    Thirty Years with HIV Infection—Nonprogression Is Still Puzzling: Lessons to Be Learned from Controllers and Long-Term Nonprogressors

    In the early days of the HIV epidemic, it was observed that a minority of the infected patients did not progress to AIDS or death and maintained stable CD4+ cell counts. As the technique for measuring viral load became available it was evident that some of these nonprogressors in addition to preserved CD4+ cell counts had very low or even undetectable viral replication. They were therefore termed controllers, while those with viral replication were termed long-term nonprogressors (LTNPs). Genetics and virology play a role in nonprogression, but does not provide a full explanation. Therefore, host differences in the immunological response have been proposed. Moreover, the immunological response can be divided into an immune homeostasis resistant to HIV and an immune response leading to viral control. Thus, non-progression in LTNP and controllers may be due to different immunological mechanisms. Understanding the lack of disease progression and the different interactions between HIV and the immune system could ideally teach us how to develop a functional cure for HIV infection.     Osmotic Stress Blocks Mobility and Dynamic Regulation of Centriolar Satellites

    Centriolar satellites (CS) are small proteinaceous granules that cluster around the centrosome and serve as cargo vehicles for centrosomal proteins. It is generally accepted that CS support a number of canonical and specialized centrosome functions. Consequently, these highly dynamic structures are the target of regulation by several cellular signalling pathways. Two decades of research have led to the identification of a large number of molecular components and new biological roles of CS. Here, we summarize the latest advances in the continuous efforts to uncover the compositional, functional, dynamic and regulatory aspects of CS. We also report on our discovery that osmotic stress conditions render CS immobile and insensitive to remodelling. Upon a range of p38-activating stimuli, MK2 phosphorylates the CS component CEP131, resulting in 14-3-3 binding and a block to CS formation. This normally manifests as a rapid cellular depletion of satellites. In the case of osmotic stress, a potent inducer of p38 activity, CS translocation and dissolution is blocked, with the net result that satellites persist in an immobile state directly adjacent to the centrosome. Our results highlight a unique scenario where p38 activation and CS depletion is uncoupled, with potential implications for physiological and pathological osmotic stress responses

    Gut microbiota profiles of autism spectrum disorder and attention deficit/hyperactivity disorder:A systematic literature review

    Accumulating evidence has implicated an involvement of the gut-brain axis in autism spectrum disorder (ASD) and attention-deficit hyperactivity disorder (ADHD), however with highly diverse results. This systematic review aims to describe and evaluate studies investigating the gut microbiota composition in individuals with ASD or ADHD and to evaluate if variations in gut microbiota are associated with these disorders. Twenty-four articles were identified in a systematic literature search of PubMed and Embase up to July 22, 2019. They consisted of 20 studies investigating ASD and four studies investigating ADHD. For ASD, several studies agreed on an overall difference in β-diversity, although no consistent bacterial variation between all studies was reported. For ADHD, the results were more diverse, with no clear differences observed. Several common characteristics in gut microbiota function were identified for ASD compared to controls. In contrast, highly heterogeneous results were reported for ADHD, and thus the association between gut microbiota composition and ADHD remains unclear.     Impaired platelet aggregation and rebalanced hemostasis in patients with chronic hepatitis C virus infection

    Increased risk of both cardiovascular disease (CVD) and bleeding has been found in patients with chronic hepatitis C (CHC) infection, and a re-balanced hemostasis has been proposed. The aim of this study was to investigate functional whole blood coagulation and platelet function in CHC infection. The prospective study included 82 patients with CHC infection (39 with advanced liver fibrosis and 43 with no or mild liver fibrosis) and 39 healthy controls. A total of 33 patients were treated for CHC infection and achieved sustained virological response (SVR). Baseline and post-treatment blood samples were collected. Hemostasis was assessed by both standard coagulation tests and functional whole blood hemostatic assays (thromboelastograhy (TEG), and platelet aggregation (Multiplate). Patients with CHC and advanced fibrosis had impaired platelet aggregation both compared to patients with no or mild fibrosis and to healthy controls. Patients with CHC and advanced fibrosis also had lower antithrombin, platelet count, and coagulation factors II-VII-X compared to healthy controls. In contrast, TEG did not differ between groups. In treated patients achieving SVR, post-treatment platelet count was higher than pre-treatment counts (p = 0.033) and ADPtest, ASPItest, and RISTOhightest all increased post treatment (all p < 0.05). All Multiplate tests values, however, remained below those in the healthy controls. CHC-infected patients displayed evidence of rebalanced hemostasis with only partly hemostatic normalization in patients achieving SVR. The implications of rebalanced hemostasis and especially the impact on risk of CVD and bleeding warrants further studies

    Hvordan kan sædskiftet indrettes så den økologiske kornproduktion øges uden at bæredygtigheden mindskes? Dette centrale spørgsmål belyses med resultater fra et økologisk sædskifteforsøg, der i 1996/97 blev anlagt på tre lokaliteter i Danmark. Da forsyningen med kvælstof (N) kan være meget kritisk i økologisk kornproduktion blev forsøget designet med fokus på dyrkningsmæssige faktorer der har indflydelse på N-forsyningen. Tre faktorer blev undersøgt: 1) Betydning af andel af N-fikserende afgrøder i sædskiftet 2) Betydning af efterafgrøder 3) Betydning af tilførsel af husdyrgødning. Der blev målt udbytter, udvaskning af næringsstoffer samt forekomst af ukrudt. Forsøget demonstrerer vigtigheden af at undersøge dyrkningsfaktorer i en sædskiftemæssig helhed.For eksempel har tilførsel af husdyrgødning ikke kun effekt i den afgrøde hvor den tilføres. Efterfølgende afgrøder kan også blive påvirkede. Flere af disse langsigtede effekter er knyttet til kløvergræsmarker og N-fikserende efterafgrøder, som er væsentlige elementer i økologiske sædskifter.     Incomplete Immune Recovery in HIV Infection: Mechanisms, Relevance for Clinical Care, and Possible Solutions

    Treatment of HIV-infected patients with highly active antiretroviral therapy (HAART) usually results in diminished viral replication, increasing CD4+ cell counts, a reversal of most immunological disturbances, and a reduction in risk of morbidity and mortality. However, approximately 20% of all HIV-infected patients do not achieve optimal immune reconstitution despite suppression of viral replication. These patients are referred to as immunological nonresponders (INRs). INRs present with severely altered immunological functions, including malfunction and diminished production of cells within lymphopoetic tissue, perturbed frequencies of immune regulators such as regulatory T cells and Th17 cells, and increased immune activation, immunosenescence, and apoptosis. Importantly, INRs have an increased risk of morbidity and mortality compared to HIV-infected patients with an optimal immune reconstitution. Additional treatment to HAART that may improve immune reconstitution has been investigated, but results thus far have proved disappointing. The reason for immunological nonresponse is incompletely understood. This paper summarizes the known and unknown factors regarding the incomplete immune reconstitution in HIV infection, including mechanisms, relevance for clinical care, and possible solutions

    Calanus finmarchicus basin scale life history traits and role in community carbon turnover during spring

    The copepod Calanus finmarchicus was investigated in four Subpolar Basins, Labrador, Irminger, Iceland, and Norwegian Seas, during spring, covering the time of ascent, grazing, and initiation of reproduction in the area. Lipid content, spawning activity, and stage composition and vital rates, such as egg and faecal pellet production were measured and linked to environmental parameters. Specific egg- and faecal pellet production rates varied with diatom biomass and were negatively correlated with temperature. Comparison of the various biological indicators revealed different life history traits C. finmarchicus has adopted in the different basins. In Labrador Sea, the females have invested in large eggs compared to the remaining basins. Labrador and Irminger Sea C. finmarchicus invest in size that we propose to be adaptation to cope with warmer overwintering habitats resulting in larger potential lipid storage capacity, while the Iceland and Norwegian Sea females can invest their remaining lipid storage in spring to fuel lipid-driven egg production. Grazing pressure on the phytoplankton community was estimated and compared between copepod and two dominating groups of protozooplankton; ciliates and heterotrophic dinoflagellates. Despite approximately the same biomass in the upper 100 m, the grazing impact of the protozoan grazers was an order of magnitude higher than the C. finmarchicus dominated mesozooplankton. This illustrates the importance to also include the smallest grazers when studying the spring bloom in high latitude marine ecosystems if the fate of the primary production should be fully understood.publishedVersio
