    In vitro effects of metals and pesticides on dehydrogenase activity in microbial community of cowpea (Vigna unguiculata) rhizoplane

    Effects of heavy metals and pesticides on cowpea (Vigna unquiculata) rhizoplane microbial community  were assessed in vitro via dehydrogenase activity. The microbial community was exposed to various concentrations of heavy metals and pesticides in a nutrient broth-glucose-2,3,5-triphenyl chloride (TTC) medium. At 0.2 mM, iron and cadmium stimulated the dehydrogenase activity of the microbialcommunity. For all the metal ions, there was progressive inhibition with each successive increase in the concentration of metal ion, reaching near 100% at 0.6, 0.8, 1.2, 0.12 and 12 mM for cobalt, cadmium,iron, mercury and nickel, respectively. Between 0.2 and 0.4 mM, zinc sharply inhibited dehydrogenase activity and at concentration above 0.4 mm, inhibition of dehydrogenase activity became lesspronounced. The order of toxicity is Hg2+ > Co2+ > Cd2+ > Zn2+ > Fe2+ > Ni2+. The herbicides Cotrazine (Atrazine 80W) and Northrin®10EC stimulated dehydrogenase activity of the microbial community at 0.2% and inhibited it at higher concentrations. The median inhibitory concentrations (IC50s) of Cotrazine(Atrazine 80W) and Northrin®10EC were 0.552 ± 0.028 and 0.593 ± 0.051%, respectively. The dehydrogenase activity varied significantly (p < 0.05) with the type and concentrations of metals orpesticides. The result indicates that the heavy metals and pesticides are potentially toxic to V. unquiculata root surface microorganisms. In soil, this toxicity may affect nitrogen fixation processes and by extrapolation affect crop yield

    The Human Pancreas as a Source of Protolerogenic Extracellular Matrix Scaffold for a New-generation Bioartificial Endocrine Pancreas

    OBJECTIVES: Our study aims at producing acellular extracellular matrix scaffolds from the human pancreas (hpaECMs) as a first critical step toward the production of a new-generation, fully human-derived bioartificial endocrine pancreas. In this bioartificial endocrine pancreas, the hardware will be represented by hpaECMs, whereas the software will consist in the cellular compartment generated from patient's own cells. BACKGROUND: Extracellular matrix (ECM)-based scaffolds obtained through the decellularization of native organs have become the favored platform in the field of complex organ bioengineering. However, the paradigm is now switching from the porcine to the human model. METHODS: To achieve our goal, human pancreata were decellularized with Triton-based solution and thoroughly characterized. Primary endpoints were complete cell and DNA clearance, preservation of ECM components, growth factors and stiffness, ability to induce angiogenesis, conservation of the framework of the innate vasculature, and immunogenicity. Secondary endpoint was hpaECMs’ ability to sustain growth and function of human islet and human primary pancreatic endothelial cells. RESULTS: Results show that hpaECMs can be successfully and consistently produced from human pancreata and maintain their innate molecular and spatial framework and stiffness, and vital growth factors. Importantly, hpaECMs inhibit human naïve CD4+ T-cell expansion in response to polyclonal stimuli by inducing their apoptosis and promoting their conversion into regulatory T cells. hpaECMs are cytocompatible and supportive of representative pancreatic cell types. DISCUSSION: We, therefore, conclude that hpaECMs has the potential to become an ideal platform for investigations aiming at the manufacturing of a regenerative medicine-inspired bioartificial endocrine pancreas

    Potent Antioxidant and Genoprotective Effects of Boeravinone G, a Rotenoid Isolated from Boerhaavia diffusa

    Background and Aims: Free radicals are implicated in the aetiology of some gastrointestinal disorders such as gastric ulcer, colorectal cancer and inflammatory bowel disease. In the present study we investigated the antioxidant and genoprotective activity of some rotenoids (i.e. boeravinones) isolated from the roots of Boerhaavia diffusa, a plant used in the Ayurvedic medicine for the treatment of diseases affecting the gastrointestinal tract. Methods/Principal Findings: Antioxidant activity has been evaluated using both chemical (Electron Spin Resonance spectroscopy, ESR) and Caco-2 cells-based (TBARS and ROS) assays. DNA damage was evaluated by Comet assay, while pERK 1/2 and phospho-NF-kB p65 levels were estimated by western blot. Boeravinones G, D and H significantly reduced the signal intensity of ESR induced by hydroxyl radicals, suggesting a scavenging activity. Among rotenoids tested, boeravinone G exerted the most potent effect. Boeravinone G inhibited both TBARS and ROS formation induced by Fenton's reagent, increased SOD activity and reduced H 2O 2-induced DNA damage. Finally, boeravinone G reduced the levels of pERK 1 and phospho-NF-kB p65 (but not of pERK 2) increased by Fenton's reagent. Conclusions: It is concluded that boeravinone G exhibits an extraordinary potent antioxidant activity (significant effect in the nanomolar range). The MAP kinase and NF-kB pathways seem to be involved in the antioxidant effect of boeravinone G. Boeravinone G might be considered as lead compound for the development of drugs potentially useful against those pathologies whose aetiology is related to ROS-mediated injuries

    First report of Theileria annulata in Nigeria : findings from cattle ticks in Zamfara and Sokoto States

    Background: Ticks and tick-borne pathogens (TBPs) represent a significant economic burden to cattle farming in sub-Saharan Africa including Nigeria. However, in the northern part of this country, where the largest livestock population resides, little is known about the contemporary diversity of ticks and TBPs. This area is particularly vulnerable to climate change, undergoing marked transformation of habitat and associated flora and fauna that is also likely to include ticks. This study aimed to document the occurrence of tick species and Apicomplexan TBPs in cattle from north-western Nigeria. Methods: In 2017, ticks were collected from cattle in Zamfara and Sokoto States and identified morphologically. Additionally, a subset of ticks was screened molecularly for the detection of apicomplexan DNA. Results: A total of 494 adult ticks were collected from 80 cattle in Zamfara and 65 cattle in Sokoto State. Nine tick species were encountered, among which the presence of one, Hyalomma turanicum, had not previously been recorded in Nigeria. Hyalomma rufipes was the most prevalent tick infesting cattle in Zamfara State (76%), while Hyalomma dromedarii was the most prevalent in Sokoto State (44%), confirming the widespread transfer of this species from camels onto livestock and its adaptation to cattle in the region. Of 159 ticks screened, 2 out of 54 (3.7%) from Zamfara State and 29 out of 105 (27.6%) from Sokoto State harboured DNA of Theileria annulata, the agent of tropical theileriosis. Conclusions: This study confirms the presence of a broad diversity of tick species in cattle from north-western Nigeria, providing the first locality records for Zamfara State. The occurrence of H. turanicum indicates a distribution of this tick beyond northern Africa. This study provides the first report for T. annulata in Nigerian ticks. Given its enormous burden on livestock farming in north Africa and across Asia, further investigations are needed to better understand its epidemiology, vector transmission and potential clinical significance in cattle from northern Nigeria and neighbouring Sahelian countries

    The computer virus scourge continues to be a problem the Information Technology (IT), industries must address. Computer virus is a malicious program codes, which can replicate and spread infections into large number of possible hosts and cause damage to computer programs, files, databases and data, in general. This paper focuses on the 21st century virus scourges in the IT industries and the ways of combating them vis- a- vis, the surveys carried out on the types of computer viruses and how these malicious programs infect the computers, the consequences of such infections, the possible measures of prevention and recovery from the damages. However, findings show that, the defensive mechanisms developed were confirmed good and efficient in combating the computer virus attack and reducing the occurrence of the infection.     Evaluation of Bacterial Diversity in Palm Wine by 16S rDNA Analysis of Community DNA

    Bacterial diversity and fermentation dynamics in palm wine, a traditional alcoholic fermented beverage, collected from upright palm trees from Idiaba community, Abeokuta, Ogun State, Nigeria were evaluated by DNA based method using the 16S rDNA of the microbial community to verify and complement previous reports, improve our understanding and document yet unreported, uncultured microbial diversity associated with palm wine. The 16S rRNA gene fragments were amplified from microbial community and genomic DNA of isolates, by Polymerase Chain Reaction (PCR) using universal primers; and sequenced. The partial sequences were identified by comparison with sequences deposited in the non-redundant nucleotide database of National Center for Biotechnology Information(NCBI). This analysis revealed that 32 community clones were identified as Lactobacillus sp, Lactobacillus casei strain zhang, Lactobacillus plantarum, Leuconostoc mesenteriodes ssp dextranicum, Leuconcostoc lactis, Pediococcus parvulus strain Bpe-299, Acetobacter pomorum, Acetobacter pasteurianus, Gluconobacter oxydans, Acinetobacter calcoaceticus, Enterobacterium bacterium, Acidovorax sp, Comamonas sp, Bacillus subtilis, Staphylococcus piscifermentans and uncultured bacteria clone D1-78. The results showed that bacterial diversity in the palm wine sample is dominated by Lactobacillus and Leuconostoc species as reportedby previous workers and uncultured bacteria clone D1-78 (1 clone) was detected for the first time in palm wine.Keywords: Palm wine, 16S rRNA gene, community DNA, PCR, lactic acid bacteria

    Comparative study of factors affecting birth weight of Muturu, N'dama and Boran cattle in South eastern Nigeria

    Data on two hundred and thirty five (235) calves consisting of 45 Muturu, 140 N'dama and 50 Boran were subjected to factorial analyses over the period January 1997 to April 2002, during which some factors affecting birth weight were studied. Results indicated that the mean birth weights of Muturu, N'dama and Boran calves were 14.04±2.60, 13.89±0.28 and 24.50±0.70 kg respectively.
