    Fabrication and Evaluation of Cellulose-Alginate-Hydroxyapatite Beads for the Removal of Heavy Metal Ions from Aqueous Solutions

In the present study, the potential of synthesized mixed cellulose, alginate and hydroxyapatite beads for the efficient removal of Ni (II) and Cu (II) ions from aqueous solutions was investigated. Cellulose, alginate and hydroxyapatite are known for their individual adsorption capacity. Beads were prepared in different ratios of these materials. The prepared beads were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscope (SEM) and thermogravimetric analysis (TGA). FTIR and XRD analysis showed characteristic peaks assigned to cellulose, alginate and hydroxyapatite. Thermal stability was observed to increase with increase of hydroxyapatite percentage in beads. SEM images showed increased surface porosity and roughness with the increase of cellulose percentage. The prepared beads were used for the removal of Ni (II) and Cu (II) ions from aqueous solutions and the process was optimized with respect to pH, contact time, adsorbent dose and initial concentration of metal ions. The values of the coefficient of determination (R2) of the Langmuir and Freundlich adsorption model indicated that the adsorbed Cu (II) and Ni (II) ions form monolayer coverage on the adsorbent surface. In kinetic analysis, Pseudo-second-order model fitted the kinetic experimental data well, as it showed high R2 value; above 0.9990

    Enhancement of diterpenoid steviol glycosides by co-overexpressing SrKO and SrUGT76G1 genes in Stevia rebaudiana Bertoni.

    Stevia rebaudiana (stevia) contains commercially important steviol glycosides, stevioside and rebaudioside A, these compounds have insulinotropic and anti-hyperglycemic effect. Steviol, stevioside and rebaudioside-A have taste modulation and insulin potentiation activity. Stevia leaves are composed of steviol (2-5%), stevioside (4-13%) and rebaudioside-A (1-6%). Stevioside has after-taste bitterness, rebaudioside-A is sweetest in taste among all the glycosides present. Therefore, lower ratio of rebaudioside-A to stevioside has bitter after-taste, which makes stevia plants unpalatable. By over-expressing the genes, SrUGT76G1 and SrKO, we propose to increase the ratio of RebA to stevioside in stevia. Various lines were generated and amongst them, seven lines had both the transgenes present. Co-overxpresion of SrUGT76G1 and SrKO led to the increased concentration of RebA in all the seven transgenic lines (KU1-KU7) than control plant and RebA to stevioside ratio also increased significantly. Steviol, stevioside and RebA showed a differential concentration in all the seven lines, but the pattern was the same in all of them and the ratio of RebA to stevioside increased dramatically. In transgenic line 2 (KU2), RebA showed a steep increase in concentration 52% the rebaudioside-A to stevioside ratio increased from 0.74 (control) to 2.83. In overall all the lines, RebA showed a positive correlation with steviol and stevioside. Overexpression of SrKO led to an increase in steviol which increased the stevioside, overexpression of SrUGT76G1 ultimately increased RebA concentration. In conclusion, concentration of RebA increased significantly with co- overexpression of SrUGT6G1 and SrKO genes. Lines with increased RebA are more palatable and commercially viable

    Genetics And Interrelationship of Yield and Yield Related Attributes in Some Genotypes of Safflower (Carthamus Tinctorius L.) Under Rainfed Conditions

    In order to estimate variability, heritability, genetic advance and relationship of grain yield with other yield related attributes, the experiments were carried out in field research Area of Barani Agricultural Research Institute, Chakwal, Pakistan during two rabi growing seasons 2011-12 and 2012-13. The experimental materials were comprised of 40 genotypes of safflower and were sown in randomized complete block design with three replications each year. The analysis of variance showed that all the safflower genotypes were significantly different for all the traits studied except days to physiological maturity. Among the attributes studied the estimates of phenotypic coefficient of variation (PCV) were generally higher than the genetic coefficient of variation (GCV).  Highest GCV was found in grain filing rate plant-1 (49.34 & 48.25) followed by seed yield plant-1 (49.09 & 48.18) during 2011-12 & 2012-13 respectively. Grain filing rate plant-1 showed maximum PCV (49.78 & 48.96) followed by seed yield plant-1 (49.64 & 49.44) during both years (2011-12 & 2012-13) respectively. The high heritability coupled with high genetic advance was measured in biological yield plant-1, harvest index (%), number of heads plant-1, and seed yield plant-1. These results revealed that high estimates of heritability and genetic advance for these traits would be helpful for the plant breeders to select the suitable combination and to achieve the enviable level of seed yield potential in safflower under arid environment conditions. The correlation results of two year studies revealed that seed yield plant-1 had significant and positive relationship with days to maturity, plant height, biological yield plant-1, number of seeds head-1, 1000-grain weight, grain filing rate plant-1, effective head weight, number of primary branches  plant-1 and plant height. On the other hand, days to 50% flowering, days to physiological maturity, number of seeds head-1, seed weight head-1 and 1000- grain weight showed low heritability with low genetic advance.     Significant predictors influencing the adoption of ChatGPT usage in the academia in Sindh, Pakistan: extension of UTAUT model

    The innovative ChatGPT AI-based application is considered a supplemental tool for learners to assist and guide broader academic information and research opportunities in various domains. This study uses the modified UTAUT model to investigate the significant predictors influencing the learners’ intention to adopt ChatGPT in academia in Sindh, Pakistan. The learners’ intention to adopt the ChatGPT tool is assessed based on significant predictors such as DS, HCI, PE, EE, SI, and FC as key indicators influencing the ChatGPT adoption. The research model and suggested hypotheses were addressed using a quantitative technique. Data gathered from 497 respondents as learners from top-ranked public institutions in Sindh, Pakistan, was examined using the PLS-SEM approach. This research found that all significant predictors influencing learners’ intention to adopt the ChatGPT tool and all predictors have a more significant impact. This research study can help administration and IT experts incorporate the skills and techniques associated with the ChatGPT tool in education in Sindh, Pakistan

    pH-Responsive Liposomes of Dioleoyl Phosphatidylethanolamine and Cholesteryl Hemisuccinate for the Enhanced Anticancer Efficacy of Cisplatin

    The current study aimed to develop pH-responsive cisplatin-loaded liposomes (CDDP@PLs) via the thin film hydration method. Formulations with varied ratios of dioleoyl phosphatidylethanolamine (DOPE) to cholesteryl hemisuccinate (CHEMS) were investigated to obtain the optimal particle size, zeta potential, entrapment efficiency, in vitro release profile, and stability. The particle size of the CDDP@PLs was in the range of 153.2 ± 3.08-206.4 ± 2.26 nm, zeta potential was -17.8 ± 1.26 to -24.6 ± 1.72, and PDI displayed an acceptable size distribution. Transmission electron microscopy revealed a spherical shape with ~200 nm size. Fourier transform infrared spectroscopic analysis showed the physicochemical stability of CDDP@PLs, and differential scanning calorimetry analysis showed the loss of the crystalline nature of cisplatin in liposomes. In vitro release study of CDDP@PLs at pH 7.4 depicted the lower release rate of cisplatin (less than 40%), and at a pH of 6.5, an almost 65% release rate was achieved compared to the release rate at pH 5.5 (more than 80%) showing the tumor-specific drug release. The cytotoxicity study showed the improved cytotoxicity of CDDP@PLs compared to cisplatin solution in MDA-MB-231 and SK-OV-3 cell lines, and fluorescence microscopy also showed enhanced cellular internalization. The acute toxicity study showed the safety and biocompatibility of the developed carrier system for the potential delivery of chemotherapeutic agents. These studies suggest that CDDP@PLs could be utilized as an efficient delivery system for the enhancement of therapeutic efficacy and to minimize the side effects of chemotherapy by releasing cisplatin at the tumor site

    Neurochemistry refers to the chemical processes that occur in the brain and nervous system. This section of study determines how neurochemicals influence the network of neural operation. The brain transfers numerous chemical information via neurons to communicate. The main role of neurochemistry activities takes place in the brain, which allows it to perform numerous actions. Foundation of brain is a little bit different from man to man and several things can play a role in the levels of various neurotransmitters in the brain. It is supposed that differences in brain chemistry may accountable for a variety of behavioral disorders. A particular cell called neurons is the basis of brain. Neurotransmitters have the capability that it can trigger when ordered to do so, along with receptors for specific neurotransmitters. By sending messages with neurotransmitters to signal various cell activities, brain perform its functions. Neurotransmitter spreads chemical messages from neuron to neuron to broadcast certain work and thus it works. A neuron may accept many chemical messages, both positive and negative from the other neurons contiguous it. They are accountable to get the neuron to reply in different ways, or they may work combine to produce a certain effect. Since all of this occurs just within a split second, the neurotransmitter must be cleared away rapidly so that the same receptors can be activated again and again. Psychoactive drugs work by briefly influencing a man's neurochemistry, which thusly causes changes in a man's mind-set, cognition, perception and behavior. Neuropeptides are endogenous protein molecules that are utilized for neuronal signaling. These molecules exert more prolonged and diverse effects on behavior than neurotransmitters.     Effect of Ajwa date pits powder (Phoenix dactylifera L.) on body composition, lipid profile and blood pressure in patients with hyperlipidemia: A randomized clinical trial

    Objective: To evaluate the effect of Ajwa dates pit powder (ADP) on lipid profile, body composition and blood pressure in patients with hyperlipidemia.Materials and Methods: This randomized controlled clinical study was carried out on 40 patients with total cholesterol >200 mg/dl, triglycerides >150 mg/dl and BMI >25, of either sex, aged 30-50 years, who were recruited through written consent. The patients were divided into two groups (n=20 each): the ADP and the control group (CG). All patients received the doctor’s prescribed class A statin (Rosuvastatin/ Atorvastatin) 10 mg/day, while 2.7 g ADP was given on daily basis before breakfast with lukewarm water for 40 days and the control group received the same amount of wheat flour. Body composition, blood pressure and lipid profile were determined at baseline, and after 20 and 40 days. Data were analyzed by using SPSS and GraphPad Prism.Results: ADP significantly reduced body weight (p<0.001), BMI (p<0.001), fat mass, body fat percentage, visceral fat area and waist circumference compared to the control group. Similarly, ADP significantly (p=0.000) decreased the serum level of total cholesterol and low-density lipoprotein. Conclusion: ADP may have the potential to improve dyslipidemia and obesity

    Preparation, Characterization, and Pharmacological Investigation of Withaferin-A Loaded Nanosponges for Cancer Therapy; In Vitro, In Vivo and Molecular Docking Studies

In parallel, nanotechnology approaches with various pharmacological properties offer efficacious clinical outcomes. The use of new artificial variants of nanosponges (NS) as a transporter of chemotherapeutic drugs to target cells has emerged as a very promising tool. Therefore, in this research, ethylcellulose (EC) NS were prepared using the ultrasonication assisted-emulsion solvent evaporation technique. Withaferin-A (WFA), an active ingredient in Withania somnifera, has been implanted into the nanospongic framework with enhanced anticancer properties. Inside the polymeric structure, WFA was efficiently entrapped (85 ± 11%). The drug (WFA) was found to be stable within polymeric nanosponges, as demonstrated by Fourier transform infrared (FTIR) spectroscopy and differential scanning calorimetry (DSC) studies. The WFA-NS had a diameter of 117 ± 4 nm and zeta potential of −39.02 ± 5.71 mV with a polydispersity index (PDI) of 0.419 ± 0.073. In addition, scanning electron microscopy (SEM) revealed the porous surface texture of WFA-NS. In vitro anticancer activity (SRB assay) results showed that WFA–NS exhibited almost twice the anticancer efficacy against MCF-7 cells (IC50 = 1.57 ± 0.091 µM), as quantified by flow cytometry and comet tests. Moreover, fluorescence microscopy with DAPI staining and analysis of DNA fragmentation revealed apoptosis as a mechanism of cancer cell death. The anticancer activity of WFA-NS was further determined in vivo and results were compared to cisplatin. The anticancer activity of WFA-NS was further investigated in vivo, and the data were consistent to those obtained with cisplatin. At Day 10, WFA-NS (10 mg/kg) significantly reduced tumour volume to 72 ± 6%, which was comparable to cisplatin (10 mg/kg), which reduced tumour volume to 78 ± 8%. Finally, the outcomes of molecular modeling (in silico) also suggested that WFA established a stable connection with nanosponges, generating persistent hydrophobic contacts (polar and nonpolar) and helping with the attractive delayed-release features of the formulation. Collectively, all the findings support the use of WFA in nanosponges as a prototype for cancer treatment, and opened up new avenues for increasing the efficacy of natural product-derived medications
