    A Granger Causality Analysis of Groundwater Patterns over a Half-Century

    Groundwater depletion in many areas of the world has been broadly attributed to irrigation. However, more formal, data-driven, causal mechanisms of long-term groundwater patterns have not been assessed. Here, we conducted the first Granger causality analysis to identify the causes of groundwater patterns using the rice-producing parishes of Louisiana, USA, as an example. Trend analysis showed a decline of up to 6 m in groundwater level over 51 years. We found that no single cause explained groundwater patterns for all parishes. Causal linkages were noted between groundwater and area harvested, number of irrigation wells, summer precipitation totals, and drought. Bi-directional linkages were noted between groundwater and rice yield, suggesting feedback between both time series. Causal linkages were absent between groundwater and many drivers where significant correlations were noted, highlighting the importance of using robust causal relationships over illusive correlations to detect the cause. These results advance our understanding of groundwater dynamics and can reveal a key connection between food and groundwater

    A Bayesian Framework to Unravel Food, Groundwater, and Climate Linkages: A Case Study from Louisiana

    Advancing our understanding of the connections among groundwater, food, and climate is critical to meet global food demands while optimizing water resources usage. However, our understanding of the linkages among groundwater, food, and climate is still limited. Here, we offer a Bayesian framework to simulate crop yield at a regional scale and quantify its relationships and associated uncertainty with climate, groundwater, agricultural, and energy-related variables. We implemented the framework in the rice-producing regions of Louisiana from 1960-2015. To build a parsimonious model, we used a probability-based variable selection approach to detect the key drivers of rice yield. Rice yield increased, groundwater declined, and area planted declined or did not change over 56yrs. The number of irrigation wells, groundwater level, air temperature, and area planted were found to be the key drivers of rice yield. The regression coefficients showed that rice yield was positively related to groundwater level, and negatively related to area planted and the number of irrigation wells. The limited influence of N fertilizer was noted on rice yield for the period when fertilizer data were available. The inverse relationship between rice yield and area planted pointed to the adaption of efficient crop management practices that maintained or increased yield, despite the decline in area planted. The farmers\u27 ability to install irrigation wells during droughts sustained the yields over long-term but not short-term. This decline in rice yield in response to drought over the short-term might explain the negative relation between yield and irrigation wells. Overall, this work highlighted the uncertainty in relationships between rice yield and key drivers and quantified the intimate connection between food and groundwater. This work may have implications for managing two highly competing commodities (i.e., groundwater and food) in agricultural regions

    Investigation of the Effect of Built Orientation on Mechanical Properties and Total Cost of FDM Parts

    AbstractFused deposition modeling (FDM) is one of the rapid prototyping methods that produce prototypes from plastic materials such as acrylonitrile butadiene styrene (ABS) by laying tracks of semi-molten plastic filament onto a platform in a layer wise manner from bottom to top. In FDM, one of the critical factor is to select the build up orientation of the model since it affects the different areas of the model like main material, support material, built up time, total cost per part and most important the mechanical properties of the part. In view of this, objective of the present study was to investigate the effect of the built-up orientation on the mechanical properties and total cost of the FDM parts. Experiments were carried out on STRATASYS FDM type rapid prototyping machine coupled with CATALYST software and ABS as main material. Tensile and Flexural specimens were prepared as per the ASTM standard with different built-up orientation and in three geometrical axes. It can be concluded from the experimental analysis that built orientation has significant affect on the tensile, flexural and total cost of the FDM parts. These conclusions will help the design engineers to decide on proper build orientation, so that FDM parts can be fabricated with good mechanical properties at minimum manufacturing cost

    Molecular Characterization of Heat Shock Protein 70-1 Gene of Goat (Capra hircus)

    Heat shock protein 70 (HSP 70) plays a vital role by bestowing cytoprotection against diverse kinds of stresses. The ubiquitous HSP 70 proteins are the most abundant and temperature sensitive among all the HSPs. The present paper has characterized HSP70-1 cDNA in goat (Capra hircus). Total RNA isolated from goat peripheral blood mononuclear cells was reverse transcribed to cDNA that was used for amplification of HSP 70-1 gene. PCR product (1926 bp) was cloned in pGEM-T easy vector and sequenced. Sequence analysis revealed 1926-bp-long open reading frame of HSP 70-1 gene encoding 641 amino acids in goat, as reported in cattle. At nucleotide level, goat HSP 70-1 was found to be 96–99% similar to that of sheep (partial), cattle, and buffalo whereas the similarity at amino acid level was 95–100%. Nonsynonymous substitutions exceeding synonymous substitutions indicate the evolution of this protein through positive selection among domestic animals. Goat and sheep appear to have diverged from a common ancestor in phylogenetic analysis. Predicted protein structures of goat HSP 70 protein obtained from deduced amino acid sequence indicated that the functional amino acids involved in chaperoning through ATPase hydrolytic cycle and in uncoating of clathrin coated vesicles are highly conserved

    Anti-nociceptive effect of seed extract of Acacia tortilis in rodents

    Background: Management of pain is a primary clinical concern for any pathology in medical field. Addiction liability of opioids and troublesome gastrointestinal side effects of NSAIDs   leads to intensive research for compound with lesser side effects.The aim of the study to evaluate the anti-nociceptive activity of Acacia Tortilis Seed Extract (ATE) in experimental animals.Methods: First of all, animals were randomly allocated into four groups of six animals each. In acetic acid induced writhing test model, Group I (NC) served as vehicle control received saline/Tween 80 0.1%, 10ml/kg BW orally, group II (ATE-100) and III (ATE-200) received ATE in dose of 100 and 200mg/kg BW orally respectively and group IV received the standard drug diclofenac sodium in dose of 50 mg/kg BW orally. Group I to IV were same in rest of three experimental models. One additional group of standard drugs (group V) morphine sulfate in dose of 5 mg/kg BW subcutaneously (SC) was allocated for screening method hot plate and tail flick tests.  In Formalin induced paw licking test, three additional groups (group V) morphine sulfate in dose of 5mg/kg BW SC, group VI- morphine+naloxone (5mg/kg SC +2mg/kg intra-peritoneally (IP) and group VII - ATE+ naloxone (200mg/kg BW orally +2mg/kg BW IP) were also made.Results: The ATE when administered orally in dose of 100 and 200mg/ kg body weight (BW), produced significant analgesic activity (P <0.01) in acetic acid induced writhing syndrome and late phase of formalin test. In the hot plate test in mice and tail flick test in rats, ATE in same doses also showed significant analgesic activity (P <0.05) which is almost equally efficacious to standard drug diclofenac sodium (50mg/kg BW orally) but far less efficacious than morphine sulfate (5mg/kg BW subcutaneous).ATE (200mg/Kg BW orally) activity did not blocked by naloxone (2mg/kg intra-peritoneal).Conclusions: ATE possesss significant anti-nociceptive activity as evidenced in all the animal models of nociception. It might exert its effect through the peripheral mechanism of analgesic action possibly by interference in biosynthesis, release and/or action of prostaglandins and leukotrienes

    Hemoglobin receptor in Leishmania is a hexokinase located in the flagellar pocket

    Hb endocytosis in Leishmania is mediated through a 46-kDa protein located in the flagellar pocket. To understand the nature of the Hb receptor (HbR), we have purified the 46-kDa protein to homogeneity from Leishmania promastigote membrane. Purified HbR specifically binds Hb. The gene for HbR was cloned, and sequence analysis of the full-length HbR gene indicates the presence of hexokinase (HK) signature sequences, ATP-binding domain, and PTS-II motif. Four lines of evidence indicate that HbR in Leishmania is a hexokinase: 1) the recombinant HbR binds Hb, and the Hb-binding domain resides in the N terminus of the protein; 2) recombinant proteins and cell lysate prepared from HbR-overexpressing Leishmania promastigotes show enhanced HK activity in comparison with untransfected cells; 3) immunolocalization studies using antibodies against the N-terminal fragment (Ld-HbR-&#916;C) of Ld-HbR indicate that this protein is located in the flagellar pocket of Leishmania; and 4) binding and uptake of 125I-Hb by Leishmania is significantly inhibited by anti-Ld-HbR-&#916;C antibody and Ld-HbR-&#916;C, respectively. Taken together, these results indicate that HK present in the flagellar pocket of Leishmania is involved in Hb endocytosis

    Optimizing wetland restoration to improve water quality at a regional scale

    Published by IOP Publishing Ltd. Excessive phosphorus (P) export to aquatic ecosystems can lead to impaired water quality. There is a growing interest among watershed managers in using restored wetlands to retain P from agricultural landscapes and improve water quality. We develop a novel framework for prioritizing wetland restoration at a regional scale. The framework uses an ecosystem service model and an optimization algorithm that maximizes P reduction for given levels of restoration cost. Applying our framework in the Lake Champlain Basin, we find that wetland restoration can reduce P export by 2.6% for a budget of 50Mand5.150 M and 5.1% for a budget of 200 M. Sensitivity analysis shows that using finer spatial resolution data for P sources results in twice the P reduction benefits at a similar cost by capturing hot-spots on the landscape. We identify 890 wetlands that occur in more than 75% of all optimal scenarios and represent priorities for restoration. Most of these wetlands are smaller than 7 ha with contributing area less than 100 ha and are located within 200 m of streams. Our approach provides a simple yet robust tool for targeting restoration efforts at regional scales and is readily adaptable to other restoration strategies

    In this study, "Real-Time Information Access in Urban Environments: A User Interaction Study Using the Real-Time Information Test," participant data revealed a diverse group with an average age of 31, a balanced gender distribution, varying education levels (40% Bachelor's, 20% Master's, 40% PhD), and an average of 6 years of experience with urban navigation. The findings of the Real-Time Information Test (RTIT) showed an average job completion time of 140 seconds and a low average error count of 1.2, demonstrating competency in interacting with real-time information systems. Furthermore, the User Satisfaction Survey found an average of 8.4 overall satisfaction ratings, 8.4 user-friendliness ratings, and 7.8 information accuracy ratings, indicating excellent user experiences. These results highlight user variety, increases in job efficiency and accuracy, and high user satisfaction, all of which contribute to a comprehensive knowledge of real-time information access in urban contexts, with implications for system advancements and urban planning
