Über die Regulation der Calcium-Homöostase und den Einfluss von STIM-Proteinen auf die Alzheimer-Pathologie
Weltweit erkrankt alle drei Sekunden ein Mensch an einer Demenzerkrankung. Morbus Alzheimer
bildet hierbei in 50 bis 60 % der Fälle die Ursache und stellt somit die häufigste Demenzform dar.
Makropathologisch ist die Krankheit durch eine gleichmäßige Hirnrindenatrophie mit Erweiterungen
der Liquorräume gekennzeichnet. Das histopathologische Korrelat liegt in einer massiven
Neuronendegeneration mit extrazellulären Ablagerungen von Aβ – Plaques und intrazellulären
hyperphosphorylierten neurofibrillären Tau – Bündeln. Aβ entsteht im Zuge einer amyloidogenen APP
– Prozessierung durch sequenzielle Schnitte der β – und γ – Sekretasen. Presenilin – Proteine stellen
dabei das katalytische Zentrum der γ – Sekretase dar. Neben einem amyloidogenen APP –
Prozessierungsweg wird durch den Schnitt der α – Sekretase der nicht – amyloidogene Weg eingeleitet.
Das dabei anfallende sAPPα wird in den Extrazellulärraum freigesetzt und kann neuroprotektive
Eigenschaften entfalten. Die Literatur berichtet von einer gestörten 2+ - Homöostase bei Morbus
Alzheimer. Sowohl APP als auch Aβ, sAPPα und Preseniline greifen in die Regulation des 2+ -
Haushaltes ein. Um die intrazelluläre 2+ - Konzentration ( [2+]) zu regulieren, sind viele
Mechanismen erforderlich, die den 2+ - Transport ĂĽber die Plasmamembran sowie zwischen dem
Zytosol und den intrazellulären Zellkompartimenten ermöglichen. Dazu gehören u.a. die
spannungsgesteuerten (VGCC) und die speichergesteuerten 2+ - Kanäle (CRAC), die den SOCE (engl.
store operated 2+ - entry) aktivieren. Im Rahmen dieser Arbeit wurde folglich der Einfluss von APP,
seinen Prozessierungsprodukten und von Presenilin – Proteinen auf CavB, CavD und CavG als Vertreter
der VGCC – Familie sowie auf ORAI1, ORAI2, ORAI3, STIM1, STIM2, STIM2.1 und STIM2.2 als SOCE –
Komponenten untersucht. Anschließend erfolgte die Analyse der reversen Regulation von den α -, β –
und γ – Sekretasen durch STIM1 und STIM2. Schließlich wurden humane post mortem Hirnproben der
Analyse der Genexpression von CavB, CavD, ORAI1, ORAI2, ORAI3, STIM1 und STIM2 unterzogen.
Zunächst wurde der Einfluss von Presenilin - Proteinen auf das intrazelluläre 2+ - Signal in
unterschiedlichen murinen und humanen Zelllinien untersucht. Während in Bezug auf Presenilin 1
keine eindeutige Aussage bzgl. der Regulation von SOCE gemacht werden konnte, schienen die
Ergebnisse in Bezug auf Presenilin 2 für einen SOCE – aktivierenden Einfluss zu sprechen. Die
Untersuchungen der [2+] konnten keinen Einfluss von Presenilin1 auf die Aktivität von SOCE
aufzeigen, jedoch sprachen die signifikanten Veränderungen der Genexpression von STIM – und ORAI
– Proteinen in Presenilin1 – und Presenilin1/2 – defizienten Zellen für einen modulierenden Einfluss
von Presenilin1 auf der Genebene. Presenilin2 scheint SOCE ĂĽber eine forcierte Speicherentleerung
des endoplasmatischen Retikulums potenzieren zu können. In der Literatur häufen sich die Belege für
einen SOCE – hemmenden Einfluss von Presenilin2. Hierbei übt am ehesten Presenilin1 einen
ihhibierenden Einfluss auf die Funktion von Presenilin2 aus, weswegen die gestörte
Speicherentleerung des endoplasmatischen Retikulums zu seiner 2+ -Ăśberladung fĂĽhrt und folglich
der SOCE beeinträchtigt werden kann. Im Rahmen dieser Arbeit konnte ein SOCE – hemmender
Einfluss sowohl auf der Gen – als auch auf der Aktivitätsebene durch AICD/AL2ICD nachgewiesen
werden. AICD/AL2ICD sind hingegen Spaltprodukte, die im Zuge der APP – Prozessierung durch die γ –
Sekretase anfallen. Demnach scheint Presenilin1 und/oder Presenilin2 zum einen direkt ĂĽber die
Regulation der Speicher des endoplasmatischen Retikulums und zum anderen indirekt ĂĽber die
Produktion von genregulatorischen AICD/AL2ICD den SOCE zu modulieren.
Des Weiteren wurde in dieser Arbeit ein Hinweis auf den SERCA – fördernden Einfluss von AICD/AL2ICD
entdeckt. Die Literatur berichtet von einer Aktivierung der SERCA – Pumpe durch Presenilin1 und
Presenilin2 im Zuge einer direkten Interaktion. In dieser Arbeit lagen Hinweise auf eine zusätzliche
indirekte Aktivierung der SERCA ĂĽber die Bildung von AICD/AL2ICD vor. Somit scheint AICD/AL2ICD
einen wichtigen Regulator der 2+ - Homöostase darzustellen. AICD/AL2ICD schützt demnach vor
einer 2+ - Ăśberladung der Zelle, indem sie den 2+ - Influx aus dem Extrazellularraum durch die
Hemmung von SOCE kontrolliert und über die Aktivierung der SERCA – Pumpe 2+ - Ionen in die
intrazellulären Speicher bringt. Beides führt zu einer Senkung der [2+] .
Im Rahmen dieser Arbeit konnte außerdem nachgewiesen werden, dass STIM – Proteine direkt, 2+
- unabhängig und indirekt über die Regulation der [2+] die Funktion der α – Sekretase sowohl auf
der Protein - als auch auf der Aktivitätsebene steigern, wohingegen die Funktion der γ – Sekretase auf
der Gen -, Protein – und Aktivitätsebene herunterreguliert wird. Demnach übt STIM1 und STIM2.2
einen inhibierenden Einfluss auf die γ – Sekretase aus, wohingegen STIM2 diese fördert. Bezüglich der
Funktion der β – Sekretase lagen nur indirekte Hinweise auf einen hemmenden Einfluss von STIM vor.
Die Untersuchung der Genexpression von CavD wies auf eine mögliche Regulation sowohl durch Aβ als
auch durch AICD hin. Während APP – defiziente Zellen, die folglich weder Aβ noch AICD exprimieren,
sowie Zellen mit herunterreguliertem Fe65, einem Translokationspartner von AICD, eine Reduktion
der Genexpression gezeigt haben, wiesen Zellen mit einer überschießenden Aβ – Produktion ebenfalls
eine Reduktion der Genexpression auf. Presenilin1 – defiziente Zellen konnten keinen Unterschied
verglichen zur Kontrolle zeigen. Somit scheint AICD die Genexpression von CavD zu fördern,
wohingegen Aβ diese hemmt. Die Genexpression von CavB wird am ehesten durch Aβ reguliert, wobei
es die Genexpression von CavB inhibiert. Im Fall von CavG bewirkt AICD ebenfalls am ehesten eine
Reduktion der Genexpression.
Zusätzlich wurde in humanen post mortem Hirnproben untersucht, inwiefern die Genexpression von
den oben angeführten ORAI -, STIM – und VGCC – Proteinen in AD verändert ist. Gegenüber der
Kontrollkohorte konnte eine signifikante Steigerung aller Gene nachgewiesen werden. Die
Genexpression von CavB, CavD, ORAI1, ORAI3, STIM1 und vermutlich STIM2 erfährt mit steigendem
Alter bei Vorliegen von AD eine Reduktion, wobei CavD unabhängig von AD mit dem Alter negativ
korreliert. AuĂźerdem konnte fĂĽr CavB, ORAI1, ORAI3 und STIM1 eine positive Korrelation zwischen
dem Braak – Stadium I und den Stadien V- VI ermittelt werden, wohingegen STIM2 nur zwischen den
Stadien I und VI positiv korrelierte. In Bezug auf die Allelkonstellation von ApoE ergaben die
Untersuchungen der Genexpression eine Steigerung der Gene CavB, CavD und ORAI1 bei zusätzlichem
ε4 – Allel, wohingegen die Genexpression von CavD bei zusätzlichem ε3 – Allel abnahm. Es lagen keine
Hinweise auf einen Zusammenhang mit dem Geschlecht vor.There is one person every three seconds who get dementia worldwide. There are 50 – 60 % of all cases
of dementia that go to the Alzheimer’s disease, therefore Alzheimer’s disease makes up the most
common cause of dementia. Macroscopic hallmarks are the equally distributed atrophy of the cortices
and the widening of the liquor spaces, whereas the histopathological changes are represented by a
massive neurodegeneration accompanied by extracellular amyloid (Aβ) deposits and intracellular
hyperphosphorylated neurofibrillary tau tangles. Aβ arises as a product of the amyloidogenic APP
processing by sequential cleavage of β – and γ – secretases. Presenilins as one of the subunits of the γ
– secretase exert hereby the catalytic activity. α – secretase is the next enzyme that processes the non
– amyloidogenic APP proteolysis with sAPPα being secreted in the extracellular space and having
neuroprotective properties. There is evidence of a disturbed 2+ - homeostasis in AD. APP as well as
Aβ, sAPPα and presenilins seem to be involved in the regulation of the 2+ - household. To balance
the intracellular 2+ concentration ([2+]) plenty of agents are required to mediate the 2+
transport across the plasma membrane and between the cytosol and intracellular cell compartments.
The voltage gated 2+ - channels (VGCC) and the 2+ release – activated channels (CRAC)
mediating the store operated 2+ entry (SOCE) are some of them. In scope of this work the influence
of APP, it’s cleavage products and presenilins was analyzed on CavB, CavD and CavG being
representatives of the VGCC family and on ORAI1, ORAI2, ORAI3, STIM1, STIM2, STIM2.1 and STIM2.2
as SOCE components. Subsequently the reverse regulation of the α -, β – and γ – secretases by STIM1
and STIM2 was analyzed. In the end the gene expression of CavB, CavD, ORAI1, ORAI2, ORAI3, STIM1
and STIM2 was investigated in the humane post mortem cerebral samples.
First of all the influence of presenilins on the 2+ signaling has been tested in different mouse and
humane cell lines. While no exact conclusions could be made regarding presenilin 1, the results
concerning presenilin 2 indicated a SOCE activating effect. Presenilin1 seems to be not involved in the
regulation of the acitivity of SOCE, however there is a clue that presenilin1 interfere with the gene
regulation as it was seen in presenilin1 – and presenilin1/2 – deficient cell lines. Presenilin2 seems to
facilitate SOCE by intensifying the emptying of the endoplasmatic reticulum – storages. There is a body
of evidence that presenilin1 inhibits SOCE. So presenilin1 seems to have an inhibitory effect on the
functioning of presenilin2 leading to a 2+ overload in endoplasmatic reticulum by minimizing its
storage emptying. Next discovered was the SOCE inhibitory effect of AICD/AL2ICD that was confirmed
at the gene and the acitivity level. In turn AICD/AL2ICD are cleavage products of APP that arise in the
course of proteolysis by γ – secretase. Therefore presenilin 1 and 2 apparently modulate SOCE directly
by regulating the storages of endoplasmatic reticulum and indirectly by producing the gene regulatory
AICD/AL2ICD.
Furthermore a SERCA pump activating effect of AICD/AL2ICD could be shown in the course of the work.
The literature reports about the SERCA activating effect of presenilins by direct interaction. So
presenilins can activate the SERCA pump by producing AICD/AL2ICD additionally. Consequently,
AICD/AL2ICD play an important role in the regulation of the 2+ - homeostasis by preveting the cell
against a 2+ overload. Firstly, AICD/AL2ICD controls the 2+ influx across the plasma membrane
by inhibiting SOCE and secondly it stores 2+ in the intracellular cell compartiments by activating the
SERCA pump.
Investigating the protein and acitivity level it could be shown that STIM proteins are able to activate
the α – secretase both directly, 2+ - independently as well as indirectly,
2+ - dependetly, whereas the functioning of the γ – secretase as shown on the gene, protein and activity level could be down regulated. Furthermore STIM1 and STIM2.2 seem to inhibit, whereas STIM2 seems to facilitate the γ –secretase. Referring to β – secretase only some clues were given for STIMs to inhbit it’s functioning.
Analyzing the gene expression of CavD evidence was given of Aβ and AICD being involved in its
regulation. While APP – deficient cells being avoid of Aβ and AICD as well as cells with down regulated
AICD translocation partner Fe65 has shown a reduction of the gene expression, cells with exaggerated
Aβ production presented a reduction of the gene expression as well. In addition, cells not expressing
presenilin1 couldn’t show any difference compared to the control cells. So it seems AICD potentiates
the gene expression of CavD, whereas Aβ is likely to reduce it. CavB seems to be regulated rather by
Aβ, whereas there is an inhibitory effect being observed. Regarding CavG there is a clue, AICD reduces
it’s gene expression, as well.
Furthermore the gene expression of mentioned above ORAI -, STIM – and VGCC – proteins was
analyzed in the humane post mortem cerebral samples of patients with Alzheimer’s disease. In
comparison to the control cohort all of genes showed a significant elevation. CavB, CavD, ORAI1,
ORAI3, STIM1 and most likely STIM2 are experiencing a reduction by aging when having Alheimer’s
disease, whereas CavD is getting reduced with growing age independently of Alzheimer’s disease.
Besides that a positive correlation could be shown for CavB, ORAI1, ORAI3 and STIM1 regarding the
Braak stage I and V - VI, whereas STIM2 proved a positive correlation between the stages I and VI.
Analyzing the importance of the ApoE allels combination, an increase of the gene expression of CavB,
CavD and ORAI1 could be proved in case of an additional ε4 – allel. In case of an additional ε3 - allel
the gene expression of CavD is getting reduced. No correlation between the gender and gene
expression of the investigated genes could be detected
Tocotrienol Affects Oxidative Stress, Cholesterol Homeostasis and the Amyloidogenic Pathway in Neuroblastoma Cells: Consequences for Alzheimer’s Disease
One of the characteristics of Alzheimer´s disease (AD) is an increased amyloid load and an enhanced level of reactive oxidative species (ROS). Vitamin E has known beneficial neuroprotective effects, and previously, some studies suggested that vitamin E is associated with a reduced risk of AD due to its antioxidative properties. However, epidemiological studies and nutritional approaches of vitamin E treatment are controversial. Here, we investigate the effect of α-tocotrienol, which belongs to the group of vitamin E, on AD-relevant processes in neuronal cell lines. In line with the literature, α-tocotrienol reduced the ROS level in SH-SY5Y cells. In the presence of tocotrienols, cholesterol and cholesterol esters, which have been shown to be risk factors in AD, were decreased. Besides the unambiguous positive effects of tocotrienol, amyloid-β (Aβ) levels were increased accompanied by an increase in the activity of enzymes responsible for Aβ production. Proteins and gene expression of the secretases and their components remained unchanged, whereas tocotrienol accelerates enzyme activity in cell-free assays. Besides enhanced Aβ production, tocotrienols inhibited Aβ degradation in neuro 2a (N2a)-cells. Our results might help to understand the controversial findings of vitamin E studies and demonstrate that besides the known positive neuroprotective properties, tocotrienols also have negative characteristics with respect to AD
Vitamin D and Its Analogues Decrease Amyloid-β (Aβ) Formation and Increase Aβ-Degradation
Alzheimer’s disease (AD) is characterized by extracellular plaques in the brain, mainly consisting of amyloid-β (Aβ), as derived from sequential cleavage of the amyloid precursor protein. Epidemiological studies suggest a tight link between hypovitaminosis of the secosteroid vitamin D and AD. Besides decreased vitamin D level in AD patients, an effect of vitamin D on Aβ-homeostasis is discussed. However, the exact underlying mechanisms remain to be elucidated and nothing is known about the potential effect of vitamin D analogues. Here we systematically investigate the effect of vitamin D and therapeutically used analogues (maxacalcitol, calcipotriol, alfacalcidol, paricalcitol, doxercalciferol) on AD-relevant mechanisms. D2 and D3 analogues decreased Aβ-production and increased Aβ-degradation in neuroblastoma cells or vitamin D deficient mouse brains. Effects were mediated by affecting the Aβ-producing enzymes BACE1 and γ-secretase. A reduced secretase activity was accompanied by a decreased BACE1 protein level and nicastrin expression, an essential component of the γ-secretase. Vitamin D and analogues decreased β-secretase activity, not only in mouse brains with mild vitamin D hypovitaminosis, but also in non-deficient mouse brains. Our results further strengthen the link between AD and vitamin D, suggesting that supplementation of vitamin D or vitamin D analogues might have beneficial effects in AD prevention
