Novel functional anti-HER3 monoclonal antibodies with potent anti-cancer effects on various human epithelial cancers

Resistance of progressive cancers against chemotherapy is a serious clinical problem. In this context, human epidermal growth factor receptor 3 (HER3) can play important roles in drug resistance to HER1- and HER2- targeted therapies. Since clinical testing of anti-HER3 monoclonal antibodies (mAbs) such as patritumab could not show remarkable effect compared with existing drugs, we generated novel mAbs against anti-HER3. Novel rat mAbs reacted with HEK293 cells expressing HER3, but not with cells expressing HER1, HER2 or HER4. Specificity of mAbs was substantiated by the loss of mAb binding with knockdown by siRNA and knockout of CRISPR/Cas9-based genome-editing. Analyses of CDR sequence and germline segment have revealed that seven mAbs are classified to four groups, and the binding of patritumab was inhibited by one of seven mAbs. Seven mAbs have shown reactivity with various human epithelial cancer cells, strong internalization activity of cell-surface HER3, and inhibition of NRG1 binding, NRG1-dependent HER3 phosphorylation and cell growth. Anti-HER3 mAbs were also reactive with in vivo tumor tissues and cancer tissue-originated spheroid. Ab4 inhibited in vivo tumor growth of human colon cancer cells in nude mice. Present mAbs may be superior to existing anti-HER3 mAbs and support existing anti-cancer therapeutic mAbs

Predicting Law School Enrollment: The Strategic Use of Financial Aid to Craft a Class

    No full text
In this study, we explore what factors predict student decisions to enroll at law schools and how the probability of enrollment varies across students with various profiles and conditions. To find the predictors of enrollment and differences in the probability of enrollment across groups, we employ a logistic regression model using the institutional data obtained from one of the top-ranked law schools in the nation. After estimating the logistic regression model, the probabilities of enrollment are calculated for students with specific profiles and conditions based on the coefficients generated by the logistic regression analysis. The findings reveal many factors that are associated with the probability of enrollment at this law school. Particularly, students with higher academic qualifications, underrepresented minority status, the most selective undergraduate school, STEM background, and previous applicant status have a lower probability of enrollment compared to their respective counterparts. Simulation analysis findings show that the increase in financial aid does not increase the probability of enrollment for URM students and that out-of-state and international students are more sensitive to financial aid increases than in-state students. Admissions and enrollment management offices at individual institutions could apply this exercise with their own data to understand who is more or less likely to enroll and how their students with various profiles respond differently to various financial aid offers and recruitment efforts. It is our hope that this article is used as an example to other law schools to leverage their institutional data to create enrollment models that will help make more effective admission decision making

Leading for Equity from Where You Are: How Leaders in Different Roles Engage in Shared Equity Leadership

    No full text
In this report, the fourth in the On Shared Equity Leadership series, we describe the ways that leaders in different campus roles contribute to shared equity leadership (SEL) efforts. SEL is a leadership approach that scales diversity, equity, and inclusion (DEI) work and creates culture change by connecting individual and organizational transformation. Individuals embrace a personal journey toward critical consciousness to become equity-oriented leaders. Collectively, leaders embody a set of values and enact a set of practices that form new relationships and understandings, ultimately working to dismantle current systems and structures that inhibit equitable outcomes. This report examines roles from both a functional perspective (e.g., faculty, student affairs, and DEI-specific roles) and from a positional view (e.g., senior-level, mid-level, and groundlevel leaders), and it highlights the ways in which different values and practices are especially important for leaders in particular roles
