    The finger flexors occlusion threshold in sport-climbers: an exploratory study on its indirect approximation

    Blood flow partially determines specific climbing endurance (SCE) as it mediates oxygen bio-availability in the finger flexors. Blood flow is related to occlusion threshold (OT), which is defined as the contraction intensity at which intramuscular pressure exceeds perfusion blood pressure resulting in the cessation of local blood flow. The OT is represented as an inflection point on a force-time graph when isometric force is registered and applied through maximal and continuous tests. Endurance time (ET) to exhaustion is influenced by the relative isometric applied force and is different for each climber. The aim of this study was to explore whether an approximation of the finger flexors OT in sport climbers through records of ET to exhaustion at different isometric relative intensities was possible. We measured maximum finger hang ETs at 6 intensities ranging from 85% to 35% maximal force in 34 sport climbers of advanced and elite level. The values obtained were analysed by two different methods in an attempt to determine a change in the shape of the curve in the intensity-ET relationship graphs that approximated the OT for each climber. The results suggest that the finger flexors OT could be different among climbers, regardless of their strength and ability level. The presented methods do not accurately reflect the OT, but could indicate the intensity at which blood flow is restored in the active muscles. This is the first study to indirectly approximate the finger flexors OT in sport-climbers, a parameter that could be essential to assess SCE

    A new performance threshold in sport climbing: A change in how climbing trainers work?

    Objectives: Previous research has shown that the finger flexor's occlusion threshold (OT) could be different among sport climbers when expressed as a percentage of their maximum finger force (OT%), and that there is no association between the OT% and the climberś ability level. The aim of the present study was to evaluate the possible association between the relative finger force applied at the OT% (rff-OT%) and climbing ability level. Equipment and methods: WWe approximated the finger flexor's OT of 34 sport climbers by finger hang endurance test analyses at different intensities between 35% and 85% of their individual maximum finger force on a previously individually adapted edge depth, and we valued their rff-OT% as the relative force they could perform at that intensity. Results: We found a high correlation between the rff-OT% and climbing ability in elite climbers. These findings suggest that having an OT at the highest possible percentage is critical, in addition to having a high relative finger force, as this would enable climbers to express relative force at a wider range of intensities with favorable metabolic conditions

    This informal session led by the Patient Experience Research Centre (PERC), in collaboration with Positively UK, invited people living with, affected by, or working in HIV to share their experience, views, questions and concerns on accessing HIV care during COVID-19. The aim of the call was to gather feedback on specific areas to help guide a proposed qualitative (interview-based study) looking to explore experiences, specifically on: 1. Challenges and concerns in managing HIV care during COVID-19 2. Challenges in the provision of HIV care during COVID-19 3. Opportunities presented for HIV care during COVID-19 We also wished to inspire new ways to rapidly engage and involve communities remotely during a public health emergency, through strengthening partnerships with existing groups (in this case, Positively UK)

    We extend the algebraic-eikonal approach to medium energy proton scattering from odd-mass nuclei by combining the eikonal approximation for the scattering with a description of odd-mass nuclei in terms of the interacting boson-fermion model.     Enabling effective operational decision making on a Combined Heat and Power System using the 5C architecture

    The use of Cyber Physical Systems (CPS) to optimise industrial energy systems is an approach which has the potential to positively impact on manufacturing sector energy efficiency. The need to obtain data to facilitate the implementation of a CPS in an industrial energy system is however a complex task which is often implemented in a non-standardised way. The use of the 5C CPS architecture has the potential to standardise this approach. This paper describes a case study where data from a Combined Heat and Power (CHP) system located in a large manufacturing company was fused with grid electricity and gas models as well as a maintenance cost model using the 5C architecture with a view to making effective decisions on its cost efficient operation. A control change implemented based on the cognitive analysis enabled via the 5C architecture implementation has resulted in energy cost savings of over €7400 over a four-month period, with energy cost savings of over €150,000 projected once the 5C architecture is extended into the production environment

    Doxazosin induces apoptosis in PTEN–positive androgen-independent PC cells via inhibition of Akt activation

    Doxazosin, a selective α1-adrenoreceptor antagonist widely used in the management of benign prostatic hyperplasia (BPH), has been shown to induce apoptosis in androgen-independent (AI) and PTEN (phosphatases and tensin homolog)-negative prostate cancer (PC) cells. The objective of this study was to assess the effects of doxazosin on the growth of PC cells with a functional PTEN/PI3K/Akt pathway in relation to cell androgen sensitivity and Bcl-2 expression. Material and methods. The DU145 cell line and two derivatives: 1) DKC9 [DU145 (Bcl2 +/+)] and 2) DAR19 [DU145 (AR+/+)] were used. The effect of doxazosin on i) cell proliferation and ii) Akt phosphorylation was measured using the MTT assay and Western blotting, respectively. Results. Doxazosin caused significant concentration, but not time-dependent, decrease in cell viability. The threshold of sensitivity to the compound differed between the cell lines and was the lowest in the parental cell line. In DAR 19 cells, AR-mediated signaling further increased cell resistance to doxazosin. In all cell lines, doxazosin-induced apoptosis was rescued by EGF stimulation and, in DAR19 cells, by AR activation. Doxazosin reduced basal and EGF-/DHT-induced Akt phosphorylation. Conclusions. This study demonstrates that doxazosin induces apoptosis in PTEN-positive AI PC cells, at least partially, via Akt deactivation and that over-expression of Bcl-2 or AR increases cell resistance to the drug. These results imply that translational potential of doxazosin depends on phenotypic characteristics of PC cells and provide evidence for limitations to its application in hormone refractory tumors
