    Hyperbaric oxygen preconditioning attenuates hyperglycemia enhanced hemorrhagic transformation after transient MCAO in rats

    <p>Abstract</p> <p>Background</p> <p>Hemorrhagic transformation (HT) can be a devastating complication of ischemic stroke. Hyperbaric oxygen preconditioning (HBO-PC) has been shown to improve blood-brain barrier (BBB) permeability in stroke models. The purpose of this study is to examine whether HBO-PC attenuates HT after focal cerebral ischemia, and to investigate whether the mechanism of HBO-PC against HT includes up-regulation of antioxidants in hyperglycemic rats.</p> <p>Methods</p> <p>Male Sprague-Dawley rats (280-320 g) were divided into the following groups: sham, middle cerebral artery occlusion (MCAO) for 2 h, and MCAO treated with HBO-PC. HBO-PC was conducted giving 100% oxygen at 2.5 atm absolute (ATA), for 1 h at every 24 h interval for 5 days. At 24 h after the last session of HBO-PC, rats received an injection of 50% glucose (6 ml/kg intraperitoneally) and were subjected to MCAO 15 min later. At 24 h after MCAO, neurological behavior tests, infarct volume, blood-brain barrier permeability, and hemoglobin content were measured to evaluate the effect of HBO-PC. Western blot analysis of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) was evaluated at multiple time-points before and after MCAO.</p> <p>Results</p> <p>HBO-PC improved neurological behavior test, and reduced infarction volume, HT and Evans blue extravasation in the ipsilateral hemisphere at 24 h after MCAO. Western blot analysis failed to demonstrate up-regulation of Nrf2 in HBO-PC group before and after MCAO. Paradoxically, HBO-PC decreased HO-1 expression at 24 h after MCAO, as compared with htMCAO group.</p> <p>Conclusions</p> <p>HBO-PC improved neurological deficits, infarction volume, BBB disruption, and HT after focal cerebral ischemia. However, its mechanism against focal cerebral ischemia and HT may not include activation of Nrf2 and subsequent HO-1 expression.</p

    Proof of principle for a high sensitivity search for the electric dipole moment of the electron using the metastable a(1)[^3\Sigma^+] state of PbO

    The metastable a(1)[^3\Sigma^+] state of PbO has been suggested as a suitable system in which to search for the electric dipole moment (EDM) of the electron. We report here the development of experimental techniques allowing high-sensitivity measurements of Zeeman and Stark effects in this system, similar to those required for an EDM search. We observe Zeeman quantum beats in fluorescence from a vapor cell, with shot-noise limited extraction of the quantum beat frequencies, high counting rates, and long coherence times. We argue that improvement in sensitvity to the electron EDM by at least two orders of magnitude appears possible using these techniques.Comment: 5 pages, 3 figure

    Correlation between Arctic river discharge and sea ice formation in Laptev Sea using sea surface salinity from SMOS satellite

    European Geosciences Union (EGU) General Assembly 2020, 4-8 May 2020During the last 3 decades, the Arctic rivers have increased their discharge around 10%, mainly due to the increase of the global atmospheric temperature. The increase of the river discharge carries higher loads of dissolved organic matter (DOM) and suspended matter (SM) entering to the Arctic Ocean. This results in increased absorption of solar energy in the mixed layer, which can potentially contribute to the general sea ice retreat. Observation based studies (e.g. Bauch et al., 2013) showed correlation between river water discharge and local sea ice melting on the Laptev sea shelf due to the change on the ocean heat. Previous studies are based with a limited number of observations, both in space and in time. Thanks to the ESA SMOS (Soil Moisture and Ocean Salinity) and NASA SMAP (Soil Moisture Active Passive) missions we have daily the sea surface salinity (SSS) maps from the Arctic, which permit to observe the salinity variations due to the river discharges. The Arctic sea surface salinity products obtained from SMOS measurements have been improved considerable by the Barcelona Expert Center (BEC) team thanks to the project Arctic+Salinity, funded by ESA. The new version of the product (v3) covers the years from 2011 up to 2018, have a spatial resolution of 25km and are daily maps with 9 day averages. The Arctic+ SSS maps provide a better description of the salinity gradients and a better effective spatial resolution than the previous versions of the Arctic product, so the salinity fronts are better resolved. The quality assessment of the Arctic+SSS product is challenging because, in this region, there are scarce number of in-situ measurements. The high effective spatial resolution of the Arctic+ SSS maps will permit to study for the first time scientific physical processes that occurs in the Arctic. We will explore if a correlation between the Lena and Ob rivers discharge with the sea ice melting and freeze up is observed with satellite data, as already stated with in-situ measurements by Bauch et al. 2013.     Assessing the predictive performance of population pharmacokinetic models for intravenous polymyxin B in critically ill patients

    Polymyxin B (PMB) has reemerged as a last-line therapy for infections caused by multidrug-resistant gram-negative pathogens, but dosing is challenging because of its narrow therapeutic window and pharmacokinetic (PK) variability. Population PK (POPPK) models based on suitably powered clinical studies with appropriate sampling strategies that take variability into consideration can inform PMB dosing to maximize efficacy and minimize toxicity and resistance. Here we reviewed published PMB POPPK models and evaluated them using an external validation data set (EVD) of patients who are critically ill and enrolled in an ongoing clinical study to assess their utility. Seven published POPPK models were employed using the reported model equations, parameter values, covariate relationships, interpatient variability, parameter covariance, and unexplained residual variability in NONMEM (Version 7.4.3). The predictive ability of the models was assessed using prediction-based and simulation-based diagnostics. Patient characteristics and treatment information were comparable across studies and with the EVD (n = 40), but the sampling strategy was a main source of PK variability across studies. All models visually and statistically underpredicted EVD plasma concentrations, but the two-compartment models more accurately described the external data set. As current POPPK models were inadequately predictive of the EVD, creation of a new POPPK model based on an appropriately powered clinical study with an informed PK sampling strategy would be expected to improve characterization of PMB PK and identify covariates to explain interpatient variability. Such a model would support model-informed precision dosing frameworks, which are urgently needed to improve PMB treatment efficacy, limit resistance, and reduce toxicity in patients who are critically ill

    Risk-Return Relationship in a Complex Adaptive System

    For survival and development, autonomous agents in complex adaptive systems involving the human society must compete against or collaborate with others for sharing limited resources or wealth, by using different methods. One method is to invest, in order to obtain payoffs with risk. It is a common belief that investments with a positive risk-return relationship (namely, high risk high return and vice versa) are dominant over those with a negative risk-return relationship (i.e., high risk low return and vice versa) in the human society; the belief has a notable impact on daily investing activities of investors. Here we investigate the risk-return relationship in a model complex adaptive system, in order to study the effect of both market efficiency and closeness that exist in the human society and play an important role in helping to establish traditional finance/economics theories. We conduct a series of computer-aided human experiments, and also perform agent-based simulations and theoretical analysis to confirm the experimental observations and reveal the underlying mechanism. We report that investments with a negative risk-return relationship have dominance over those with a positive risk-return relationship instead in such a complex adaptive systems. We formulate the dynamical process for the system's evolution, which helps to discover the different role of identical and heterogeneous preferences. This work might be valuable not only to complexity science, but also to finance and economics, to management and social science, and to physics

    A hydrogel based localized release of colistin for antimicrobial treatment of burn wound infection

    There is an urgent unmet medical need for new treatments for wound and burn infections caused by multidrug-resistant (MDR) Gram-negative ‘superbugs’, especially the problematic Pseudomonas aeruginosa. In this work we report the incorporation of colistin, a potent lipopeptide into a self-healable hydrogel (via dynamic imine bond formation) following the chemical reaction between the amine groups present in glycol chitosan and an aldehyde modified poly (ethylene glycol) (PEG). The storage module (G’) of the colistin-loaded hydrogel ranged from 1.3 kPa to 5.3 kPa by varying the amount of the cross-linker and colistin loading providing different options for topical wound healing. The majority of the colistin is released from the hydrogel within 24 h and remains active as demonstrated by both antibacterial in vitro disk diffusion and time-kill assays. Moreover and pleasingly, the colistin-loaded hydrogel performed almost equally well as native colistin against both the colistin-sensitive and also colistin-resistant P. aeruginosa strain in the in vivo animal 'burn' infection model despite exhibiting a slower killing profile in vitro. Based on this antibiotic performance along with the biodegradability of the product, we believe the colistin-loaded hydrogel to be a potential localized wound-healing formulation to treat burn wounds against microbial infection

    Cue-Elicited Craving in Heroin Addicts at Different Abstinent Time: An fMRI Pilot Study

    Objective: We evaluated the effect of short-term and long-term heroin abstinence on brain responses to heroin-related cues using functional magnetic resonance imaging (fMRI). Methods: Eighteen male heroin addicts following short-term abstinence and 19 male heroin addicts following long-term abstinence underwent fMRI scanning while viewing heroin-related and neutral images. Cue-elicited craving and withdrawal symptoms in the subjects were measured. Results: Following short-term abstinence, greater activation was found in response to heroin cues compared to neutral cues in bilateral temporal, occipital, posterior cingulate, anterior cingulate, thalamus, cerebellum, and left hippocampus. In contrast, activations in bilateral temporal and occipital and deactivations in bilateral frontal, bilateral parietal, left posterior cingulate, insula, thalamus, dorsal striatum, and bilateral cerebellum were observed following long-term abstinence. Direct comparisons between conditions showed greater brain reactivity in response to smoking cues following short-term abstinence. In addition, short-term abstinence had more serious withdrawal symptoms than the long-term. Conclusion: The present findings indicate that compared to short-term, long-term abstinence manifests less serious withdrawal symptoms and significantly decreases neural responses to heroin-related cues in brain regions subserving visual sensory processing, attention, memory, and action planning. These findings suggest that long-term abstinence can decrease the salience of conditioned cues, thereby reducing the risk of relapses. The study's limitations are noted

    Characterization of occupational exposures to cleaning products used for common cleaning tasks-a pilot study of hospital cleaners

    Background: In recent years, cleaning has been identified as an occupational risk because of an increased incidence of reported respiratory effects, such as asthma and asthma-like symptoms among cleaning workers. Due to the lack of systematic occupational hygiene analyses and workplace exposure data, it is not clear which cleaning-related exposures induce or aggravate asthma and other respiratory effects. Currently, there is a need for systematic evaluation of cleaning products ingredients and their exposures in the workplace. The objectives of this work were to: a) identify cleaning products' ingredients of concern with respect to respiratory and skin irritation and sensitization; and b) assess the potential for inhalation and dermal exposures to these ingredients during common cleaning tasks. Methods: We prioritized ingredients of concern in cleaning products commonly used in several hospitals in Massachusetts. Methods included workplace interviews, reviews of product Materials Safety Data Sheets and the scientific literature on adverse health effects to humans, reviews of physico-chemical properties of cleaning ingredients, and occupational hygiene observational analyses. Furthermore, the potential for exposure in the workplace was assessed by conducting qualitative assessment of airborne exposures and semi-quantitative assessment of dermal exposures. Results: Cleaning products used for common cleaning tasks were mixtures of many chemicals, including respiratory and dermal irritants and sensitizers. Examples of ingredients of concern include quaternary ammonium compounds, 2-butoxyethanol, and ethanolamines. Cleaning workers are at risk of acute and chronic inhalation exposures to volatile organic compounds (VOC) vapors and aerosols generated from product spraying, and dermal exposures mostly through hands. Conclusion: Cleaning products are mixtures of many chemical ingredients that may impact workers' health through air and dermal exposures. Because cleaning exposures are a function of product formulations and product application procedures, a combination of product evaluation with workplace exposure assessment is critical in developing strategies for protecting workers from cleaning hazards. Our task based assessment methods allowed classification of tasks in different exposure categories, a strategy that can be employed by epidemiological investigations related to cleaning.     Improved BEC SMOS Arctic Sea Surface Salinity product v3.1

The Argo program is part of the Global Ocean Observing SystemMeasuring salinity from space is challenging since the sensitivity of the brightness temperature (TB) to sea surface salinity (SSS) is low (about 0.5 K psu−1), while the SSS range in the open ocean is narrow (about 5 psu, if river discharge areas are not considered). This translates into a high accuracy requirement of the radiometer (about 2–3 K). Moreover, the sensitivity of the TB to SSS at cold waters is even lower (0.3 K psu−1), making the retrieval of the SSS in the cold waters even more challenging. Due to this limitation, the ESA launched a specific initiative in 2019, the Arctic+Salinity project (AO/1-9158/18/I-BG), to produce an enhanced Arctic SSS product with better quality and resolution than the available products. This paper presents the methodologies used to produce the new enhanced Arctic SMOS SSS product (Martínez et al., 2019) . The product consists of 9 d averaged maps in an EASE 2.0 grid of 25 km. The product is freely distributed from the Barcelona Expert Center (BEC, http://bec.icm.csic.es/, last access: 25 January 2022) with the DOI number https://doi.org/10.20350/digitalCSIC/12620 (Martínez et al., 2019).
