Tests multiples pour la comparaison des probabilités de survenue d'une infidélité de transcription dans des ARNm sains et cancéreux
International audienceL'implication d'erreurs de transcription dans l'hétérogénéité du cancer avait jusqu'alors été peu considérée. En effet, la transcription est supposée fidèle et contrôlée par un système complexe de vérification. Afin d'étudier l'hétérogénéité des séquences d'ARNm issus de tissus sains et cancéreux de 17 gènes d'intérêt, les probabilités de survenue d'une substitution de base ont été comparées à chaque position des séquences des transcrits à l'aide d'une procédure de tests multiples. Pour cela, les séquences Expressed Sequences Tags, qui sont des copies partielles des ARNm d'un gène, ont été utilisées et un modèle prenant en compte l'erreur de séquençage inhérente à ces données a été proposé. Enfin, l'estimateur Location Based Estimator du nombre moyen de tests faux positifs a été étendu au cas de statistiques de tests discrètes. Cette étude préliminaire a ainsi permis de mettre en évidence les positions des ARNm plus fréquemment sujettes à des substitutions dans les tissus cancéreux que dans les tissus sains et d'introduire la notion d'infidélité de transcription chez l'Homme
Rôle de la structure quaternaire de la Glycéraldéhyde 3-phosphate Déshydrogénase (GAPDH) phosphorylante de Bacillus stearothermophilus dans la révélation de l'activité enzymatique et les propriétés de coopérativité de fixation du cofacteur
Non disponible / Not availableNotre étude a consisté à étudier les propriétés des gapdh dimeriques générées par ingénierie protéique, à les relier à leur état oligometrique et à les comparer à celles de l'enzyme tetramerique de type sauvage. A partir des structures cristallographiques des formes Apo- et holoenzyme de la gapdh de b. Stearothermophilus connues à haute résolution et grâce à l'outil modélisation moléculaire, des mutations appropriées ont été introduites aux interfaces r et q de la gapdh tetramerique de type sauvage et ont permis de générer des gapdh mutées produites sous forme strictement dimerique ou sous deux formes dimère et tétramère en équilibre. Notre étude a montré que les dimères générés sont très probablement de type o-p, en raison du plus grand nombre de contacts interatomiques s'établissant le long de l'interface p, du plus petit nombre de résidus hydrophobes exposés au solvant et de leur meilleure affinité pour le nadp comparée à celle pour le nad. Les gapdh dimeriques générées sont inactives tandis que les formes tetrameriques générées possèdent une activité enzymatique variable suivant les cas. Cette absence d'activité peut-être attribuée à une modification de la conformation du site actif, comparée à celle dans le tétramère. La fixation du nad par la gapdh de type sauvage et les gapdh tetrameriques mutées peut être décrite par un modèle à au moins 2 constantes de dissociation, indiquant que ces enzymes sont en fait constituées de 2 dimères indépendants. Cela n'est pas en contradiction avec la structure cristallographique de la gapdh de b. Stearothermophilus qui peut être décrite comme un dimère de dimères. Les gapdh dimeriques testées conservent des propriétés de coopérativité de fixation du cofacteur, ce qui indique que l'interface p est essentielle pour transmettre l'information structurale. L'étude de l'équilibre existant entre les formes dimerique et tetramerique du mutant (n180g-e276g) a montré une apparente non totale réversibilité de l'équilibre
Transcriptional frameshifts contribute to protein allergenicity
Transcription infidelity (TI) is a mechanism that increases RNA and protein diversity. We found that single-base omissions (i.e., gaps) occurred at significantly higher rates in the RNA of highly-allergenic legumes. Transcripts from peanut, soybean, sesame, and mite allergens contained a higher density of gaps than those of non-allergens. Allergen transcripts translate into proteins with a cationic carboxy-terminus depleted in hydrophobic residues. In mice, recombinant TI variants of the peanut allergen Ara h 2, but not the canonical allergen itself, induced, without adjuvant, the production of anaphylactogenic specific IgE (sIgE) binding to linear epitopes on both canonical and TI segments of the TI variants. The removal of cationic proteins from bovine lactoserum markedly reduced its capacity to induce sIgE. In peanut-allergic children, the sIgE reactivity was directed toward both canonical and TI segments of Ara h 2 variants. We discovered two novel peanut allergens because of their RNA-DNA divergence gap patterns and TI peptide amino-acid composition. Finally, we showed that the sIgE of children with IgE-negative milk allergy targeted cationic proteins in lactoserum. We propose that it is not the canonical allergens, but their TI variants, that initiate sIgE isotype switching, while both canonical and TI variants elicit clinical allergic reactions

Misfolded forms of glyceraldehyde-3-phosphate dehydrogenase interact with GroEL and inhibit chaperonin-assisted folding of the wild-type enzyme
We studied the interaction of chaperonin GroEL with different misfolded forms of tetrameric phosphorylating glyceraldehyde-3-phosphate dehydrogenase (GAPDH): (1) GAPDH from rabbit muscles with all SH-groups modified by 5,5′-dithiobis(2-nitrobenzoate); (2) O-R-type dimers of mutant GAPDH from Bacillus stearothermophilus with amino acid substitutions Y283V, D282G, and Y283V/W84F, and (3) O-P-type dimers of mutant GAPDH from B. stearothermophilus with amino acid substitutions Y46G/S48G and Y46G/R52G. It was shown that chemically modified GAPDH and the O-R-type mutant dimers bound to GroEL with 1:1 stoichiometry and dissociation constants Kd of 0.4 and 0.9 μM, respectively. A striking feature of the resulting complexes with GroEL was their stability in the presence of Mg-ATP. Chemically modified GAPDH and the O-R-type mutant dimers inhibited GroEL-assisted refolding of urea-denatured wild-type GAPDH from B. stearothermophilus but did not affect its spontaneous reactivation. In contrast to the O-R-dimers, the O-P-type mutant dimers neither bound nor affected GroEL-assisted refolding of the wild-type GAPDH. Thus, we suggest that interaction of GroEL with certain types of misfolded proteins can result in the formation of stable complexes and the impairment of chaperonin activity
Detection of IgE-reactive proteins in hydrolysed dog foods
BACKGROUND
Commercial hydrolysed diets are used for the diagnosis of food allergy in dogs. The cleaved parent proteins are presumed to be too small to elicit an allergic response by reacting with allergen-specific immunoglobin E (IgE).
OBJECTIVES
To evaluate three commercial hydrolysed dog diets for proteins.
ANIMALS
Sera were collected from dogs with suspected food allergy.
METHODS
Two batches of each hydrolysed diet were examined by electrophoresis and visualized by Coomassie blue, silver nitrate staining and IgE immunoblotting.
RESULTS
From two to five proteins, ranging from 21 to 67 kDa, were detected in all three diets evaluated. Circulating IgE antibodies targeting these proteins were detected by immunoblotting of dog sera. Six different carbohydrate proteins were identified by mass spectrometry; maize/potato granule-bound starch synthase-1, soybean glycinin, soybean β-conglycinin α chain, potato aspartic protease inhibitor, rice glutelin type B1 and soybean sucrose-binding protein. Four of these proteins have been described as allergens in humans.
CONCLUSIONS
Some commercial hydrolysed diets contain carbohydrate proteins. Some dogs have circulating IgE antibodies targeting these proteins. The clinical significance of these findings is unknown
