    Mapping glacial rock flour deposits in Tasersuaq, southern West Greenland

    Global population has increased rapidly in recent decades. So far, it has been possible to feed the growing population by using more and more land for agriculture, using irrigation and artificial fertilisers and by improving the efficiency of agriculture. Recently the growth of the global agricultural area has slowed. However, the need for food will continue to grow markedly in coming years. This demand can no longer be met by using increasingly more land for agriculture, and in many areas it is not possible to increase crop production by irrigation (Wise 2013). Large areas in the tropics are characterised by strongly depleted soils with low concentrations of nutrients such as nitrogen, phosphorous and potassium. In such areas, the yield of crop per hectare is much lower than the theoretical yield using optimal fertilising (Ray et al. 2013). Reducing the gap between real and potential crop productivity offers the best solution to achieve food security for the world’s rapidly growing population. Poor soil quality in the tropics is largely due to the rapid weathering of minerals and leaching of dissolved nutrients in the warm and humid climate. If weathered minerals are not replaced by new minerals, for example due to volcanic activity, then soil fertility continues to decline over time. Therefore, it is necessary to use increasing amounts of fertilisers to feed growing populations in the tropics. Most nutrients come from geological deposits; the only exception is nitrogen, which can be extracted from the atmosphere. Nutrients that are mined constitute a limited resource. Hence the known occurrences of phosphorous can only cover the current demand for a few decades (van Vuuren et al. 2010). In recent years, investigations have been conducted to see if the productivity of nutrient-poor soils can be improved by the application of glacial rock flour from Greenland. Rock flour in southern West Greenland consists of fine-grained silt, formed by the grinding of bedrock by stones and boulders embedded in the basal part of glaciers. Preliminary results indicate that plants cultivated in soils with rock flour can achieve increased growth (M.T. Rosing, unpublished data 2019). However, the research is still in its early days and many questions remain. We do not know why adding rock flour to soil results in increased growth. Maybe the silt fraction improves the soil properties. Also we do not know if it is feasible to mine rock flour and transport it to the tropics. As a first step towards answering some of these questions, our aim here was to simply map and sample the glacial rock flour in Tasersuaq, a large proglacial lake in southern West Greenland, c. 105 km north-east of Nuuk

    Thermal delousing has become the most applied method for treatment against salmon lice. However, the temperature range used is strongly aversive for salmonids, and the method is associated with increased mortality. Treatment temperature × duration combinations should be tailored to maximise delousing efficiency and minimize welfare impacts on the host fish. We tested the detachment rate of sessile, pre-adult and adult male and female salmon lice (Lepeophtheirus salmonis) as a function of ambient temperature (11–16 °C), exposure temperature (28–36 °C), and exposure duration (0–120 s). Dead Atlantic salmon (Salmo salar) hosts were used to avoid negative fish welfare and detachment of lice due to fish behaviour. Within the range tested, higher exposure temperatures were associated with higher detachment rates among pre-adult and adult lice, while no sessile lice detached at any temperature. Moreover, no treatment combination detached 100% of lice of any stage, and at 28 °C, detachment of adult females was negligible. Most detachments occurred within the first 30 s of exposure. We conclude that for a given delousing efficiency, lower temperatures must be compensated for by considerably longer exposure durations. This may be a higher risk for the welfare of the host fish than higher temperatures and shorter exposure durations.publishedVersio

    Deep tissue optical imaging of upconverting nanoparticles enabled by exploiting higher intrinsic quantum yield through use of millisecond single pulse excitation with high peak power

    We have accomplished deep tissue optical imaging of upconverting nanoparticles at 800 nm, using millisecond single pulse excitation with high peak power. This is achieved by carefully choosing the pulse parameters, derived from time-resolved rate-equation analysis, which result in higher intrinsic quantum yield that is utilized by upconverting nanoparticles for generating this near infrared upconversion emission. The pulsed excitation approach thus promises previously unreachable imaging depths and shorter data acquisition times compared with continuous wave excitation, while simultaneously keeping the possible thermal side-effects of the excitation light moderate. These key results facilitate means to break through the general shallow depth limit of upconverting-nanoparticle-based fluorescence techniques, necessary for a range of biomedical applications, including diffuse optical imaging, photodynamic therapy and remote activation of biomolecules in deep tissues

    Decreased IL7Rα and TdT expression underlie the skewed immunoglobulin repertoire of human B-cell precursors from fetal origin

    Newborns are unable to mount antibody responses towards certain antigens. This has been related to the restricted repertoire of immunoglobulin (Ig) genes of their B cells. The mechanisms underlying the restricted fetal Ig gene repertoire are currently unresolved. We here addressed this with detailed molecular and cellular analysis of human precursor-B cells from fetal liver, fetal bone marrow (BM), and pediatric BM. In the absence of selection processes, fetal B-cell progenitors more frequently used proximal V, D and J genes in complete IGH gene rearrangements, despite normal Ig locus contraction. Fewer N-nucleotides were added in IGH gene rearrangements in the context of low TdT and XRCC4 expression. Moreover, fetal progenitor-B cells expressed lower levels of IL7Rα than their pediatric counterparts. Analysis of progenitor-B cells from IL7Rα-deficient patients revealed that TdT expression and N-nucleotides additions in Dh-Jh junctions were dependent on functional IL7Rα. Thus, IL7Rα affects TdT expression, and decreased expression of this receptor underlies at least in part the skewed Ig repertoire formation in fetal B-cell precursors. These new insights provide a better understanding of the formation of adaptive immunity in the developing fetus

    Sex Determination and Isotopic Analysis of the Nivåfjord Mesolithic Burials, Zealand, Denmark

    Since 1992 the prehistoric Nivåfjord in northeast Zealand, Denmark, has yielded an appreciable number of inhumation burials and cremations dating to the Mesolithic, especially the sites of Nivå 10 and Nivågård. Unfortunately, the micro-region is characterised by poor organic preservation, restricting the successful application of biomolecular techniques to human remains, including large-scale radiocarbon dating programmes as well as both stable isotope and ancient DNA analyses. Here, we apply an alternative technique, an acid etch peptide-based method, to determine the sex of eight individuals from Nivå 10 as well as the Nivågård child. Moreover, we revisit the utility of stable carbon (δ13C), nitrogen (δ15N) and sulfur (δ34S) isotope analysis of human tissues to reconstruct the life histories and diets of 10 individuals from Nivå 10 as well as the Nivågård child. To contextualise further, we sampled 14 Capreolus capreolus and three Sus scrofa from the Nivågård site for stable isotope analysis. We demonstrate that sex can successfully be determined from contexts susceptible to poor organic preservation, and show that the Nivågård child spent a proportion of its life outside a sea spray-influenced environment, and consumed significant quantities of marine protein as demonstrated by its δ13C and δ34S values
