Drought affects the heat-hardening capacity of alpine plants as indicated by changes in xanthophyll cycle pigments, singlet oxygen scavenging, α-tocopherol and plant hormones
AbstractAlpine environments in Europe are increasingly affected by more erratic precipitation patterns, and more frequent drought and heat waves. Heat-hardening capacity is a key feature for survival of these abiotic stress factors, but it is poorly understood how heat and drought affect plant performance when combined. The main objectives of this study were (1) to determine maximum heat hardening capacity in 14 selected plant species and (2) to study how alpine plants respond to combined heat and drought stress compared to heat alone. (3) For risk assessment maximum leaf temperatures were measured in the field and (4) important methodological aspects of testing heat tolerance were evaluated. Heat hardening capacity was assessed by Tc, the heat threshold of photosystem II (PS II), and by heat tolerance tests based on visual inspection of leaf tissue damage or potential quantum efficiency of PS II (Fv/Fm). A purpose-built Heat Tolerance Testing System (HTTS) was used, which allows for controlled heat exposure of whole plants under nearly natural conditions. Additionally, in two species from contrasting habitats, Senecio incanus and Primula minima, the dynamics of heat hardening was studied during and after 8days exposure to heat (H), or to a combination of heat and severe drought (H+D) within a light-transmissive heat hardening chamber at the alpine field site. In both species, H treatment significantly increased heat tolerance (LT50), determined by the HTTS, to 58.0°C and 54.9°C, respectively, and was accompanied by elevated production of abscisic acid (ABA) and salicylic acid (SA), whereas jasmonic acid (JA) levels decreased. Under H+D the LT50 was only 56.5°C and 51.6°C, respectively, and levels of ABA were higher in S. incanus and SA lower in both species in comparison to H. Changes in xanthophyll cycle pigments, α-tocopherol and carotenoids:chlorophyll ratio were more pronounced in P. minima than in S. incanus. In P. minima both H and H+D significantly increased singlet oxygen (1O2) scavenging capacity, determined by electron paramagnetic resonance spectroscopy (EPR). In the field, the maximum half-hourly mean (HHM) leaf temperature of P. minima (32.2°C) was significantly lower than of S. incanus (46.5°C, a potentially harmful temperature). We conclude that the investigated species are well adapted to the prevailing temperature conditions in the field. They also possess an outstanding heat hardening capacity, but this can be curtailed when heat is combined with drought. As drought further increases leaf temperatures, the risk of suffering lethal heat damage of some species may increase in the future, particularly at south exposed, ruderal alpine sites with uncertain water supply

Objectives. To investigate self-reported health-related quality of life
(HrQoL) in children and adolescents with chronic medical conditions compared
with siblings/peers. Methods. Group 1 (6 treatment centers) consisted of 74
children/adolescents aged 8â16 years with hereditary bleeding disorders (HBD),
12 siblings, and 34 peers. Group 2 (one treatment center) consisted of 70
children/adolescents with stroke/transient ischemic attack, 14 siblings, and
72 peers. HrQoL was assessed with the ârevised KINDer
LebensqualitĂ€tsfragebogenâ (KINDL-R) questionnaire. Multivariate analyses
within groups were done by one-way ANOVA and post hoc pairwise single
comparisons by Studentâs -tests. Adjusted pairwise comparisons were done by
hierarchical linear regressions with individuals nested within treatment
centers (group 1) and by linear regressions (group 2), respectively. Results.
No differences were found in multivariate analyses of self-reported HrQoL in
group 1, while in group 2 differences occurred in overall wellbeing and all
subdimensions. These differences were due to differences between patients and
peers. After adjusting for age, gender, number of siblings, and treatment
center these differences persisted regarding self-worth () and friend-related
wellbeing (). Conclusions. In children with HBD, HrQoL was comparable to
siblings and peers. In children with stroke/TIA HrQoL was comparable to
siblings while peers, independently of relevant confounder, showed better
self-worth and friend-related wellbeing

Over-Detection of Melanoma-Suspect Lesions by a CE-Certified Smartphone App: Performance in Comparison to Dermatologists, 2D and 3D Convolutional Neural Networks in a Prospective Data Set of 1204 Pigmented Skin Lesions Involving Patientsâ Perception
The exponential increase in algorithm-based mobile health (mHealth) applications (apps) for melanoma screening is a reaction to a growing market. However, the performance of available apps remains to be investigated. In this prospective study, we investigated the diagnostic accuracy of a class 1 CE-certified smartphone app in melanoma risk stratification and its patient and dermatologist satisfaction. Pigmented skin lesions â„ 3 mm and any suspicious smaller lesions were assessed by the smartphone app SkinVisionÂź (SkinVisionÂź B.V., Amsterdam, the Netherlands, App-Version 6.8.1), 2D FotoFinder ATBMÂź master (FotoFinder ATBMÂź Systems GmbH, Bad Birnbach, Germany, Version 3.3.1.0), 3D VectraÂź WB360 (Canfield Scientific, Parsippany, NJ, USA, Version 4.7.1) total body photography (TBP) devices, and dermatologists. The high-risk score of the smartphone app was compared with the two gold standards: histological diagnosis, or if not available, the combination of dermatologistsâ, 2D and 3D risk assessments. A total of 1204 lesions among 114 patients (mean age 59 years; 51% females (55 patients at high-risk for developing a melanoma, 59 melanoma patients)) were included. The smartphone appâs sensitivity, specificity, and area under the receiver operating characteristics (AUROC) varied between 41.3â83.3%, 60.0â82.9%, and 0.62â0.72% according to two study-defined reference standards. Additionally, all patients and dermatologists completed a newly created questionnaire for preference and trust of screening type. The smartphone app was rated as trustworthy by 36% (20/55) of patients at high-risk for melanoma, 49% (29/59) of melanoma patients, and 8.8% (10/114) of dermatologists. Most of the patients rated the 2D TBP imaging (93% (51/55) resp. 88% (52/59)) and the 3D TBP imaging (91% (50/55) resp. 90% (53/59)) as trustworthy. A skin cancer screening by combination of dermatologist and smartphone app was favored by only 1.8% (1/55) resp. 3.4% (2/59) of the patients; no patient preferred an assessment by a smartphone app alone. The diagnostic accuracy in clinical practice was not as reliable as previously advertised and the satisfaction with smartphone apps for melanoma risk stratification was scarce. MHealth apps might be a potential medium to increase awareness for melanoma screening in the lay population, but healthcare professionals and users should be alerted to the potential harm of over-detection and poor performance. In conclusion, we suggest further robust evidence-based evaluation before including market-approved apps in self-examination for public health benefits
The problematic legacy of victim specimens from the Nazi era: Identifying the persons behind the specimens at the Max Planck Institutes for Brain Research and of Psychiatry
Although 75 years have passed since the end of World War II, the Max Planck Society (Max-Planck Gesellschaft, MPG), successor to the Kaiser Wilhelm Society (Kaiser-Wilhelm-Gesellschaft, KWG), still must grapple with how two of its foremost institutesâthe KWI of Psychiatry in Munich and the KWI for Brain Research in Berlin-Buchâamassed collections of brains from victims of Nazi crimes, and how these human remains were retained for postwar research. Initial efforts to deal with victim specimens during the 1980s met with denial and, subsequently, rapid disposal in 1989/1990. Despite the decision of the MPGâs president to retain documentation for historical purposes, there are gaps in the available sources. This article provides preliminary results of a research program initiated in 2017 (to be completed by October 2023) to provide victim identifications and the circumstances of deaths

The production of tt⟠, W+bb⟠and W+cc⟠is studied in the forward region of protonâproton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98±0.02 fbâ1 . The W bosons are reconstructed in the decays WââÎœ , where â denotes muon or electron, while the b and c quarks are reconstructed as jets. A search for the rare decays and is performed in a data set corresponding to an integrated luminosity of 3.0 fb collected by the LHCb detector in proton-proton collisions at centre-of-mass energies of 7 and 8 TeV. Decay candidates with pion pairs that have invariant mass in the range 0.5-1.3 GeV/ and with muon pairs that do not originate from a resonance are considered. The first observation of the decay and the first evidence of the decay are obtained and the branching fractions are measured to be and , where the third uncertainty is due to the branching fraction of the decay , used as a normalisation.A search for the rare decays Bs0âÏ+ÏâÎŒ+ÎŒâ and B0âÏ+ÏâÎŒ+ÎŒâ is performed in a data set corresponding to an integrated luminosity of 3.0 fbâ1 collected by the LHCb detector in protonâproton collisions at centre-of-mass energies of 7 and 8 TeV . For the first time several observables are measured in the dielectron mass squared () interval between 0.002 and 1.120. The angular observables and which are related to the polarisation and to the lepton forward-backward asymmetry, are measured to be and , where the first uncertainty is statistical and the second systematic. The angular observables and which are sensitive to the photon polarisation in this range, are found to be and .
