    Electrocatalytic performance of PbO2 films in the degradation of dimethoate insecticide

    This study was performed to find the best experimental conditions for the electrochemical removal of the insecticide dimethoate (C5H12NO3PS2) from aqueous solutions using a lead dioxide niobium anode. The process was studied under galvanostatic polarisation mode. The influence of applied current density (10–50 mA·cm–2), initial chemical oxygen demand COD0 (100–550 mg·ℓ–1), temperature (30–70°C) and pH (3–11) on COD and instantaneous current efficiency (ICE) was studied. The results showed that almost 90% of COD removal was achieved under optimal experimental conditions, indicating that electrochemical oxidation on a PbO2 anode is a suitable method for treatment of water polluted with dimethoate. It was found that the decay of COD generally followed a pseudo first-order kinetic and the oxidation rate was favoured by increasing the applied current density, temperature, pH and initial COD. The greatest COD removal (90%) was obtained when using an applied current density of 50 mA·cm–2, COD0 = 320 mg·ℓ–1, pH = 11, T = 70°C and electrolysis time = 8 h.Keywords: Electrochemical degradation; hydroxyl radicals; organic pollutants; lead dioxide; wastewate

    Growth factor concentrations and their placental mRNA expression are modulated in gestational diabetes mellitus: possible interactions with macrosomia

    <p>Abstract</p> <p>Background</p> <p>Gestational diabetes mellitus (GDM) is a form of diabetes that occurs during pregnancy. GDM is a well known risk factor for foetal overgrowth, termed macrosomia which is influenced by maternal hypergycemia and endocrine status through placental circulation. The study was undertaken to investigate the implication of growth factors and their receptors in GDM and macrosomia, and to discuss the role of the materno-foeto-placental axis in the <it>in-utero </it>regulation of foetal growth.</p> <p>Methods</p> <p>30 women with GDM and their 30 macrosomic babies (4.75 ± 0.15 kg), and 30 healthy age-matched pregnant women and their 30 newborns (3.50 ± 0.10 kg) were recruited in the present study. Serum concentrations of GH and growth factors, <it>i.e</it>., IGF-I, IGF-BP3, FGF-2, EGF and PDGF-B were determined by ELISA. The expression of mRNA encoding for GH, IGF-I, IGF-BP3, FGF-2, PDGF-B and EGF, and their receptors, <it>i.e</it>., GHR, IGF-IR, FGF-2R, EGFR and PDGFR-ÎČ were quantified by using RT-qPCR.</p> <p>Results</p> <p>The serum concentrations of IGF-I, IGF-BP3, EGF, FGF-2 and PDGF-B were higher in GDM women and their macrosomic babies as compared to their respective controls. The placental mRNA expression of the growth factors was either upregulated (FGF-2 or PDGF-B) or remained unaltered (IGF-I and EGF) in the placenta of GDM women. The mRNA expression of three growth factor receptors, <it>i.e</it>., IGF-IR, EGFR and PDGFR-ÎČ, was upregulated in the placenta of GDM women. Interestingly, serum concentrations of GH were downregulated in the GDM women and their macrosomic offspring. Besides, the expression of mRNAs encoding for GHR was higher, but that encoding for GH was lower, in the placenta of GDM women than control women.</p> <p>Conclusions</p> <p>Our results demonstrate that growth factors might be implicated in GDM and, in part, in the pathology of macrosomia via materno-foeto-placental axis.</p

    Towards quality analysis for document oriented bases

    International audienceDocument-oriented bases allow high flexibility in data representation which facilitates a rapid development of applications and enables many possibilities for data structuring. Nevertheless, the structural choices remain crucial because of their impact on several aspects of the document base and application quality, e.g, memory print, data redundancy, readability and maintainability. Our research is motivated by quality issues of document-oriented bases. We aim at facilitating the study of the possibilities of data structuring and providing objective metrics to better reveal the advantages and disadvantages of each solution with respect to user needs. In this paper, we propose a set of structural metrics for a JSON compatible schema abstraction. These metrics reflect the complexity of the structure and are intended to be used in decision criteria for schema analysis and design process. This work capitalizes on experiences with MongoDB, XML and software complexity metrics. The paper presents the definition of the metrics together with a validation scenario where we discuss how to use the results in a schema recommendation perspective

    Genetic Heterogeneity of 72 Patients With Mucopolysaccharidosis In Tunisia

    The mucopolysaccharidoses (MPS) are a group of lysosomal storage diseases, presenting with a progressive multisystem disorder, with extreme clinical heterogeneity. The purpose of this study was to investigate six genes involved in different types of mucopolysaccharidosis (MPS), a group of lysosomal storage diseases. Mutation screening was performed for a total of 72 MPS Tunisian patients with MPS I (n=43), MPS II (n=5), MPS IIIA (n=7), MPS IIIB (n=3), MPS IIIC (n=2), or MPS IVA (n=12). The methodological approaches included genomic PCR sequencing, RT-PCR for MPS IVA and tetra-primer ARMS PCR assay for MPSI. The present study revealed one novel splice site mutation in one MPSI patient, in addition to the 11 previously Tunisian mutations reported for the first time by our research team: three in the IDUA gene (MPS I) including two missense, one nonsense four in the SGSH gene (MPS IIIA) including a mutation involving the start codon, one small duplication, one small deletion and a large deletion of exons 1 to 5 two in the NAGLU gene (MPS IIIB) including one missense mutation and one nonsense mutation two in the HGSNAT gene (MPS IIIC) including one missense and one nonsense mutation, two in the GALNS gene including one missense and one splicing mutation. These data demonstrate the remarkable mutational heterogeneity characterizing all types of MPS tested, although high consanguinity rate in the Tunisian population. We report biochemical and molecular aspects of these patients to prevent (e.g. genetic counseling, prenatal diagnosis) the recurrence of affected child since these diseases are still not rare in our area and as specific therapy is not available in our country. nbs

    Purification and characterization of a novel angiotensin I-converting enzyme-inhibitory peptide derived from Alaska pollack skins

    Hydrolysates containing angiotensin I-converting enzyme (ACE)-inhibitory peptide were prepared from protein of Alaska pollack skins using alcalase and trypsin. The protein hydrolysate was separated by ultrafiltration, Sephadex G-25 gel filtration chromatography and reversed phase high-performance liquid chromatography (HPLC), from which a novel purified peptide was obtained. Both random coil structure and beta-sheet in the purified peptide were revealed in Fourier transform infrared spectrum. The amino sequence of the purified peptide was identified as GPLGVP, VLYPVK, VFLENVLR, and FEEF by HPLC-Q-TOF-MS (HPLC-quadrupole time-of-flight mass spectrometry). The peptide GPLGVP whose molecular weight was 538.31 Da showed the highest ACE inhibitory activity (IC50 = 105.8 mu M). The purified peptide featured a noncompetitive inhibition kinetic mechanism was shown in the Lineweaver-Burk plots and was susceptible to enzymes as indicated in the studies on stability of gastrointestinal proteases. Moreover, the peptide GPLGVP can combine ACE catalytic pocket through hydrogen bonds and other forces with high binding power as disclosed in molecular docking simulation, which provides the inhibitory effect of GPLGVP on ACE
