    The effects of eccentric and concentric resistance exercise on metabolic health

University of Missouri--Columbia 2010.The purpose of this study was to 1) compare the effects of single sessions of eccentric (ECC) and concentric (CON) contractions, and 2) determine the effects of 12 wks of ECC and CON resistance training (RT) on parameters of metabolic health. Thirty sedentary and obese individuals were randomly assigned to complete a single session of either CON (n=15) or ECC (n=15) resistance exercise consisting of 3 sets of 10 repetitions of bilateral leg extension and leg curl at 75% of CON or ECC one repetition maximum (1RMCON/1RMECC). Outcome measures were assessed at baseline (0 h) and 1, 24, 48, and 72 h post-exercise. Twenty-five of the 30 subjects also completed 12 wks of CON (n=13) or ECC (n=12) RT. Training consisted of whole-body RT 3-4 d/wk at 60-75% of 1RMCON or 1RMECC. Outcome measures were assessed pre- and post-training. The single session of ECC exercise produced significantly greater muscle degradation than CON exercise. The single session of exercise significantly reduced plasma TG and lipoproteins; however, there were no differences in these reductions between groups. No significant differences were found for plasma cytokines, glucose, or insulin. RT resulted in significant reductions in body fat%, waist circumference, diastolic blood pressure, and mid-thigh adipose tissue (AT) and significant increases in muscular strength, lean body mass, and mid-thigh total- and high density-muscle with no significant group differences. There were no significant changes in abdominal AT, plasma TG, lipoproteins, cytokines, glucose, or insulin following RT. Although a single session of ECC contractions resulted in greater skeletal muscle degradation than CON, the effects on plasma metabolic variables were similar in the two groups. Both CON and ECC RT produce beneficial alterations in metabolic health.Includes bibliographical reference

    Obsessive-Compulsive Disorder (OCD) is a debilitating, chronic condition characterized by recurrent intrusive thoughts followed by persistent behavioral urges and time-consuming rituals that attempt to neutralize anxiety and distress caused by those unwanted obsessive thoughts (American Psychiatric Association, 2013). This research study aims to explore the effectiveness of Couples-Based ERP in decreasing individual symptoms of OCD and evaluating the impact of treatment on relationship satisfaction. Using the Yale-Brown Obsessive Compulsive Scale (Y-BOCS) and the Dyadic-Adjustment Scale (DAS) to measure our constructs, we hypothesize that Couples-Based ERP will be effective in not only decreasing OCD symptoms, but also increasing overall relationship satisfaction

    Gaining S-T-E-A-M: A General Athletic Department Social Media Strategy

    In the 10 years since the invention of Facebook, social media sites have become an indispensable part of the marketing and communications strategy employed by a broad spectrum of organizations, including university athletic departments. While social media is almost universally used, a review of academic literature suggests the study of deployment of social media resources, and analysis of their effectiveness, is still very much in preliminary stages. Professional literature on social media use is out in front of peer-reviewed research. Therefore, we use Funk’s framework for social media practices as a point of departure, offering a social media strategy specifically for university athletic departments, grounded in Social Marketing Theory. Using a case study of Old Dominion University, a mid-sized, U.S. college athletic department, the authors analyze the 40 social media pages run by the department in comparison to guidelines created from the Funk framework and the growing body of academic literature, conduct interviews with practitioners in the athletic department, and a focus group of fans. Using this data, the authors create a case study-based list of best practices, known by the acronym S-T-E-A-M, which could assist similar university athletic departments in their use of social media

    An Examination of Post-Traumatic Stress Symptoms and Aggression Among Children with a History of Adverse Childhood Experiences

    Childhood aggression is associated with many deleterious outcomes and is a common reason for psychiatric referral (Card and Little 2006; Gurnani, Ivanov, and Newcorn 2016). One factor associated with childhood aggression is Adverse Childhood Experiences (ACEs; Felitti et al. 1998). However, existing research remains equivocal on which characteristics of ACEs (e.g., cumulative impact, typology, etc.) are significantly elated to aggression, especially when considering differential effects of ACEs on proactive aggression (PA) and reactive aggression (RA; Dodge and Coie, 1987). Post-traumatic stress symptoms (PTSS) are a common negative sequalae of ACEs and are characterized by disruptions in several cognitive, emotional, and behavioral processes similar to those associated with both RA and PA (e.g., Marsee 2008). As such, the examination of PTSS as an underlying mechanism of influence on the relation between ACEs, PA, and RA is warranted. The present study fills several gaps in the literature by examining ACE characteristics that might be related to PTSS, PA, and RA while also examining direct and indirect effects on the relation between ACEs, PTSS and PA and RA. Results indicated the type of ACE, specifically child maltreatment ACEs (CM-ACEs), was most strongly related to all outcome variables. Therefore, CM-ACEs were included in a path analysis with PTSS, PA, and RA. Results indicated a significant indirect effect for PTSS on the relation between CM-ACEs and RA (β = .18, p \u3c .01) but not PA. Findings have several implications for future research and clinical practice, especially for children with an extensive history of CM-ACEs

    Mitochondria-Penetrating Peptides

    SummaryMitochondria are important targets for cancer chemotherapy and other disease treatments. Gaining access to this organelle can be difficult, as the inner membrane is a barrier limiting diffusive transport. A mitochondrial molecular carrier would be a boon to the development of organelle-specific therapeutics. Here, we report a significant advance in the development of mitochondrial transporters—synthetic cell-permeable peptides that are able to enter mitochondria. Efficient uptake of these mitochondria-penetrating peptides (MPPs) is observed in a variety of cell types, and organellar specificity is attained with sequences that possess specific chemical properties. The MPPs identified are cationic, but also lipophilic; this combination of characteristics facilitates permeation of the hydrophobic mitochondrial membrane. The examination of a panel of MPPs illustrates that mitochondrial localization can be rationally controlled and finely tuned by altering lipophilicity and charge

    Pathways from Child Maltreatment to Proactive and Reactive Aggression: The Role of Post-Traumatic Stress Symptom Clusters

    Objective: Childhood aggression is related to a myriad of negative concurrent and long-term outcomes. To mitigate the risks associated with childhood aggression, it is important to understand risk factors that might predispose one to aggressive behaviors. One risk factor commonly associated with aggression is the experience of child maltreatment. A common outcome associated with child maltreatment is the development of post-traumatic stress symptoms (PTSS). Several prevailing theoretical models of both post-traumatic stress and aggression indicate that these constructs have similar underlying cognitive, behavioral, and emotional mechanisms. Therefore, the present study examined the relations between and among child maltreatment, PTSS clusters, and proactive and reactive aggression in children. Method: Children between the ages of 6 and 14 who were enrolled in a residential treatment program completed self-report measures to evaluate variables of interest. These variables were included as multiple outcomes in a path analysis model in which individual PTSS clusters were examined as potential multiple mediators of the relations between child maltreatment and proactive and reactive aggression. Results: Direct effects of child maltreatment and PTSS clusters on aggression were observed. Significant indirect effects of the intrusion PTSS cluster on the relation between child maltreatment and reactive aggression was found. Conclusions: Findings suggest that symptoms associated with these specific PTSS clusters might help explain the relation between child maltreatment and reactive aggression and therefore present important implications for clinical practice and future research

    Dopamine D2 receptor signaling on iMSNs is required for initiation and vigor of learned actions

    Striatal dopamine D2 receptors (D2Rs) are important for motor output. Selective deletion of D2Rs from indirect pathway-projecting medium spiny neurons (iMSNs) impairs locomotor activities in a task-specific manner. However, the role of D2Rs in the initiation of motor actions in reward seeking and taking is not fully understood, and there is little information about how receptors contribute under different task demands and with different outcome types. The iMSN-D2Rs modulate neuronal activity and synaptic transmission, exerting control on circuit functions that may play distinct roles in action learning and performance. Selective deletion of D2Rs on iMSNs resulted in slower action initiation and response rate in an instrumental conditioning task, but only when performance demand was increased. The iMSN-Drd2KO mice were also slower to initiate swimming in a T-maze procedural learning task but were unimpaired in cognitive function and behavioral flexibility. In contrast, in a Pavlovian discrimination learning task, iMSN-Drd2KO mice exhibited normal acquisition and extinction of rewarded responding. The iMSN-Drd2KO mice showed performance deficits at all phases of rotarod skill learning. These findings reveal that dopamine modulation through iMSN-D2Rs influences the ability to self-initiate actions, as well as the willingness and/or vigor with which these responses are performed. However, these receptors seem to have little influence on simple associative learning or on stimulus-driven responding. The loss of normal D2R roles may contribute to disorders in which impaired dopamine signaling leads to hypokinesia or impaired initiation of specific voluntary actions

    Advancing the speed, sensitivity and accuracy of biomolecular detection using multi-length-scale engineering

    Rapid progress in identifying disease biomarkers has increased the importance of creating high-performance detection technologies. Over the last decade, the design of many detection platforms has focused on either the nano or micro length scale. Here, we review recent strategies that combine nano- and microscale materials and devices to produce large improvements in detection sensitivity, speed and accuracy, allowing previously undetectable biomarkers to be identified in clinical samples. Microsensors that incorporate nanoscale features can now rapidly detect disease-related nucleic acids expressed in patient samples. New microdevices that separate large clinical samples into nanocompartments allow precise quantitation of analytes, and microfluidic systems that utilize nanoscale binding events can detect rare cancer cells in the bloodstream more accurately than before. These advances will lead to faster and more reliable clinical diagnostic devices

    Intercalative Stacking: A Critical Feature of DNA Charge-Transport Electrochemistry

    In electrochemistry experiments on DNA-modified electrodes, features of the redox probe that determine efficient charge transport through DNA-modified surfaces have been explored using methylene blue (MB^+) and Ru(NH_3)_6^(3+) as DNA-binding redox probes. The electrochemistry of these molecules is studied as a function of ionic strength to determine the necessity of tight binding to DNA and the number of electrons involved in the redox reaction; on the DNA surface, MB^+ displays 2e^-/1H^+ electrochemistry (pH 7) and Ru(NH^3)_6^(3+) displays 1e^- electrochemistry. We examine also the effect of electrode surface passivation and the effect of the mode (intercalation or electrostatic) of MB^+ and Ru(NH_3)_6^(3+) binding to DNA to highlight the importance of intercalation for reduction by a DNA-mediated charge-transport pathway. Furthermore, in experiments in which MB^+ is covalently linked to the DNA through a σ-bonded tether and the ionic strength is varied, it is demonstrated that intercalative stacking rather than covalent σ-bonding is essential for efficient reduction of MB^+. The results presented here therefore establish that efficient charge transport to the DNA-binding moiety in DNA films requires intercalative stacking and is mediated by the DNA base pair array

    Associations of Objectively Measured Light-Intensity Activity on Metabolic Risk Factors in Female Adults

    Metabolic syndrome (MetSy) has been defined as a collection of metabolic irregularities that increase the risk of cardiovascular disease, type 2 diabetes, and all-cause mortality. Women are at an even greater risk for developing MetSy when compared to men with prevalence increasing with age. Moderate to vigorous physical activity (MVPA) has shown consistent association with decreases in risk of MetSy; however, light physical activity (LPA) has been studied less often in comparison. PURPOSE: The purpose of this study was to examine relationships between accelerometer-determined LPA and MetSy risk factors (waist circumference (WC), glucose, high density lipoprotein (HDL), triglycerides, systolic blood pressure (SBP), and diastolic blood pressure (DPB)) across women of different age groups. METHODS: A cross-sectional study design was used to examine 68 female participants, ages 20-65, employed in sedentary careers working in an urban area in the south central United States. Venous blood samples were taken from the antecubital area of the arm from all participants who had fasted at least eight hours prior to their scheduled appointment times. Physical activity data was collected for seven days via hip-worn accelerometers with LPA being measured based on Freedson cut-points (100-1951 counts per minute). MetSy was diagnosed based on the criteria set forth by the National Cholesterol Education Program in 2002. Data were analyzed using regression analysis with SPSS version 20.0. Between-subject analysis was conducted for four age groups (group1=20-35; group2=36-45; group3=46-55; group4=55 and older) using LPA, WC, glucose, HDL, triglycerides, SBP, and DPB. RESULTS: Between-subject regression analysis showed HDL as a significant MetSy risk factor in a full model (p=0.02) and a model with age groups as the only independent variable (p=0.00). Post-hoc Tukey tests showed significance differences in HDL among groups 4 and 2 (confidence interval (CI): 4.03, 35.92; p=0.00), and groups 4 and 3 (CI: 6.06, 39.63; p=0.00). Significance dropped below the apriori level of 0.05 in a model based on LPA. Post-hoc Tukey tests revealed non-significant (α\u3e0.05) associations between LPA levels (high, medium, low) and all MetSy values. CONCLUSION: The relationship between LPA and MetSy risk factors has been studied less frequently than MVPA in relation to MetSy risk factors. Based on these findings LPA is not significantly related with these risk factors. Future studies should continue to explore the relationship between LPA and MetSy risk factors among both men and woman, and in different settings
