Quantifying the Differences in Binding Affinity of Reduced Glutathione for Glutathione S-Transferase at pH 6.5 and 8.5 Using Isothermal Titration Calorimetry
The binding affinity between an enzyme and its substrate is often dependent on the pH of the local environment. Knowing the pH at which reduced glutathione (GSH) binds with the highest affinity to the enzyme glutathione S-transferase (GST) is useful for determining the optimal pH for purification of GST-fusion proteins during GST-affinity chromatography. In this study, GST of the species Schistosoma japonicum was purified, quantified, and utilized to study its binding interaction with GSH at pH 6.5 and 8.5 via isothermal titration calorimetry (ITC). After protein expression, extraction, and purification, the GST concentration was quantified using QubitTM fluorometry. Thermodynamic properties and a dissociation constant (KD) for each experiment were obtained utilizing the MicroCal PEAQ-ITC Analysis Software for the binding of GSH to GST at pH 6.5 and 8.5. A Good Idea is Not Enough: Understanding the Challenges of Entrepreneurship Communication
This paper addresses a less-investigated issue of innovations: entrepreneurship communication. Business and marketing studies demonstrate that new product development processes do not succeed on good technical invention alone. To succeed, the invention must be appropriately communicated to a market and iterated through dialogue with potential stakeholders.
We explore this issue by examining communication-related challenges, abilities and barriers from the perspectives of innovators trying to enter an unfamiliar, foreign market. Specifically, we summarize results of a set of studies conducted in the Gyeonggi Innovation Program (GIP), an entrepreneurship program formed by a partnership between the University of Texas at Austin and Gyeonggi-Do Province in South Korea. Through the GIP, Korean entrepreneurs attempt to expand domestically successful product ideas to the American market. The study results demonstrate that these innovators must deal with a broad range of challenges, particularly (1) developing deeper understanding of market needs, values, and cultural expectations, and (2) producing pitches with the structure, claims and evidence, and engagement strategies expected by American stakeholders. These studies confirm that a deeper understanding of successful new product development (NPD) projects requires not only a culturally authentic NPD process model, but also communication-oriented research.
The GIP approach offers insights into good programmatic concept and effective methods for training engineers to become entrepreneurs. Yet we also identify potential improvements for such programs. Finally, we draw implications for studying entrepreneurship communication.IC2 Institut
Intermediate phase of the one dimensional half-filled Hubbard-Holstein model
We present a detailed numerical study of the Hubbard-Holstein model in one
dimension at half filling, including full finite-frequency quantum phonons. At
half filling, the effects of the electron-phonon and electron-electron
interactions compete, with the Holstein phonon coupling acting as an effective
negative Hubbard onsite interaction U that promotes on-site electron pairs and
a Peierls charge-density wave state. Most previous work on this model has
assumed that only Peierls or U>0 Mott insulator phases are possible at half
filling. However, there has been speculation that a third metallic phase exists
between the Peierls and Mott phases. We present results confirming the
intermediate metallic phase, and show that the Luttinger liquid correlation
exponent K_rho>1 in this region, indicating dominant superconducting pair
correlations. We explore the full phase diagram as a function of onsite Hubbard

Analytical study of the optimum geometric configuration of a space shuttle materials laboratory
A steady state, collisionless flow analysis was made of the density distribution within a hemisphere-disc system due to independent, uniformly distributed internal gas sources. The model was used to estimate the density within a molecular shield, deployed from the shuttle orbiter, which contained internal experiments having a prescribed gas source. Contour plots of the density distribution within the system were presented for disc-to-hemisphere radius ratios of .1, .3, .5, .7, and for disc-to-hemisphere surface emission flux density ratios of .01, 1, 100. The hemisphere-disc system was compared to the empty hemisphere, and it was found that if the disc emission flux density was the same as the hemisphere and the disc radius was not greater than 1/3 of the hemisphere radius, the increase in density at the center of the hemisphere-disc system was less than 50%
A catch-free stock assessment model with application to goliath grouper (Epinephelus itajara) off southern Florida
Many modern stock assessment methods provide the machinery for determining the status of a stock in relation to certain reference points and for estimating how quickly a stock can be rebuilt. However, these methods typically require catch data, which are not always available. We introduce a model-based framework for estimating reference points, stock status, and recovery times in situations where catch data and other measures of absolute abundance are unavailable. The specif ic estimator developed is essentially an age-structured production model recast in terms relative to pre-exploitation levels. A Bayesian estimation scheme is adopted to allow the incorporation of pertinent auxiliary information such as might be obtained from meta-analyses of similar stocks or anecdotal observations. The approach is applied to the population of goliath grouper (Epinephelus itajara) off southern Florida, for which there are three indices of relative abundance but no reliable catch data. The results confirm anecdotal accounts of a marked decline in abundance during the 1980s followed by a substantial increase after the harvest of goliath grouper was banned in 1990. The ban appears to have reduced fishing pressure to between 10% and 50% of the levels observed during the 1980s. Nevertheless, the predicted fishing mortality rate under the ban appears to remain substantial, perhaps owing to illegal harvest and depth-related release mortality. As a result, the base model predicts that there is less than a 40% chance that the spawning biomass will recover to a level that would produce a 50% spawning potential ratio
