    Dietary carbohydrate source influences molecular fingerprints of the rat faecal microbiota

    BACKGROUND: A study was designed to elucidate effects of selected carbohydrates on composition and activity of the intestinal microbiota. Five groups of eight rats were fed a western type diet containing cornstarch (reference group), sucrose, potato starch, inulin (a long- chained fructan) or oligofructose (a short-chained fructan). Fructans are, opposite sucrose and starches, not digestible by mammalian gut enzymes, but are known to be fermentable by specific bacteria in the large intestine. RESULTS: Animals fed with diets containing potato starch, or either of the fructans had a significantly (p < 0.05) higher caecal weight and lower caecal pH when compared to the reference group, indicating increased fermentation. Selective cultivation from faeces revealed a higher amount of lactic acid bacteria cultivable on Rogosa agar in these animals. Additionally, the fructan groups had a lower amount of coliform bacteria in faeces. In the inulin and oligofructose groups, higher levels of butyrate and propionate, respectively, were measured. Principal Component Analysis of profiles of the faecal microbiota obtained by Denaturing Gradient Gel Electrophoresis (DGGE) of PCR amplified bacterial 16S rRNA genes as well as of Reverse Transcriptase-PCR amplified bacterial 16S rRNA resulted in different phylogenetic profiles for each of the five animal groups as revealed by Principal Component Analysis (PCA) of band patterns. CONCLUSION: Even though sucrose and cornstarch are both easily digestible and are not expected to reach the large intestine, the DGGE band patterns obtained indicated that these carbohydrates indeed affected the composition of bacteria in the large gut. Also the two fructans resulted in completely different molecular fingerprints of the faecal microbiota, indicating that even though they are chemically similar, different intestinal bacteria ferment them. Comparison of DNA-based and RNA-based profiles suggested that two species within the phylum Bacteroidetes were not abundant in numbers but had a particularly high ribosome content in the animals fed with inulin

    Effects of apples and specific apple components on the cecal environment of conventional rats: role of apple pectin

    <p>Abstract</p> <p>Background</p> <p>Our study was part of the large European project ISAFRUIT aiming to reveal the biological explanations for the epidemiologically well-established health effects of fruits. The objective was to identify effects of apple and apple product consumption on the composition of the cecal microbial community in rats, as well as on a number of cecal parameters, which may be influenced by a changed microbiota.</p> <p>Results</p> <p>Principal Component Analysis (PCA) of cecal microbiota profiles obtained by PCR-DGGE targeting bacterial 16S rRNA genes showed an effect of whole apples in a long-term feeding study (14 weeks), while no effects of apple juice, purĂ©e or pomace on microbial composition in cecum were observed. Administration of either 0.33 or 3.3% apple pectin in the diet resulted in considerable changes in the DGGE profiles.</p> <p>A 2-fold increase in the activity of beta-glucuronidase was observed in animals fed with pectin (7% in the diet) for four weeks, as compared to control animals (P < 0.01). Additionally, the level of butyrate measured in these pectin-fed animal was more than double of the corresponding level in control animals (P < 0.01). Sequencing revealed that DGGE bands, which were suppressed in pectin-fed rats, represented Gram-negative anaerobic rods belonging to the phylum <it>Bacteroidetes</it>, whereas bands that became more prominent represented mainly Gram-positive anaerobic rods belonging to the phylum <it>Firmicutes</it>, and specific species belonging to the <it>Clostridium </it>Cluster XIVa.</p> <p>Quantitative real-time PCR confirmed a lower amount of given <it>Bacteroidetes </it>species in the pectin-fed rats as well as in the apple-fed rats in the four-week study (P < 0.05). Additionally, a more than four-fold increase in the amount of <it>Clostridium coccoides </it>(belonging to Cluster XIVa), as well as of genes encoding butyryl-coenzyme A CoA transferase, which is involved in butyrate production, was detected by quantitative PCR in fecal samples from the pectin-fed animals.</p> <p>Conclusions</p> <p>Our findings show that consumption of apple pectin (7% in the diet) increases the population of butyrate- and ÎČ-glucuronidase producing <it>Clostridiales</it>, and decreases the population of specific species within the <it>Bacteroidetes </it>group in the rat gut. Similar changes were not caused by consumption of whole apples, apple juice, purĂ©e or pomace.</p

    Effect of gluon-exchange pair-currents on the ratio G(E(P))/G(M(P))

    The effect of one-gluon-exchange (OGE) pair-currents on the ratio ÎŒpGEp/GMp\mu_p G_E^p/G_M^p for the proton is investigated within a nonrelativistic constituent quark model (CQM) starting from SU(6)×O(3)SU(6) \times O(3) nucleon wave functions, but with relativistic corrections. We found that the OGE pair-currents are important to reproduce well the ratio ÎŒpGEp/GMp\mu_p G_E^p/G_M^p. With the assumption that the OGE pair-currents are the driving mechanism for the violation of the scaling law we give a prediction for the ratio ÎŒnGEn/GMn\mu_n G_E^n/G_M^n of the neutron.Comment: 5 pages, 4 figure

    Begleit- und Wirkungsforschung zur Hightech-Strategie : Ex-Post-Evaluierung der Fördermaßnahmen BioChance und BioChancePlus im Rahmen der Systemevaluierung

    Der vorliegende Bericht stellt die Ergebnisse der Evaluation der BMBF-Fördermaßnahmen BioChance und BioChancePlus dar. Beide zielten als VorlĂ€ufer der Förderinitiative "KMUinnovativ: Biotechnologie" darauf ab, innovative und anspruchsvolle Forschungsvorhaben von kleinen und mittleren Unternehmen in der Biotechnologie zu ermöglichen. Die beiden Fördermaßnahmen unterschieden sich in Förderziel und Design: WĂ€hrend BioChance ab 1999 darauf abzielte, neu gegrĂŒndete Firmen zu unterstĂŒtzen, ging es in BioChancePlus ab 2003 darum, die weitere Entwicklung junger Biotechnologie- Unternehmen und deren risikoreichen Projekte voranzutreiben. Die vorliegende Analyse zeigt, dass BioChance und BioChancePlus eine angemessene Reaktion auf die Schwierigkeiten waren, mit denen die dedizierten Biotechnologie-Unternehmen in Deutschland in den Jahren 1999-2009 zu kĂ€mpfen hatten. Die Fördermaßnahmen wurden in ihrem Design jeweils adĂ€quat weiterentwickelt und den Erfordernissen der Zielgruppe entsprechend angepasst. BioChance und BioChancePlus haben ihre Zielgruppen in hohem Maße erreicht: Von BioChance profitierten 15% der jungen Biotechnologiefirmen in Deutschland, der Nachfolger BioChancePlus erreichte 40% seiner Zielgruppe. Insgesamt erhielten 260 Unternehmen eine Zuwendung. 85% davon wurden nur einmal gefördert. Die öffentliche Förderung stellte eine wichtige, jedoch keineswegs die dominierende Finanzierungsquelle fĂŒr die Unternehmen dar. So flossen im Zeitraum 2000-2009 rund 3 Mrd. Euro an VC-Investitionen in die Biotechnologie-Branche, wĂ€hrend sich die öffentliche Förderung auf ca. 5% dieser Summe belief. Bei BioChance erhielten 17% der eingereichten AntrĂ€ge eine Förderung, bei BioChancePlus waren es 29%. Insgesamt wurden durch die Maßnahme BioChance etwa 36 Millionen Euro und durch BioChancePlus 133 Millionen Euro an Fördergeldern gewĂ€hrt

    VEGF preconditioning leads to stem cell remodeling and attenuates age-related decay of adult hippocampal neurogenesis

    Several factors are known to enhance adult hippocampal neurogenesis but a factor capable of inducing a long-lasting neurogenic enhancement that attenuates age-related neurogenic decay has not been described. Here, we studied hippocampal neurogenesis following conditional VEGF induction in the adult brain and showed that a short episode of VEGF exposure withdrawn shortly after the generation of durable new vessels (but not under conditions where newly made vessels failed to persist) is sufficient for neurogenesis to proceed at a markedly elevated level for many months later. Continual neurogenic increase over several months was not accompanied by accelerated exhaustion of the neuronal stem cell (NSC) reserve, thereby allowing neurogenesis to proceed at a markedly elevated rate also in old mice. Neurogenic enhancement by VEGF preconditioning was, in part, attributed to rescue of age-related NSC quiescence. Remarkably, VEGF caused extensive NSC remodelling manifested in transition of the enigmatic NSC terminal arbor onto long cytoplasmic processes engaging with and spreading over even remote blood vessels, a configuration reminiscent of early postnatal "juvenile" NSCs. Together, these findings suggest that VEGF preconditioning might be harnessed for long-term neurogenic enhancement despite continued exposure to an "aged" systemic milieu

    Big sugar in southern Africa : rural development and the perverted potential of sugar/ethanol exports

    This paper asks how investment in large-scale sugar cane production has contributed, and will contribute, to rural development in southern Africa. Taking a case study of the South African company Illovo in Zambia, the argument is made that the potential for greater tax revenue, domestic competition, access to resources and wealth distribution from sugar/ethanol production have all been perverted and with relatively little payoff in wage labour opportunities in return. If the benefits of agro-exports cannot be so easily assumed, then the prospective 'balance sheet' of biofuels needs to be re-examined. In this light, the paper advocates smaller-scale agrarian initiatives

    GeantV: Results from the prototype of concurrent vector particle transport simulation in HEP

    Full detector simulation was among the largest CPU consumer in all CERN experiment software stacks for the first two runs of the Large Hadron Collider (LHC). In the early 2010's, the projections were that simulation demands would scale linearly with luminosity increase, compensated only partially by an increase of computing resources. The extension of fast simulation approaches to more use cases, covering a larger fraction of the simulation budget, is only part of the solution due to intrinsic precision limitations. The remainder corresponds to speeding-up the simulation software by several factors, which is out of reach using simple optimizations on the current code base. In this context, the GeantV R&D project was launched, aiming to redesign the legacy particle transport codes in order to make them benefit from fine-grained parallelism features such as vectorization, but also from increased code and data locality. This paper presents extensively the results and achievements of this R&D, as well as the conclusions and lessons learnt from the beta prototype.Comment: 34 pages, 26 figures, 24 table

    Nucleon Charge and Magnetization Densities from Sachs Form Factors

    Relativistic prescriptions relating Sachs form factors to nucleon charge and magnetization densities are used to fit recent data for both the proton and the neutron. The analysis uses expansions in complete radial bases to minimize model dependence and to estimate the uncertainties in radial densities due to limitation of the range of momentum transfer. We find that the charge distribution for the proton is significantly broad than its magnetization density and that the magnetization density is slightly broader for the neutron than the proton. The neutron charge form factor is consistent with the Galster parametrization over the available range of Q^2, but relativistic inversion produces a softer radial density. Discrete ambiguities in the inversion method are analyzed in detail. The method of Mitra and Kumari ensures compatibility with pQCD and is most useful for extrapolating form factors to large Q^2.Comment: To appear in Phys. Rev. C.
