    Background and purpose At present, more than 20 different forms of limb-girdle muscular dystrophies (LGMDs) are known (at least 7 autosomal dominant and 14 autosomal recessive). Although these different forms show some typical phenotypic characteristics, the existing clinical overlap makes their differential diagnosis difficult. Limb-girdle muscular dystrophy type 2 (LGMD2A) is the most prevalent LGMD in many European as well as Brazilian communities and is caused by mutations in the gene CAPN3. Laboratory testing, such as calpain immunohistochemistry and Western-blot analysis, is not totally reliable, since up to 20% of molecularly confirmed LGMD2A show normal content of calpain 3 and a third of LGMD2A biopsies have normal calpain 3 proteolytic activity in the muscle. Thus, genetic testing is considered as the only reliable diagnostic criterion in LGMD2A. Material and methods In an attempt to find a correlation between genotype and muscle pathology in limb-girdle muscular dystrophy 2A we performed histopathological investigation of a group of 31 patients subdivided according to the type of pathologic CAPN3 gene mutation. Results In all biopsies typical features of muscular dystrophy such as fiber necrosis and regeneration, variation in fiber size and fibrosis were noted. Lobulated fibers were often encountered in the muscle biopsies of LGMD2A patients. Such fibers were more frequent in patients with 550delA mutation. Conclusions These findings may be helpful in establishing diagnostic strategies in LGMD.Wstęp i cel pracy Dotychczas opisano ponad 20 różnych form dystrofii obręczowo-kończynowej (limb girdle muscular dystrophy – LGMD) (co najmniej 7 rodzajów o dziedziczeniu autosomalnym dominującym oraz 14 o dziedziczeniu autosomalnym recesywnym). Pomimo że część z tych chorób można różnicować na podstawie obrazu klinicznego, diagnostykę utrudnia często podobieństwo objawów. Dystrofia obręczowo–kończynowa typu 2A (limb-girdle muscular dystrophy type 2 – LGMD2A), najczęstsza dystrofia mięśniowa w wielu społecznościach (np. w Europie i Brazylii), spowodowana jest przez mutacje w genie kalpainy 3 (CAPN3). Badanie immunohisto-chemiczne kalpainy czy też metodą Western blot nie są wystarczające do ustalenia właściwego rozpoznania (w odpowiednio 1/3 i 20% potwierdzonych genetycznie LGMD2A badania te wypadają prawidłowo). Podstawę rozpoznania tej miopatii stanowi badanie genetyczne. Materiał i metody W pracy przedstawiono wyniki badania zależności między genotypem a analizą histopatologiczną biopsji mięśnia u 31 chorych na LGMD2A. Chorzy podzieleni zostali na grupy według wyników badania genetycznego genu CAPN3 odpowiedzialnego za tę chorobę. Wyniki We wszystkich badanych biopsjach stwierdzano typowe zmiany dystroficzne, takie jak obecność włókien martwiczych i regenerujących, zróżnicowaną wielkość włókien oraz włóknienie. Włókna o nierównomiernym rozkładzie barwień na enzymy oddechowe (lobulated fibers) były często obserwowane w biopsjach chorych z LGMD2A. Tego typu włókna szczególnie często występowały u chorych z mutacją 550delA. Wnioski Wyniki pracy wnoszą nowe informacje ułatwiające diagnostykę LGMD

    A Survey of Avian Influenza in Tree Sparrows in China in 2011

    Tree sparrows (Passer montanus) are widely distributed in all seasons in many countries. In this study, a survey and relevant experiments on avian influenza (AI) in tree sparrows were conducted. The results suggested that the receptor for avian influenza viruses (AIVs), SAα2,3Gal, is abundant in the respiratory tract of tree sparrows, and most of the tree sparrows infected experimentally with two H5 subtype highly pathogenic avian influenza (HPAI) viruses died within five days after inoculation. Furthermore, no AIVs were isolated from the rectum eluate of 1300 tree sparrows, but 94 serological positives of AI were found in 800 tree sparrows. The serological positives were more prevalent for H5 subtype HPAI (94/800) than for H7 subtype AI (0/800), more prevalent for clade 2.3.2.1 H5 subtype HPAI (89/800) than for clade 2.3.4 (1/800) and clade 7.2 (4/800) H5 subtype HPAI, more prevalent for clade 2.3.2.1 H5 subtype HPAI in a city in southern China (82/800) than in a city in northern China (8/800). The serological data are all consistent with the distribution of the subtypes or clades of AI in poultry in China. Previously, sparrows or other passerine birds were often found to be pathogenically negative for AIVs, except when an AIV was circulating in the local poultry, or the tested passerine birds were from a region near waterfowl-rich bodies of water. Taken together, the data suggest that tree sparrows are susceptible to infection of AIVs, and surveys targeting sparrows can provide good serological data about the circulation of AIVs in relevant regions

    Host shifts and molecular evolution of H7 avian influenza virus hemagglutinin

    Evolutionary consequences of host shifts represent a challenge to identify the mechanisms involved in the emergence of influenza A (IA) viruses. In this study we focused on the evolutionary history of H7 IA virus in wild and domestic birds, with a particular emphasis on host shifts consequences on the molecular evolution of the hemagglutinin (HA) gene. Based on a dataset of 414 HA nucleotide sequences, we performed an extensive phylogeographic analysis in order to identify the overall genetic structure of H7 IA viruses. We then identified host shift events and investigated viral population dynamics in wild and domestic birds, independently. Finally, we estimated changes in nucleotide substitution rates and tested for positive selection in the HA gene. A strong association between the geographic origin and the genetic structure was observed, with four main clades including viruses isolated in North America, South America, Australia and Eurasia-Africa. We identified ten potential events of virus introduction from wild to domestic birds, but little evidence for spillover of viruses from poultry to wild waterbirds. Several sites involved in host specificity (addition of a glycosylation site in the receptor binding domain) and virulence (insertion of amino acids in the cleavage site) were found to be positively selected in HA nucleotide sequences, in genetically unrelated lineages, suggesting parallel evolution for the HA gene of IA viruses in domestic birds. These results highlight that evolutionary consequences of bird host shifts would need to be further studied to understand the ecological and molecular mechanisms involved in the emergence of domestic bird-adapted viruses

    Influenza A Viruses from Wild Birds in Guatemala Belong to the North American Lineage

    The role wild bird species play in the transmission and ecology of avian influenza virus (AIV) is well established; however, there are significant gaps in our understanding of the worldwide distribution of these viruses, specifically about the prevalence and/or significance of AIV in Central and South America. As part of an assessment of the ecology of AIV in Guatemala, we conducted active surveillance in wild birds on the Pacific and Atlantic coasts. Cloacal and tracheal swab samples taken from resident and migratory wild birds were collected from February 2007 to January 2010.1913 samples were collected and virus was detected by real time RT-PCR (rRT-PCR) in 28 swab samples from ducks (Anas discors). Virus isolation was attempted for these positive samples, and 15 isolates were obtained from the migratory duck species Blue-winged teal. The subtypes identified included H7N9, H11N2, H3N8, H5N3, H8N4, and H5N4. Phylogenetic analysis of the viral sequences revealed that AIV isolates are highly similar to viruses from the North American lineage suggesting that bird migration dictates the ecology of these viruses in the Guatemalan bird population

    Sanitary assesment of selected water reservoir's in Warsaw parks

    The research of the sanitary state in three waters’ reservoirs situated within the area of three municipal park of Warsaw: Glinianki Szczęśliwickie, Glinianki Moczydłowskie and Jeziorko Kamionkowskie. The selected sanitary parameters of water quality in Reservoirs (bacteriological and chemical) were analyzed in May and July 2007. There was described existenexistence of fecal indicators in bacteriological analysis: Escherichia coli (MPN, index), Streptococcus faecalis (MPN, index), Clostridium perfringens (general amount of CFU). There was indicated also amount of mesophiles and psychrophiles bacteria (general amount of CFU). The chemical analysis included basic indicators of water pollution (BOD5 with the addition of a nitrifi cation inhibitor, TOC, TN). Results shows that sanitary pollution of water is quite high and organic pollution is very high. This pollution probably is as a result of human activity (sewage). Investigated reservoirs contain water, which is danger for human health. Those reservoirs cannot be used as a swimming pool and are only element of the landscape which can increase it`s complexity
