    Receptor tyrosine kinases (RTKs), such as epidermal growth factor receptor (EGFR) are internalised from the plasma membrane by endocytosis and may be transported to the nucleus. EGFR, a receptor for EGF and other RTKs, HER-2 and HER-4 has an important role in signalling; it contains transactivational activity and can function as a transcription co-factor to activate gene promoters. High nuclear accumulation of imported full length EGFR is associated with an increased tumour proliferation and a reduced survival in cancer patients. However, little is known about the mechanism by which membrane-bound proteins, such as EGFR, translocate from the cell surface into the cell nucleus; how nuclear membrane proteins cross through the NPC to reach the INM. The mechanism of translocation for soluble proteins is also presently unclear. EGFR nuclear import is mediated by importin α/β. And it is exported from the nucleus by the exportin CRM1. Sec61β which may reside in the inner nuclear membrane (INM) is required for the release of EGFR from the INM into the nucleus. Nuclear transport involves binding of nuclear localisation sequences (NLSs) within the cargo to a transport receptor (karyopherins or importin). Karyopherins interact with certain nuclear pore complex (NPC) proteins (nucleoporins). Membrane proteins can access the INM through the NPC membrane: by diffusion, using classical nuclear transport factors (the importin/Ran system); or by an ATP dependent mechanism. EGFR may use the former mechanism. This work concentrates to show by electron microscopy and by Immuno-Fluorescence that upon EGF treatment, the biotinylated cell surface EGFR is trafficked to the INM through the NPC, yet remaining a membrane-bound protein. We also confirm that importin regulates EGFR nuclear transport to the INM and in addition, Sec61β is required for EGFR release to the nucleoplasm. Altogether, this study of the mechanism of EGFR nuclear-cytoplasmic import in breast cancer cells, further confirms previous reports and provides an understanding of the nature and regulation of the nuclear EGFR pathway and the mechanism by which cell-surface EGFR is shuttled in the cell cytoplasm and channelled through the Golgi and Endoplasmic Reticulum (ER) compartments and into the nucleus through the NPC

    Includes bibliographical references.In 2011, the Western Cape Department of Health (WC DoH) adopted a health plan called Healthcare 2020. The framework of the plan emphasizes the necessity to de-hospitalize care in the province. The healthcare plan has brought about a shift in care policy. That is, moving from a passive delivery to a more active delivery where patients act independently. Active care refers to care that takes into account the needs of patients. Developing rehabilitation and care skills within the Western Cape Province is a key element to meet the de-hospitalization goal. Thus, the WC DoH commissioned and funded a pilot programme to train Rehabilitation Care Workers (RCWs) in the Mitchell's Plain District. This pilot programme involved rehabilitation and support for people with disabilities (PWDs) and their families. The rationale behind the programme was that if RCWs are given the necessary training and knowledge through learning and practical sessions, they will be more skilled to offer improved quality of care and support (Schneider, 2012, Healthcare, 2020). The Disabilities Studies Division (DSD) of School of Health and Rehabilitation Sciences at University of Cape Town (UCT) designed and delivered the training, consisting of learning and practical components. The current evaluation aims at investigating implementation of the learning component of the programme

    Evaluation of mass selection for grain yield and estimation of genetic variability in three selected maize (Zea mays L.) populations

    Fourteen cycles of mass selection for grain yield in Krug(BSK) maize (Zea mays L.) population were evaluated. Estimates of genetic components of variance were obtained for comparison with those computed from the eighth cycle of half-sib and S(,1) selection conducted in the same population. The CO and the even cycles (C2 to C14), and two sets of testcrosses (Cn x CO and Cn x B73) were evaluated in nine environments to estimate progress from mass selection. Unselected S(,1) lines derived from the original, mass, half-sib, and S(,1) selected populations wer evaluated in four environments to obtain the estimates of genetic variance;Significant, but low (0.50 q/ha), linear response for grain yield was obtained from mass selection for grain yield. Increased yield was associated with delayed silk emergence, increased stalk breakage, increased grain moisture, and greater ear height. Estimates of genetic variability for grain yield indicated a decrease of genetic variance for the S(,1) derived population and no change for mass and half-sib derived populations. For many traits, half-sib and S(,1) derived populations had similar estimates of genetic variance. The mass selected population had increased genetic variability for silk date, ear height, stalk lodging, and grain moisture. Estimates of heritability were relatively high for most traits;Although significant linear response was observed from mass selection, the mean performance of S(,1) progenies derived from the mass selected population were 6.85% lower yielding than S(,1) lines from the original population. Reduced yield for the mass selection derived lines was accompanied by delayed silking, decreased ears per plant, increased ear height, and greater stalk breakage. The S(,1) progenies from half-sib and S(,1) selected populations, however, had increased grain yield, reduced ear height, root lodging, and stalk breakage, and increased ears per plant;Predicted genetic gains computed for each trait in each population indicated that S(,1) selection was superior to mass and half-sib selection methods
