    Practical uses of genetic profile assessment in athletic training – an illustrative case study

    Recent studies suggested that several potential genes may explain athletic success. However, while genetic assessment will probably become part of future talent identification, at present, genetic testing predictive value is poor, mainly because athletic success depends on a combination of genetic, physiological, behavioral and environmental factors (including coaching, medical, nutritional, psychological, equipment, facilities and administrative aspects). However, one should consider genetic testing not only for talent identification or sport event selection, but also for possible assistance in the training process itself. In the present case study we show an example of potential practical use of genetic profile assessment for improving the athletic training process. We deliberately chose a case study of a national-level athlete to show that genetic aid should not be limited to top world-class athletes

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Brief memory reactivations induce learning in the numeric domain

    Learning of arithmetic facts such as the multiplication table requires time-consuming, repeated practice. In light of evidence indicating that reactivation of encoded memories can modulate learning and memory processes at the synaptic, system and behavioral levels, we asked whether brief memory reactivations can induce human learning in the numeric domain. Adult participants performed a novel number-fact retrieval task in which they learned arbitrary numeric facts. Following encoding and a baseline test, 3 passive, brief reactivation sessions of only 40 seconds each were conducted on separate days. Learning was evaluated in a retest session. Results showed reactivations induced learning, with improved performance at retest relative to baseline test. Furthermore, performance was superior compared to a control group performing test-retest sessions without reactivations, who showed significant memory deterioration. A standard practice group completed active-retrieval sessions on 3 separate days, and showed significant learning gains. Interestingly, while these gains were higher than those of the reactivations group, subjects showing reactivation-induced learning were characterized by superior efficiency relative to standard practice subjects, with higher rate of improvement per practice time. A follow-up long-term retention experiment showed that 30 days following initial practice, weekly brief reactivations reduced forgetting, with participants performing superior to controls undergoing the same initial practice without reactivations. Overall, the results demonstrate that brief passive reactivations induce efficient learning and reduce forgetting within a numerical context. Time-efficient practice in the numeric domain carries implications for enhancement of learning strategies in daily-life settings, including in learning disorders caused by neurological conditions

    The Wingate anaerobic test cannot be used for the evaluation of growth hormone secretion in children with short stature

    Purpose: To assess the growth hormone (GH) response to the Wingate anaerobic test (WAnT) among children with short stature and suspected GH deficiency. We hypothesized that the GH response to the WAnT would be similar to the GH response to a commonly used pharmacologic provocation test. Methods: Ten children (6 males and 4 females, age range 9.0–14.9 years) participated in the study. Each participant performed 2 tests: a standard all-out WAnT, cycling for 30 s against constant resistance, and a standardized pharmacologic test (clonidine or glucagon). Blood samples for GH were collected before and 10, 30, 45, and 60 min after the beginning of exercise. In addition, we collected pre- and post-exercise blood lactate levels. Results: There was a significant increase in GH levels after the WAnT, yet in 9 of 10 participants, this increase was below the threshold for GH sufficiency. Peak GH after the WAnT was significantly lower compared to the pharmacologic GH provocation tests (with 9 of 10 demonstrating GH-sufficient response). Conclusion: The traditional WAnT cannot be used as a GH provocation test. Further research is needed to develop anaerobic exercise protocols sufficient to promote GH secretion

    Imbalance in Sirt1 Alternative Splicing in Response to Chronic Stress during the Adolescence Period in Female Mice

    Stressful unpredictable life events have been implicated in numerous diseases. It is now becoming clear that some life periods are more vulnerable than others. As adolescence is a sensitive period in brain development, the long-term effects of stress during this period could be significant. We investigated the long-term effects of exposure to unpredictable chronic mild stress in adolescent mice on alternative splicing of Sirtuin 1. One-month-old mice were exposed to 4 weeks of UCMS and examined for anxiety and cognition at the age of 2, 4 and 6 months. We found a rise in anxious behavior immediately after the exposure to stress. Notably, there was a long-term impairment of performance in cognitive tasks and an imbalance in Sirtuin 1 and TrkB receptor alternative splicing in the stress-exposed mice compared with controls. To conclude, our results show that exposure to unpredictable chronic mild stress during adolescence affects cognition in adulthood. Understanding pathways affiliated with stress may help minimize the long-term emotional effects of an unpredictable, stressful event
