    BRAF mutation in colorectal carcinomas with signet ring cell component

    WOS: 000417395100008PubMed ID: 28884045Objective: Signet ring cell carcinoma is a rare subtype of colorectal carcinoma (CRC) with an associated BRAFV600E mutation. We investigated frequencies of BRAF mutation in 28 CRCs containing variable signet ring cell component and their relation with clinicopathologic parameters. Methods: According to the presence of signet ring cell component, tumors were categorized into groups as follows: 0%-9%, 10%-24%, 25%-49%, and >50%. Genomic DNA was isolated and analyzed for BRAF V600E gene mutation by polymerase chain reaction-restriction fragment length polymorphism. Eleven of 28 cases (39.3%) showed BRAFV600E mutation, which was also confirmed by Sanger sequencing. To elucidate the importance of existence of signet ring cell component at the molecular level, we separated cases into two groups with cut-off levels of 10% and 50%, which pertain to percentages of signet ring cells. Results: Seven of 19 cases (36.8%) under the threshold of 50% and four of nine cases (44.4%) over this threshold value demonstrated BRAF mutation. Three of 7 cases (42.8%) featuring 10% were BRAF mutated. Conclusions: BRAF mutation must be closely associated with the presence of malignant signet ring cells regardless of their percentages

    Livedo reticularis is a reticular discoloration of the skin because of the vascular anatomy of the skin. The condition most commonly affects the legs. Drug induced livedo reticularis which is an acknowledged side effect of amantadine, tends to be widespread, asymptomatic, benign rash. There are also reports of livedoid eruption induced with drugs including dapsone, imatinibe, gefitinibe. We describe a case of livedo reticularis like eruption and haemotological toxicity with trimetophrim-sulfamethoxazole. The purpose of this report is to remind clinicians of this rare, benign side effect of the common prescribed medication

    Adapting the socio-cultural adaptation scale (SCAS-R) to Arabic: A study on the Syrian migrants living in Gaziantep province of Turkiye

    This study aims to investigate the validity, reliability, and Arabic language equivalence of the Socio-cultural Adaptation Scale (SCAS-R) created by Colleen Ward and Antony Kennedy (1999) and revised by Jessie Wilson (2013). A sample group of 424 Syrian Migrants (18 years and older) living in Gaziantep province of Turkiye from different neighborhoods, economic status, and socio-demographic backgrounds are included in the research, and the scale is examined via commonly used validity and reliability analysis methods. It is obtained that the Cronbach's Alpha of the items is higher than 0.7 and the corrected item-total correlations are above the threshold value (0.2) in item analysis, nearly 69% of the total variance is explained by 5 factors in Exploratory Factor Analysis (EFA), and the Goodness of Fit Indexes (chi 2/sd = 1.521, CFI = 0.958, and SRMR = 0.048) are within the good/acceptable range in Confirmatory Factor Analysis (CFA). It has been understood that the SCAS-R is a valid and reliable scale for Arabic culture. The results of this study may provide a valuable tool for policymakers, researchers, and humanitarian workers studying migration issues

    Attitudes of nursing and medical school students towards ageism

    The present study aimed to evaluate the attitudes of nursing and medical school students towards age discrimination and to determine the association between these attitudes and various variables. A total of 662 students, 328 from the Faculty of Nurse Education and 334 from the Faculty of Medicine, participated in the present descriptive and cross-sectional study.Data were collected using Data Form for socio-demographic characteristicsand Ageism Attitude Scale (AAS). Descriptive statistics, t-test, Mann-Whitney U test, Chi-square test, One-Way ANOVA, and Kruskal Wallis test were used to evaluate the data using IBM SPSS Statistics for Windows (Version 20.0).Nursing school students had higher points in restricting the life of elderly people, positive discrimination towards them and Ageism Attitude Scale, but had lower points in negative discrimination towards them compared to medical school students. While the points of positive discrimination towards elderly and Ageism Attitude Scale of the students of two schools were not significantly different (p>0.05), points of restricting the life of elderly people and negative discrimination towards elderly varied significantly between the students of two schools (p [Med-Science 2020; 9(4.000): 848-55

    Methemoglobinemia after local anesthetic (prilocaine) application: a case report

    Introduction: Circumcision is a surgical procedure performed by many societies for religious, medical and cultural purposes. One of the agents used during this procedure is prilocaine, a commonly used local anesthetic. In the literature, cases of methemoglobinemia which can be developed with the application of prilocaine in normal dose have been reported. Oxidation of hemoglobin with various oxidative stresses is called methemoglobinemia when the divalent iron in its content is converted into its trivalent form. This reaction leads to hypoxaemia in the tissue and a clinical picture of cyanosis occurs at varying degrees.Case Report: Two-and-a-half-month-old patient was referred to my hospital by a physician because of the complaint of bruising after local anesthesia with prilocaine and the measured oxygen saturation was 79%. Physical examination revealed cyanosis in the peroral region, hands and feet. It was decided that methylene blue should be administered considering the fact that the patient was given prilocaine as a local anesthetic, due to the inability to find an underlying respiratory and cardiac cause, and because of the high value of methemoglobin in blood gas. Methylene blue 2 mg/kg was given orally. About an hour later, cyanosis disappeared. The control methhemoglobin level was found to be 2.7%. Conclusion: Methemoglobinemia should be considered in the differential diagnosis of cyanosis after local anesthetic application and local anesthetics with less side effects should be preferred in early infancy.     WOS: 000294879500008This study was conducted to determine the nutritive value of live yeast culture (RumiSacc, Saccharomyces cerevisiae) and to investigate its effects on milk yield, milk composition and some blood parameters in lactating cows. Six multiparous Holstein cows were allocated to two groups of three cows and assigned randomly to one of two diets in a cross-over experiment. Daily 50 g RumiSacc was top dressed at the p.m. feeding for the treatment group. RumiSacc supplied a high protein and energy with high organic matter digestibility values (83.35%) determined by in vitro enzymatic analysis. Yeast culture supplementation significantly increased milk yield, tended to increase fat yield, protein yield and lactose yield of milk. Methylated fatty acid level of 18:3 (n-3) in milk fat was increased by yeast culture supplementation. The concentrations of methionine, phenyalanine, tyrosine, tryptophan and taurine were significantly increased with dietary inclusion of yeast culture. Live yeast culture supplementation did not affect other performance characteristics, milk quality characteristics and blood parameters. As a conclusion live yeast culture (RumiSacc, Saccharomyces cerevisiae) had high nutritive value and positive effects on milk production and some milk quality characteristics in lactating cows under field conditions
