    Interference of kallikrein 1b26 (klk1b26) translation by microRNA specifically expressed in female mouse submandibular glands: an additional mechanism for sexual dimorphism of klk1b26 protein in the glands

    <p>Abstract</p> <p>Background</p> <p>Mouse kallikrein 1b26 (klk1b26) protein is more abundant in male submandibular glands (SMGs) than in female ones. This sexual dimorphism has been thought to be due to increased mRNA synthesis stimulated by androgen. However, the klk1b26 protein level in female SMG is far less than that expected from the mRNA level, suggesting an additional mechanism for down-regulation of klk1b26 expression in female SMGs.</p> <p>Methods</p> <p>We examined the effects of small non-coding RNAs in mouse SMGs on <it>in vitro </it>translation of klk1b26 using a reticulocyte lysate system and reverse transcription (RT)-PCR for klk1b26 mRNA. Statistical analyses were performed with a computer package (Microsoft Excel).</p> <p>Results</p> <p>The microRNA (miRNA) preparation from female SMGs, but not male SMGs, interfered with the <it>in vitro </it>translation of the klk1b26 protein and inhibited the RT-PCR for klk1b26 mRNA with forward primers targeting its 5'-terminal region (between the 15th and 40th nucleotide from the 5'-terminal). The miRNA preparation from castrated mouse SMGs showed the inhibitory effect on the klk1b26 translation, but that from a 5α-dihydrotestosterone-treated female mouse SMGs did not. Synthetic miRNAs (miR-325 and miR-1497a), which have partial complementarity with klk1b26 mRNA at its 5'-terminal region (15th to 40th nucleotide position from the 5'-terminal), also interfered with the <it>in vitro </it>klk1b26 translation. When the female miRNA preparation was incubated with a 30-nucleotide-long single-strand oligoDNA (named [15th-44th]ssDNA, whose sequence corresponded to the 15th to 44th position from the 5'-terminal of klk1b26 mRNA) prior to the addition into the <it>in vitro </it>translation system, the inhibitory effect of the miRNA preparation on klk1b26 translation disappeared, while [15th-44th]ssDNA itself had no effect on the translation. Preincubation of the miRNA preparation with another single-strand DNA ([169th-198th]ssDNA, whose sequence corresponded with 169th to 198th position of klk1b26 mRNA) did not show the inhibitory effect.</p> <p>Conclusions</p> <p>The small non-coding RNA, most probably miRNA, specifically expressed in female mouse SMGs interfered with klk1b26 protein synthesis in the <it>in vitro </it>translation system. Therefore sexual dimorphism observed in klk1b26 expression in mouse SMGs is due at least in part to the female-specific small non-coding RNA in SMGs.</p

    Compact tree plus algorithms for application-level multicast communications in multihome networks

    Application-level multicast (ALM) communications replicate packets on host level to deliver them from a single source to multiple clients, so that it can efficiently realize a variety of network applications using moving pictures such as video conferences, distance learning, and video-on-demands. In this paper, we propose the CT+ (compact tree plus) algorithm for finding a better ALM routing tree in terms of delay minimization between hosts. CT+ consists of a tree construction stage from the existing CT algorithm, and a newly added iterative tree improvement stage. Then, we define the extended ALM routing problem and its heuristic algorithm ExCT+, to optimize the effectiveness of the multihome network in ALM communications by selecting multihomed hosts and connections in the ALM routing tree simultaneously. For their evaluations, we construct a network simulation model named MINET (multiple-ISP network simulator), where the topology is composed of multiple ISP backbone networks with IX connections, and the network traffic is generated by following the M/M/1 queuing process. The simulation results using MINET verify the effectiveness of our algorithms.</p

    Optical-drop wavelength assignment problem for wavelength reuse in WDM ring metropolitan area networks

    This paper presents a formulation of the optical-drop wavelength assignment problem (ODWAP) and its heuristic algorithm for WDM ring networks. The wavelength-division multiplexing (WDM) technology has been popular in communication societies for providing very large communication bands by multiple lightpaths with different wavelengths on a single optical fiber. Particularly, a double-ring optical network architecture based on the packet-over-WDM technology such as the HORNET architecture has been studied as a next generation platform for metropolitan area networks (MANs). Each node in this architecture is equipped with a wavelength-fixed optical-drop and a tunable transmitter so that a lightpath can be established between any pair of nodes without wavelength conversions. In this paper, we formulate ODWAP for efficient wavelength reuse under heterogeneous traffic in this network. Then, we propose a simple heuristic algorithm for ODWAP. Through extensive simulations, we demonstrate the effectiveness of our approach in reducing waiting times for packet transmissions when a small number of wavelengths are available to retain the network cost for MANs

    A minimal-state processing search algorithm for satisfiability problems

    The satisfiability problem (SAT) is a typical NP-complete problem where a wide range of applications has been studied. Given a set of variables U and a set of clauses C, the goal of SAT is to find a truth assignment to variables in U such that every clause in C is satisfied if it exits, or to derive the infeasibility otherwise. This paper presents an approximation algorithm, called a minimal-state processing search algorithm for SAT (MIPS-SAT). MIPS-SAT repeatedly transits minimal states in terms of the cost function for searching a solution through a construction stage and a refinement stage. The first stage greedily generates an initial state composed of as many satisfied clauses as possible. The second stage iteratively seeks a solution while keeping state minimality.     IL-18 with IL-2 protects against Strongyloides venezuelensis infection by activating mucosal mast cell–dependent type 2 innate immunity

    C57BL/6 (B6) and B6 background STAT6−/− mice pretreated with IL-18 plus IL-2 showed prominent intestinal mastocytosis and rapidly expelled implanted adult worms of the gastrointestinal nematode Strongyloides venezuelensis. In contrast, identically pretreated mast cell–deficient W/Wv mice failed to do so. Thus, activated mucosal mast cells (MMC) are crucial for parasite expulsion. B6 mice infected with S. venezuelensis third-stage larvae (L3) completed parasite expulsion by day 12 after infection, whereas IL-18−/− or IL-18Rα−/− B6 mice exhibited marked impairment in parasite expulsion, suggesting a substantial contribution of IL-18–dependent MMC activation to parasite expulsion. Compared with IL-18−/− or IL-18Rα−/− mice, S. venezuelensis L3–infected STAT6−/− mice have poorly activated MMC and sustained infection; although their IL-18 production is normal. Neutralization of IL-18 and IL-2 further reduces expulsion in infected STAT6−/− mice. These results suggest that collaboration between IL-18–dependent and Th2 cell–dependent mastocytosis is important for prompt parasite expulsion

    Novel Hybrid Hydroxyapatite Spacers Ensure Sufficient Bone Bonding in Cervical Laminoplasty

    Study Design Prospective observational study. Purpose This prospective analysis aimed to evaluate the efficacy and bone-bonding rate of hybrid hydroxyapatite (HA) spacers in expansive laminoplasty. Overview of Literature Various types of spacers or plates have been developed for expansive laminoplasty. Methods Expansive open-door laminoplasty was performed in 146 patients with cervical myelopathy; 450 hybrid HA spacers and 41 autogenous bone spacers harvested from the spinous processes were grafted into the opened side of each lamina. The patients were followed up using computed tomography (CT), and their bone-bonding rates for hybrid HA and autogenous spacers, bone-fusion rates of the hinges of the laminae, and complications associated with the implants were then examined. Results Clinical symptoms significantly improved in all patients, and no major complications related to the procedure were noted. The hybrid HA spacers exhibited sufficient bone bonding on postoperative CT. The hinges completely fused in over 95% patients within 1 year of the procedure. Only 4 spacers (0.9%) developed lamina sinking, and most expanded laminae maintained their positions without sinking or floating throughout the follow-up period. Conclusions Hybrid HA spacers contributed to high bone-fusion rates of the spacers and hinges of the laminae, and no complications were associated with their use. Cervical laminoplasty with these spacers is safe and simple, and it yields sufficient fixation strength while ensuring sufficient bone bonding during the immediate postoperative period
