    From canter to cantor: Negotiating constraints, and the perceptions of elitism in serious leisure pursuits : The experiences of a high performing athlete and artist

    Everyone experiences leisure differently but, for people who excel in a chosen field, a hobby can become a serious goal-oriented leisure pursuit. Many talented people, however, fail to reach their leisure goals due to constraints. This study explored individual life experiences of serious leisure participants. It focused on the lived experiences of individual event [horse] riders and opera singers who successfully negotiated their constraints, enabling them to reach their high performance goals. The purpose of this study was to explore positive personal strategies that individuals used to negotiate constraints in serious leisure. This was done by exploring intrapersonal, interpersonal and structural leisure constraints using an individual’s life experiences. During the process of this study, elitism in the leisure pursuits of event riding and opera singing, and the perceptions about individuals who participate in these activities, were also explored. A multidisciplinary approach was used in this study using a two stage mixed research methodology. The first stage explored the lived experiences of two individuals through a series of in-depth case study interviews, followed by interviews with their parents and coaches. Focus groups followed to establish if a wider group of participants within the same leisure pursuits experienced similar findings. The second stage of the study used a quantitative method, which consisted of a broader national survey. The survey data validated the qualitative findings and strengthened study outcomes. The findings of this study related to the opportunity-seeking skills an individual develops throughout their leisure life. These opportunity-seeking skills were linked to the likes, needs and wants an individual must have to reach a high performance level. It also found four principal ‘C Factors’ important to individual decision-making processes: conditioning, change, choice and control. Research findings revealed that early support and encouragement, however small, conditioned and motivated individuals to start and continue in a particular leisure activity. It also showed that those who had the ability to improve their talent, who personally believed in themselves, who viewed difficulties or complicated situations positively, and sought opportunities to enhance their leisure goals and change constraint outcomes, continued to succeed. Individuals had to make choices to enable them to control their goal achievement and deal with constraints throughout their leisure life. High performance success was found to be related to superior opportunity-seeking skills. Constraints arising from perceptions of elitism within serious leisure pursuits were found to be based on an individual’s life experiences and societal opinions, and not on the actual activity itself. In this study the strength of an individual’s motivation and self-belief, had a direct influence on their perceptions of constraints, and how they personally used opportunity-seeking to negotiate these constraints. Although the ability to predict which athletes or artists will become national or world class is limited, the conceptual framework developed in this study based on successful constraint negotiation strategies, could aid individuals wishing to reach a high performance level, and guide their parents and coaches to provide optimal support

    OBJECTIVE: To examine the role of interferon regulatory factor 3 (IRF-3) in the regulation of interleukin-23 (IL-23) production in patients with systemic lupus erythematosus (SLE). METHODS: Bone marrow-derived macrophages were isolated from both wild-type and IRF3(-/-) C57BL/6 mice. These cells were stimulated with the Toll-like receptor 3 (TLR-3) agonist poly(I-C), and IL-23p19 cytokine levels were analyzed by enzyme-linked immunosorbent assay. IRF-3 binding to the IL-23p19 gene promoter region in monocytes from patients with SLE and healthy control subjects was analyzed by chromatin immunoprecipitation (ChIP) assay. Luciferase reporter gene assays were performed to identify key drivers of IL-23p19 promoter activity. TANK-binding kinase 1 (TBK-1) protein levels were determined by Western blotting. RESULTS: ChIP assays demonstrated that IRF-3 was stably bound to the human IL-23p19 promoter in monocytes; this association increased following TLR-3 stimulation. Patients with SLE demonstrated increased levels of IRF-3 bound to the IL-23p19 promoter compared with control subjects, which correlated with enhanced IL-23p19 production in monocytes from patients with SLE. Investigations of the TLR-3-driven responses in monocytes from patients with SLE revealed that TBK-1, which is critical for regulating IRF-3 activity, was hyperactivated in both resting and TLR-3-stimulated cells. CONCLUSION: Our results demonstrate for the first time that patients with SLE display enhanced IL-23p19 expression as a result of hyperactivation of TBK-1, resulting in increased binding of IRF-3 to the promoter. These findings provide novel insights into the molecular pathogenesis of SLE and the potential role for TLR-3 in driving this response

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Women in the Information Technology Industry: A Western Australian View

    The information technology [IT] industry is one of the fastest growing sectors of the economy, both here in Australia and globally. This paper is an exploratory study of aspects of women’s status in the IT industry in Western Australia. What is apparent from this initial study is that the shortage of skilled personnel in WA mirrors the declining trend in the representation of women in the IT industry. This industry, once thought of as a gender neutral profession is still dominated by men. This preliminary study suggests that even though the IT industry does not exclude women, it does little to promote or employ them and the old corporate \u27boys club\u27 syndrome continues to perpetuate

    The use of IT is pervasive in all work and leisure and its importance cannot be underestimated. Higher educational institutes are increasingly marketing new IT courses - but are they attracting the students? Today there is a mix of generations within the IT industry, from Baby Boomers, Generation X’ers to the new work generation, Generation Y. This paper will discuss results from a study of university students reviewing different generations and genders and their perceptions of the IT industry. It would appear that not only are women not attracted to I.T. courses, but Generation Y students are unlikely to choose IT as a career path. The implication of these findings, is that the skills shortage problem will not be solved by the next generation of workers, generation Y, as both males and females cannot identify with careers in I.T

    Resistance to HER2-targeted anti-cancer drugs is associated with immune evasion in cancer cells and their derived extracellular vesicles

    Neuromedin U (NmU) -a neuropeptide belonging to the neuromedin family? plays a substantial role in HER2-positive breast cancer, correlating with increased aggressiveness, resistance to HER2-targeted therapies and overall significantly poorer outcome for patients. However, the mechanism through which it exerts these effects remains unclear. To elucidate this, initially we used HER2-positive breast cancer cells stably over-expressing NmU. These cells and their released extracellular vesicles (EVs) had increased amounts of the immunosuppressive cytokine TGF?1 and the lymphocyte activation inhibitor PD-L1. Furthermore, these cells also showed enhanced resistance to antibody-dependent cell cytotoxicity (ADCC) mediated by trastuzumab, indicating a role of NmU in enhancing immune evasion. All these features were also found in HER2-targeted drug-resistant cells which we previously found to express higher levels of NmU than their drug-sensitive counterparts. Interestingly, EVs from drug-resistant cells were able to increase levels of TGF?1 in drug-sensitive cells. In our neo-adjuvant clinical trial, TGF?1 levels were significantly higher in EVs isolated from the serum of patients with HER2-overexpressing breast cancers who went on to not respond to HER2-targeted drug treatment, compared with those who experienced complete or partial response. Taken together, our results report a new mechanism-of-action for NmU in HER2-overexpressing breast cancer that enhances resistance to the anti-tumor immune response. Furthermore, EV levels of TGF?1 correlating with patients\u27 response versus resistance to HER2-targeted drugs suggests a potential use of EV-TGF?1 as a minimally-invasive companion diagnostic for such treatment in breast cancer
