    Acquired resistance to classical chemotherapeutics is a major obstacle in cancer treatment. Doxorubicin is frequently used in breast cancer therapy either as single-agent or in combination with other drugs like docetaxel and cyclophosphamide. All these chemotherapies have in common that they are administered sequentially and often result in chemoresistance. Here, we mimicked this pulse therapy of breast cancer patients in an in vitro cell culture model, where the epithelial breast cancer cell line BT474 was sequentially treated with doxorubicin for several treatment cycles. In consequence, we obtained chemoresistant cells displaying a mesenchymal-like phenotype with decreased levels of miR-200c. To investigate the involvement of miR-200c in resistance formation, we inhibited and overexpressed miR-200c in different cell lines. Thereby, the cells were rendered more resistant or susceptible to doxorubicin treatment. Moreover, the receptor tyrosine kinase TrkB and the transcriptional repressor Bmi1 were identified as miR-200c targets mediating the drug resistance. Hence, we provide a mechanism of acquired resistance to doxorubicin that is caused by the loss of miR-200c. Along with this, our study demonstrates the complex network of microRNA mediated chemoresistance highlighting the challenges in cancer therapy and the importance of novel microRNA-modulating anticancer agents

    Nonlinear Integrable Ion Traps

    Quadrupole ion traps can be transformed into nonlinear traps with integrable motion by adding special electrostatic potentials. This can be done with both stationary potentials (electrostatic plus a uniform magnetic field) and with time-dependent electric potentials. These potentials are chosen such that the single particle Hamilton-Jacobi equations of motion are separable in some coordinate systems. The electrostatic potentials have several free adjustable parameters allowing for a quadrupole trap to be transformed into, for example, a double-well or a toroidal-well system. The particle motion remains regular, non-chaotic, integrable in quadratures, and stable for a wide range of parameters. We present two examples of how to realize such a system in case of a time-independent (the Penning trap) as well as a time-dependent (the Paul trap) configuration

    Ring for test of nonlinear integrable optics

    Nonlinear optics is a promising idea potentially opening the path towards achieving super high beam intensities in circular accelerators. Creation of a tune spread reaching 50% of the betatron tune would provide strong Landau damping and make the beam immune to instabilities. Recent theoretical work has identified a possible way to implement stable nonlinear optics by incorporating nonlinear focusing elements into a specially designed machine lattice. In this report we propose the design of a test accelerator for a proof-of-principle experiment.     Characterization of eDNA from the Clinical Strain Acinetobacter baumannii AIIMS 7 and Its Role in Biofilm Formation

    Release of extracellular DNA (eDNA) was observed during in vitro growth of a clinical strain of Acinetobacter baumannii. Membrane vesicles (MV) of varying diameter (20–200 nm) containing DNA were found to be released by transmission electron microscopy (TEM) and atomic force microscopy (AFM). An assessment of the characteristics of the eDNA with respect to size, digestion pattern by DNase I/restriction enzymes, and PCR-sequencing, indicates a high similarity with genomic DNA. Role of eDNA in static biofilm formed on polystyrene surface was evaluated by biofilm augmentation assay using eDNA available in different preparations, for example, whole cell lysate, cell-free supernatant, MV suspension, and purified eDNA. Biofilm augmentation was seen up to 224.64%, whereas biofilm inhibition was 59.41% after DNase I treatment: confirming that eDNA facilitates biofilm formation in A. baumannii. This is the first paper elucidating the characteristics and role of eDNA in A. baumannii biofilm, which may provide new insights into its pathogenesis

    Faecal sludge treatment by vermifiltration: proof of concept

    The objective of this study was to determine if composting worms and their cocoons are able to survive in and digest faecal sludge. Eighteen vermifilters with different worm and cocoon densities were set-up, fed with faecal sludge (from portable pour-flush toilets) and ran for 38 days. Samples of the sludge, effluent and vermicompost were analysed for pH, total solids, chemical oxygen demand, faecal coliforms and Ascaris spp. number and viability. Worm and vermicompost mass, and cocoon numbers were assessed at the end of this period. It was found that the composting worms survived in these conditions and cocoons hatched. The validity of the sludge analysis results are questioned in this paper and without these results only estimates of effluent treatment and solids conversion could be made. Although this study was not completely definitive it has shown that worms are capable of converting faecal sludge into vermicompost and cocoons hatch in its presence. This trial was undertaken in India

    Fast algorithm for calculating two-photon absorption spectra

    We report a numerical calculation of the two-photon absorption coefficient of electrons in a binding potential using the real-time real-space higher-order difference method. By introducing random vector averaging for the intermediate state, the task of evaluating the two-dimensional time integral is reduced to calculating two one-dimensional integrals. This allows the reduction of the computation load down to the same order as that for the linear response function. The relative advantage of the method compared to the straightforward multi-dimensional time integration is greater for the calculation of non-linear response functions of higher order at higher energy resolution.Comment: 4 pages, 2 figures.     Hepaticoduodenostomy as a technique for biliary anastomosis in children with choledochal cyst: An experience with 31 cases

    Objective The aim of this study was to investigate the efficacy and complications of hepaticoduodenostomy in the treatment of choledochal cyst in children. Summary background data The conventional treatment of choledochal cyst includes Roux-en-Y hepaticojejunostomy for biliary reconstruction. This procedure, however, disrupts normal bowel continuity and requires two anastomoses. We studied the technique of hepaticoduodenostomy as an effective alternative to this technique.Patients and methods A total of 31 children undergoing  hepatoduodenostomy for choledochal cyst over a period of 9 years were included in this study.Results The patients operated upon had outcomes similar to those treated by the Roux-en-Y technique in other studies.Conclusion Hepaticoduodenostomy is an effective alternative to the conventional Roux-en-Y anastomotic technique in cases of choledochal cyst in children.     Impact of co-application of biomethanated spent wash with bio-char on soil micronutrients and crop growth

    Biomethanated spentwash (BMS) and biochar are novel soil conditioners considered as value addition to the bi-products from agro-industry. The high nutrient content and organic matter of BMS and high retention of nutrient and organic carbon on biochar has potential of benefits for co-application. We studied the effect of BMS, biochar and their co-application on soybean, which is a common rotation crop in Sugarcane. A field experiment was conducted in randomized block design in vertisols. BMS was produced at Ugar sugar works ltd. and biochar from agriculture waste. Treatments comprised of BMS application@ 60, 66 and 72 m3 ha-1, as sole amendment and in combination with biochar @ 2.5 t ha-1. Post-harvest soil analysis demonstrated a non-significant effect of treatments on soil pH, electrical conductivity and micro-nutrients. However, available Zn, Fe and Mn were numerically increased for all BMS treatments. BMS and biochar or their co-application had no significant effect on germination and plant height of soybean. However the number of soybean pods and grain yield was significantly improved. Grain yield of soybean in BMS 60 m3 ha-1 plus biochar co-applied treatment was significantly higher than all other treatments. Grain yield of BMS 66 m3 ha-1 and other BMS plus BC treatments were at par with each other and significantly higher than sole biochar 2.5 t ha-1, RDF and control
