    Induction of Apoptosis and Cell Cycle Arrest in Human Colon Carcinoma Cells by Corema album Leaves

    The leaves of Corema album (Ericaceae), an endemic shrub which grows in Atlantic coastal areas of the Iberian Peninsula, are rich in flavonoids and other secondary metabolites. Silica gel column chromatography of the ethyl acetate extract from dried leaves was performed and a flavonic active fraction was obtained. The cytotoxic activity of this fraction was assessed using the colon cancer cell lines HCT116 and HT29. After 48 hours of treatment, cell viability was determined with luminescence-based ATPLite assay, showing IC50 values of 7.2 ± 0.7 and 6.8 ± 1.2 µg/mL, respectively. The study by flow cytometry revealed that the cytotoxicity of this fraction was mediated, at least in part, by induction of apoptosis and G2/M cell cycle arrest. The active fraction was then subjected to Sephadex LH-20 chromatography and two flavonoids were separated and identified as the flavanone pinocembrin and 2’,4’-dihydroxychalcone after UV, MS and NMR analysis

    Phytochemical Profile and Antibacterial Activity of Retama raetam and R. sphaerocarpa cladodes from Algeria

    Retama raetam (RR) and R. sphaerocarpa (RS) are shrubs growing in Algeria desert areas, where are commonly used as healing remedies because of their antiseptic, antipyretic and anti-diarrheal properties. Phytochemical studies have shown that these species are very rich in flavonoids (isoflavones) and alkaloids (quinolizidine and bipiperidyl). The aim of this study was to compare the chemical composition of both Retama species by GC/MS and LC/MS and to determinate their antimicrobial activity of two Retama species growing naturally in Algeria. Ten alkaloids and seven flavonoids were identified in cladodes of RR and RS. The quantitative analysis showed that the most abundant flavonoid of both the aqueous extract from RR and RS was the isoflavone genistein (610.0±2.8 and 408.0±14.1 mg/100 g respectively), whereas sparteine was the predominant alkaloid in RR and retamine in RS. The antibacterial activity of Retama extracts against standard strains was performed by minimum inhibitory concentration (MIC), and by the disc diffusion method (expressed by inhibition zone, IZ). Both Retama species showed the best activity against Staphylococcus aureus and methicillin-resistant S. aureus (MRSA), being RS aqueous extract more active than RR aqueous extract, with MIC 125 µg/mL and bactericidal activity against both strains

    The phenomenon of today’s ageing population has increased interest in the search for new active substances that delay the onset and development of neurodegenerative diseases. In this respect, the search for natural compounds, mainly phenolic compounds, with neuroprotective activity has become the focus of growing interest. Verbascoside is a phenylethanoid that has already presented several pharmacological activities. The purpose of this study is to isolate and identify verbascoside from Acanthus mollis leaves. Consequently, its neuroprotective ability through enzymatic inhibition and free radical scavenging ability has been analyzed both in vitro and in cell culture assays. The antioxidant capacity of verbascoside was evaluated in vitro through total antioxidant capacity, DPPH•, OH•, and O2•—scavenging activity assays. The effect of verbascoside on intracellular reactive oxygen species (ROS) levels of HepG2 and SH-SY5Y cell lines was studied in normal culture and under induced oxidative stress. The inhibitory ability of the phenylethanoid against several enzymes implied in neurodegenerative diseases (tyrosinase, MAO-A, and AChE) was analyzed in vitro. Verbascoside neuroprotective activity is at least in part related to its free radical scavenging ability.     The rhizomes of Acanthus mollis have traditionally been used for the treatment of several ailments involving inflammation. However, to the best of our knowledge, their chemical composition and pharmacological properties have not been studied until now. As a first approach, this study analyses the A. mollis rhizome hexane extract phytochemistry and its anti-inflammatory and antioxidant capacities in HepG2 and RAW 264.7 cell culture assays. Chemical profiling was performed with gas chromatography mass spectrometry without the modification of native molecules. Free phytosterols (such as β-sitosterol) account for 70% of detected compounds. The anti-inflammatory capacity of the rhizome extract of A. mollis is mediated by the decrease in the NO production in RAW 264.7 that has previously been stimulated with lipopolysaccharide in a dose-dependent manner. Furthermore, HepG2 pre-treatment with the rhizome extract prevents any damage being caused by oxidative stress, both through ROS scavenge and through the antioxidant cellular enzyme system. In this respect, the extract reduced the activity of glutathione peroxidase and reductase, which were stimulated under oxidative stress conditions. Our results suggest that the extract from the rhizomes of A. mollis may constitute a potential source of natural products with anti-inflammatory activity and could validate the traditional use of A. mollis

    Este proyecto pretende acercar a la comunidad educativa del centro una realidad tan próxima, compleja y dramática como la del continente africano, desde casi todas las materias. Y pretende hacerlo en coherencia con valores que identifican al centro. Los objetivos son fomentar la colaboración y participación de los alumnos intercambiando sus propias experiencias; potenciar una conciencia social y ecológica; formar integralmente a los alumnos para el mejor desarrollo de la sociedad; valorar la utilidad de la enseñanza activa y el aprendizaje cooperativo; diseñar actividades que permitan desarrollar una enseñanza interdisciplinar; posibilitar que los alumnos desarrollen estrategias globalizadoras de organización del conocimiento mediante el tratamiento de la información; favorecer el aprendizaje significativo; animar al profesorado a buscar estrategias que favorezcan la actividad didáctica; fomentar la lectura y el uso correcto de la expresión oral y escrita, además de mejorar la ortografía; extender el conocimiento y la utilización de las nuevas tecnologías de la información y la comunicación; estrechar las relaciones entre los diferentes sectores de la comunidad educativa; fomentar la participación activa en la vida social; mejorar la convivencia en el centro; desarrollar hábitos de vida saludables; provocar en los alumnos una visión positiva del centro como lugar de ocio y de enriquecimiento cultural; desarrollar y favorecer las inquietudes culturales de los alumnos; desarrollar las actitudes positivas hacia otras culturas; extender el conocimiento de lenguas extranjeras y mantener relaciones estables de colaboración con instituciones culturales y sociales. Las actividades desarrolladas se organizan en tres modalidades que son actividades globales que implican a toda la comunidad educativa como un concurso de lectura llamado África desde la biblioteca, el día del Libro, viaje de fin de curso a Túnez, apadrinar una escuela en Marruecos, una jornada cultural antes de las vacaciones de Semana Santa, talleres a cargo de la Fundación Yehudi Menuhin y Hora 31 con ciclos de charlas y conferencias; actividades durante el horario lectivo en los distintos departamentos didácticos como proyecciones de películas, composición de textos, debates y trabajos en equipo; y las actividades realizadas fuera del horario lectivo como son talleres, cineclub, coloquios, exposiciones, actividades de cooperación y asistir a diversos espectáculos. La metodología es diversa, en relación con la pluralidad de actividades que se llevan a cabo. Como indicadores del proceso de evaluación se toman para las actividades del currículo, la propia evaluación de profesores. Aunque es un indicador cuantitativo para el análisis final y junto a otros parámetros, se trabaja también la evaluación cualitativa, para que las conclusiones tengan un carácter más global de los resultados del proyecto. Por el contrario, las actividades que se realizan fuera del horario lectivo se miden con el grado de participación. Además como herramienta de evaluación individual se elaboran encuestas para alumnos y profesores para que reflejen sus opiniones personales sobre el proyecto.
