    Identity Formation in a Relational Context: A Person-Centered Analysis of Troubled Youth

    The purpose of this study was to examine the identity formation of troubled youth (8 female and 12 male adolescents, ages 14–17) in an after-school treatment program for problem behavior. To achieve this goal we (a) garnered information from 2 identity interviews given 6 to 8 weeks apart, (b) adopted a qualitative, person-centered analytical strategy to identify identity profiles, and (c) examined the identity profiles within a relational context, focusing on the developmentally salient domains of parents and peers. Analyses revealed 5 identity profiles similar to the identity statuses previously described by Marcia and others, but with unique phenomenological differences. Across the profiles, youth differed in their responses (in terms of exploration and commitment) to the “identity challenges” they encountered during this brief period. Likewise, the relational contexts of parental and peer support also varied across profiles

    Differences in extrinsic tree quality and value of fibre production following alternative vegetation management treatments in Northwestern Ontario

Forest Vegetation Simulator (FVSOntario) was used to project the total and merchantable volume to age 70 and BUCK-2 was used to optimize the resulting product mix. Projected value was based on 2009 prices for hog fuel, pulpwood and SPF (spruce–pine–fir) eastern green lumber prices. At 16 years posttreatment, gross total volumes in herbicide-treated and mechanically cut plots were significantly higher (120%–165% and 94%–98%, respectively) than that in control plots (14.73 m3 ha-1). Based on height, diameter, and taper criteria, observed tree quality did not differ among treatments. The projected value of the fibre produced was 36% to 53% higher in herbicide- treated plots and 24% to 37% higher in mechanically cut plots than in control plots ($18 486.76 per ha)

    Phylogenetics of Chloridoideae (Gramineae): a Preliminary Study Based on Nuclear Ribosomal Internal Transcribed Spacer and Chloroplast trnL–F Sequences

    The phylogeny of Chloridoideae (Gramineae) was inferred from parsimony analyses of DNA sequences from two genomes—the chloroplast trnL intron, trnL 3\u27 exon, and trnL–F intergenic spacer, and the nuclear ribosomal internal transcribed spacer region (ITS1 + 5.8S + ITS2). Eighty species representing 66 chloridoid genera were sampled, including all but four of the native New World genera. Analyses of the individual and combined data sets were performed. The phylogenies were found to be highly congruent. Of the four tribes and seven subtribes of Chloridoideae sensu Clayton and Renvoize (1986) whose phylogenetic status could be tested with our taxon sample, only Orcuttieae and Uniolinae were monophyletic. The phylogenies suggested signiïŹcant homoplasy in morphological traits, including inïŹ‚orescence type, number of ïŹ‚orets per spikelet, and number of lemma nerves. We propose a new classiïŹcation based on the three main clades in the phylogenies—tribes Cynodonteae, Eragrostideae, and Zoysieae. The Eragrostideae clade is well resolved and supported and is further divided into three subtribes, Cotteinae, Eragrostidinae, and Uniolinae. Cynodonteae include most of the genera in our study, but the clade is poorly resolved. However, a clade formed of Muhlenbergia and nine other genera is present in both phylogenies and is well resolved and supported. A number of interesting, well-supported relationships are evident in the phylogenies, including Pappophorum–Tridens ïŹ‚avus, Tragus–Willkommia, and Gouinia–Tridens muticus–Triplasis–Vaseyochloa. Except for Bouteloua, no genus represented by multiple species proved to be monophyletic in the phylogenies

    Modulating GLUT1 Expression in Retinal Pigment Epithelium Decreases Glucose Levels in the Retina: Impact on Photoreceptors and MĂŒller Glial Cells

    The retina is one of the most metabolically active tissues in the body and utilizes glucose to produce energy and intermediates required for daily renewal of photoreceptor cell outer segments. Glucose transporter 1 (GLUT1) facilitates glucose transport across outer blood retinal barrier (BRB) formed by the retinal pigment epithelium (RPE) and the inner BRB formed by the endothelium. We used conditional knockout mice to study the impact of reducing glucose transport across the RPE on photoreceptor and MĂŒller glial cells. Transgenic mice expressing Cre recombinase under control of the Bestrophin1 (Best1) promoter were bred with Glut1 flox/flox mice to generate Tg-Best1-Cre:Glut1 flox/flox mice (RPE∆Glut1). The RPE∆Glut1 mice displayed a mosaic pattern of Cre expression within the RPE that allowed us to analyze mice with ~50% (RPE∆Glut1 m ) recombination and mice with \u3e70% (RPE∆Glut1 h ) recombination separately. Deletion of GLUT1 from the RPE did not affect its carrier or barrier functions, indicating that the RPE utilizes other substrates to support its metabolic needs thereby sparing glucose for the outer retina. RPE∆Glut1 m mice had normal retinal morphology, function, and no cell death; however, where GLUT1 was absent from a span of RPE greater than 100 ”m, there was shortening of the photoreceptor cell outer segments. RPE∆Glut1 h mice showed outer segment shortening, cell death of photoreceptors, and activation of MĂŒller glial cells. The severe phenotype seen in RPE∆Glut1 h mice indicates that glucose transport via the GLUT1 transporter in the RPE is required to meet the anabolic and catabolic requirements of photoreceptors and maintain MĂŒller glial cells in a quiescent state. © 2019, American Physiological Society.     This article considers the notion that to document or inscribe our lives not only leaves a trace of our creaturely presence, but may also become a form of juris-writing, a writing that concerns and aims at Justice. Employing an expanded notion of Justice that takes it beyond the institutions of law, therefore, it asks about forms of documentality (Ferraris) that put us ‘before memory’ in Derrida’s sense. How is it possible to think curation in relation to a violent past in such a way that neither attempts to deny the lacunae nor surrenders in the face of the difficulties of such attempts? How should we consider the relation between the delimited encounter with an ‘invitation to imagine’ (Didi-Huberman) and processes of institutionalisation that build a society? What about those things that it is not possible to show, including relations of power, that arise analytically? Reflecting on current memory spaces, especially within ex-clandestine centres for detention, torture and extermination (ex-ccdtyes) in Argentina, the article offers five theses in order to consider what is at stake in the encounters staged at these sites

    Microtubule-Associated Protein 1 Light Chain 3B, (LC3B) is Necessary to Maintain Lipid-Mediated Homeostasis in the Retinal Pigment Epithelium

    Like other neurons, retinal cells utilize autophagic pathways to maintain cell homeostasis. The mammalian retina relies on heterophagy and selective autophagy to efficiently degrade and metabolize ingested lipids with disruption in autophagy associated degradation contributing to age related retinal disorders. The retinal pigment epithelium (RPE) supports photoreceptor cell renewal by daily phagocytosis of shed photoreceptor outer segments (OS). The daily ingestion of these lipid-rich OS imposes a constant degradative burden on these terminally differentiated cells. These cells rely on Microtubule-Associated Protein 1 Light Chain 3 (LC3) family of proteins for phagocytic clearance of the ingested OS. The LC3 family comprises of three highly homologous members, MAP1LC3A (LC3A), MAP1LC3B (LC3B), and MAP1LC3C (LC3C). The purpose of this study was to determine whether the LC3B isoform plays a specific role in maintaining RPE lipid homeostasis. We examined the RPE and retina of the LC3B-/- mouse as a function of age using in vivo ocular imaging and electroretinography coupled with ex vivo, lipidomic analyses of lipid mediators, assessment of bisretinoids as well as imaging of lipid aggregates. Deletion of LC3B resulted in defects within the RPE including increased phagosome accumulation, decreased fatty acid oxidation and a subsequent increase in RPE and sub-RPE lipid deposits. Age-dependent RPE changes included elevated levels of oxidized cholesterol, deposition of 4-HNE lipid peroxidation products, bisretinoid lipofuscin accumulation, and subretinal migration of microglia, collectively likely contributing to loss of retinal function. These observations are consistent with a critical role for LC3B-dependent processes in the maintenance of normal lipid homeostasis in the aging RPE, and suggest that LC3 isoform specific disruption in autophagic processes contribute to AMD-like pathogenesis. © 2018 Dhingra, Bell, Peachey, Daniele, Reyes-Reveles, Sharp, Jun, Bazan, Sparrow, Kim, Philp and Boesze-Battaglia

    Advancing Alternative Analysis: Integration of Decision Science.

    Decision analysis-a systematic approach to solving complex problems-offers tools and frameworks to support decision making that are increasingly being applied to environmental challenges. Alternatives analysis is a method used in regulation and product design to identify, compare, and evaluate the safety and viability of potential substitutes for hazardous chemicals.Assess whether decision science may assist the alternatives analysis decision maker in comparing alternatives across a range of metrics.A workshop was convened that included representatives from government, academia, business, and civil society and included experts in toxicology, decision science, alternatives assessment, engineering, and law and policy. Participants were divided into two groups and prompted with targeted questions. Throughout the workshop, the groups periodically came together in plenary sessions to reflect on other groups' findings.We conclude the further incorporation of decision science into alternatives analysis would advance the ability of companies and regulators to select alternatives to harmful ingredients, and would also advance the science of decision analysis.We advance four recommendations: (1) engaging the systematic development and evaluation of decision approaches and tools; (2) using case studies to advance the integration of decision analysis into alternatives analysis; (3) supporting transdisciplinary research; and (4) supporting education and outreach efforts

    The Extragalactic Distance Scale Key Project XXVII. A Derivation of the Hubble Constant Using the Fundamental Plane and Dn-Sigma Relations in Leo I, Virgo, and Fornax

    Using published photometry and spectroscopy, we construct the fundamental plane and D_n-Sigma relations in Leo I, Virgo and Fornax. The published Cepheid P-L relations to spirals in these clusters fixes the relation between angular size and metric distance for both the fundamental plane and D_n-Sigma relations. Using the locally calibrated fundamental plane, we infer distances to a sample of clusters with a mean redshift of cz \approx 6000 \kms, and derive a value of H_0=78+- 5+- 9 km/s/Mpc (random, systematic) for the local expansion rate. This value includes a correction for depth effects in the Cepheid distances to the nearby clusters, which decreased the deduced value of the expansion rate by 5% +- 5%. If one further adopts the metallicity correction to the Cepheid PL relation, as derived by the Key Project, the value of the Hubble constant would decrease by a further 6%+- 4%. These two sources of systematic error, when combined with a +- 6% error due to the uncertainty in the distance to the Large Magellanic Cloud, a +- 4% error due to uncertainties in the WFPC2 calibration, and several small sources of uncertainty in the fundamental plane analysis, combine to yield a total systematic uncertainty of +- 11%. We find that the values obtained using either the CMB, or a flow-field model, for the reference frame of the distant clusters, agree to within 1%. The Dn-Sigma relation also produces similar results, as expected from the correlated nature of the two scaling relations. A complete discussion of the sources of random and systematic error in this determination of the Hubble constant is also given, in order to facilitate comparison with the other secondary indicators being used by the Key Project.Comment: 21 pages, 3 figures, Accepted for publication in Ap
