells lacking esterase activity did not strongly bind antibody. The binding of a number of commercially available polyclonal IgG antibodies to non-Staphylococcus spp. was also characterized. Not all SpA-expressing species showed strong binding of mouse IgG, and one species not known to express SpA showed strong binding. Most SpA-expressing strains bound rabbit IgG antibodies to some extent, whereas only one strain bound goat IgG. To reduce or eliminate SpA-mediated IgG binding, the following products were evaluated as blocking reagents and applied prior to staining with primary or secondary antibody: normal rabbit serum, mouse IgG isotype control, goat IgG, and a commercial FcR blocking reagent. Only the FcR blocking reagent consistently reduced SpA-mediated binding of Staphylococcus spp. to antibodies against other species and could be recommended as a blocking reagent in immunoassays designed to detect non-Staphylococcus species

    Reliable enumeration of malaria parasites in thick blood films using digital image analysis

    <p>Abstract</p> <p>Background</p> <p>Quantitation of malaria parasite density is an important component of laboratory diagnosis of malaria. Microscopy of Giemsa-stained thick blood films is the conventional method for parasite enumeration. Accurate and reproducible parasite counts are difficult to achieve, because of inherent technical limitations and human inconsistency. Inaccurate parasite density estimation may have adverse clinical and therapeutic implications for patients, and for endpoints of clinical trials of anti-malarial vaccines or drugs. Digital image analysis provides an opportunity to improve performance of parasite density quantitation.</p> <p>Methods</p> <p>Accurate manual parasite counts were done on 497 images of a range of thick blood films with varying densities of malaria parasites, to establish a uniformly reliable standard against which to assess the digital technique. By utilizing descriptive statistical parameters of parasite size frequency distributions, particle counting algorithms of the digital image analysis programme were semi-automatically adapted to variations in parasite size, shape and staining characteristics, to produce optimum signal/noise ratios.</p> <p>Results</p> <p>A reliable counting process was developed that requires no operator decisions that might bias the outcome. Digital counts were highly correlated with manual counts for medium to high parasite densities, and slightly less well correlated with conventional counts. At low densities (fewer than 6 parasites per analysed image) signal/noise ratios were compromised and correlation between digital and manual counts was poor. Conventional counts were consistently lower than both digital and manual counts.</p> <p>Conclusion</p> <p>Using open-access software and avoiding custom programming or any special operator intervention, accurate digital counts were obtained, particularly at high parasite densities that are difficult to count conventionally. The technique is potentially useful for laboratories that routinely perform malaria parasite enumeration. The requirements of a digital microscope camera, personal computer and good quality staining of slides are potentially reasonably easy to meet.</p
