    Effect of two pre-freezing methods on quality of sexed semen in Ettawa Grade goat

    The aim of this study was to determine the influence of pre-freezing different procedures to the quality of Ettawa Grade goat frozen semen-sexing. The research material was semen sexing consisted of 2 layers those were top layer and bottom layer. The quality of frozen semen sexing was observed by comparing the pre-freezing technique (factory standard operating procedure: FSOP) according to the  Artificial Insemination Center operating procedure (pre-freezing by placing the straw of semen for about 4 cm above liquid nitrogen for 9 minutes) with the modification procedure (MP), pre-freezing by placing the straw of semen 16 cm above liquid nitrogen for 9 minutes and then it was lowered to 4 cm above liquid nitrogen for 9 minutes during the pre-freezing phase. The parameters observed were motility, progressive motility, hyperactivation, and sperm linearity. Data were analyzed using Student's t-test.The results of this study indicated that the quality of sexed-semen in the standard operating procedures of frozen semen compared to the treatment of modifications to the top and bottom layers were motility at the top layer (46.06 ± 7.52% vs 55.6 ± 7.78%) and bottom layer (36.82 ± 6.49% vs. 41.47 ± 6.57%); progressive top layer (16.34 ± 4.27 vs. 32.83 ± 5.9%) and bottom layer 15.97 ± 2.72% vs. 19.79 ± 3.97%); hyperactivity in the top layer (0.81 ± 0.6% vs 4.09 ± 1.98%) and the bottom layer (0.71 ± 0.68% vs. 1.50 ± 1.05%); linearity consisted of linear and non-linear, the top layer (12.19 ± 2.94 vs. 20.52 ± 3.97%) and bottom layer (12.32 ± 2.63 vs 14.70 ± 2.6); while non-linear in top layer (0.14 ± 0.2 vs 0.68 ± 0.85%) and bottom layer (0.4 ± 0.13% vs 0.34 ± 0.4%). The conclusions in this study indicated that the quality of the frozen sexed-semen that has processed by pre-freezing modification technique was better than the frozen sexed-semen obtained from the Artificial Insemination Center Standard Operating procedure

    Pengaruh Tarum (Indigofera zollingeriana) dalam Pengencer Semen terhadap Viabilitas dan Tudung Akrosom Utuh Pada Spermatozoa Kambing Peranakan Ettawa

    Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan pengencer tarum (Indigofera zollingeriana) dalam pengencer semen terhadap viabilitas dan tudung akrosom utuh semen kambing Peranakan Ettawa. Materi yang digunakan dalam penelitian ini yaitu semen segar pejantan kambing Peranakan Ettawa (PE) sebanyak 5 ekor yang berumur 2-4 tahun dengan bobot badan 74-92 kg. Komponen Bahan pengencer yang dipergunakan adalah pengencer semen dan larutan tarum. Hasil penelitian menunjukkan bahwa, penambahan larutan tarum dalam pengencer semen tidak mempengaruhi viabilitas dan tudung akrosom utuh semen kambing Peranakan Ettawa (P>0,05). Kesimpulan dalam penelitian ini adalah empat perlakuan yang diujicobakan memiliki kemampuan yang sama dalam mempertahankan viabilitas dan tudung akrosom utuh  spermatozoa kambing Peranakan Ettawa

    Seleksi Pejantan Ayam Kampung Berdasarkan Breeding Value terhadap Gerak Massa, Abnormalitas dan Motilitas Spermatozoa

    Penelitian ini bertujuan untuk mengetahui keunggulan dari masing - masing pejantan ayam kampung berdasarkan gerak massa, abnormalitas dan motilitas spermatozoa. Materi yang digunakan pada penelitian ini yaitu 10 ekor ayam kampung berumur 10-12 bulan, berdasarkan panjang taji 2-3 cm serta bobot badan berkisar 2 – 2,5 kg. Alat yang akan digunakan adalah kandang individu, tempat makan, tempat minum, tabung evendof, bekker glass, pipet, mikroskop, bunsen, object glass, cover glass dan handtally counter. Bahan yang digunakan antara lain semen, tisu, eosin 2% dan pengencer NaCl. Penelitian  menggunakan rancangan non parametrik yaitu One Way ANOVA dan Kruskal Wallis. Hasil penelitian menunjukkan bahwa volume semen dari masing-masing pejantan berbeda nyata (P?0,05), sedangkan pH, warna, konsistensi, bau, gerak massa, abnomalitas dan motilitas tidak berbeda nyata (P?0,05). Hasil analisis dari kualitatif dan kuantitatif spermatozoa masing-masing pejantan memiliki total nilai breeding value yaitu ayam 2(42), 6(42), 7(42), 8(42), 9(42), dan 10(42). Berdasarkan hasil penelitian, dapat disimpulkan bahwa ayam yang dipilih adalah dari penilian total breeding value adalah ayam 2, 6, 7, 8, 9 dan 10

    KARAKTERISTIK DAN KUALITAS SEMEN BERBAGAI GALUR AYAM KEDU (Characteristic and Cemen Quality at Various Lines of Kedu Chicken)

    ABSTRAK Ayam kedu adalah ayam lokal yang mempunyai ciri–ciri khas dan telah lama terdapat di Desa Kedu, Kecamatan Kedu–Kabupaten Temanggung. Tujuan penelitian adalah mengamati derajat keasaman (pH), daya hidup dan mortalitas semen ayam Kedu hitam daging hitam (HH), Kedu putih daging putih (PP) dan Kedu hitam daging putih (HP). Serta untuk mengetahui karakteristik semen dari berbagai galur warna ayam Kedu.Materi yang digunakan dalam penelitian ini adalah semen yang diambil dari 15 ekor ayam kedu jantan, terdiri dari 5 ekor ayam kedu hitam daging hitam (HH), 5 ekor ayam kedu hitam daging putih (HP) dan 5 ayam kedu putih daging putih (PP), dengan kisaran umur 8-12 bulan. Hasil penelitian menunjukkan bahwa rata-rata abnormalitas sperma ayam kedu HH, HP dan PP adalah : 12,87%, 11,84% dan 11,82% tidak berbeda nyata. Karakteristik semen dari berbagai galur warna ayam kedu yang meliputi volume, konsentrasi dan abnormalitas tidak berbeda nyata. Hasil uji statistik terhadap derajat keasaman (pH), daya hidup dan persentase mortalitas semen berdasarkan warna bulu ayam Kedu tidak menunjukkan perbedaan (P≥0,05). Hasil penelitian dapat disimpulkan bahwa derajat keasaman (pH), daya hidup semen dan mortalitas semen ayam Kedu berdasarkan warna bulu tidak berbeda. ABSTRACT Kedu chicken is local chicken which have typical characteristic and there was in countryside of Kedu, district of Kedu-Temanggung regency. The aim of research was to perceive degree of acidity (pH), energy live and cemen mortality of black Kedu chicken of black flesh (HH), White Kedu of white meat (PP) and of Kedu black of white meat (HP). And also to know cemen characteristic from various chicken colour galur of Kedu. Items which used in this research was taken away from by cemen of 15 kedu chicken male, consist of 5 Kedu chicken of kedu black of black flesh (HH), 5 kedu chicken black of white meat (HP) and 5 Kedu chicken white of white meat (PP), with of old age was 8-12 months. The result of research indicated that mean of abnormality Kedu chicken sperm of HH, HP and of PP was : 12,87%, 11,84% and 11,82% non differ significant. The cemen characteristic from various chicken colour lines of kedu covering volume, and concentration of abnormality was not differ significant. Result of statistical test to acidity degree (pH), energy live and percentage of cemen mortality pursuant to quill colour of Kedu was not show difference (P≥0,05). Result of research can be concluded that acidity degree (pH), life energy cemen and cemen mortality of Kedu chicken pursuant to fur colour was not differ.     The aims of this research were to observed the effect of zinc (Zn), magnesium (Mg) and selenium (Se) to the abundance, color and spinnbarkeit of cervical mucus. The materials used 10 Timor deer with BCS (Body Condition Score) 2-3,25, poel 2 and had different estrous phase. The parameters were observed the abundance, color and spinnbarkeit of cervical mucus. The data of abundance and color of cervical mucus were analyzed by Mann Whitney U-test and the data of spinnbarkeit were analyzed by t-test. The treatments were zinc (Zn), selenium (Se) and magnesium (Mg) supplementation (T1) on mineral block form and control (T0). The results showed that Zn, Se, and Mg supplementation had no significant effect (P>0,05) to the abundance, color and spinnbarkeit of cervical mucus Timor deer on estrous phase. It was caused by the high zinc level on feed, so the cervical mucus profile had no difference between T0 and T1. Key words : Mineral Supplementation, Cervical Mucus, Timor Dee
