    Development of a New Measurement Method to Evaluate the Physical Properties of Granules for Dry Powder Inhalation Produced by New Spouted Bed Type Binderless Granulator

    The spouted bed type of binderless granulator is one of the most appropriate methods available for producing granules of dry powder inhalation (DPI) from cohesive fine powder. However the compressive strength of produced granule is too small to be measure by conventional method. In this work a new apparatus was developed to evaluate the strength of such a soft granule. Load and displacement curve of the granule were measured by new method of compressive strength measurement. To observe the compression process of the granule, photographs were taken with a microscopic camera. It was clarified that this measurement method was remarkably useful to evaluate the compressive strength of soft granule

    Myt1 protein kinase is essential for Golgi and ER assembly during mitotic exit

    Myt1 was originally identified as an inhibitory kinase for Cdc2 (Cdk1), the master engine of mitosis, and has been thought to function, together with Wee1, as a negative regulator of mitotic entry. In this study, we report an unexpected finding that Myt1 is essential for Golgi and endoplasmic reticulum (ER) assembly during telophase in mammalian cells. Our analyses reveal that both cyclin B1 and cyclin B2 serve as targets of Myt1 for proper Golgi and ER assembly to occur. Thus, our results show that Myt1-mediated suppression of Cdc2 activity is not indispensable for the regulation of a broad range of mitotic events but is specifically required for the control of intracellular membrane dynamics during mitosis

    Depletion of Primordial Germ Cells (PGCs) by X-irradiation to Extraembryonic Region of Chicken Embryos and Expression of Xenotransplanted Quail PGCs

    The generation of germline chimeras by the transfer of primordial germ cells (PGCs) requires incorporation of the PGCs of the donor into the gonadal tissue of the recipient embryo. We investigated the utility of soft x-irradiation with application of a lead (12-3 x 0.25 mm, similar to 0.1 g) shield to the embryo proper for the production of chicken-quail germline chimeras. Chicken embryos shielded during irradiation for 120 s (similar to 7.2 Gy) at stages 13 to 17 showed a hatchability of 35% (106/301). whereas the hatchability of unshielded embryos was 26% (27/105). The relative Population of gonadal PGCs Lit stage 30 for embryos irradiated at stage 13 with or Without shielding was 13 and 5%. respectively, of the value for nonirradiated controls. Chicken embryos irradiated at stages 13 or 14 with or Without shielding and transfused with quail embryonic blood containing PGCs each exhibited similar to 130 relative population of donor PGCs in the left gonad at stage 30. Xenotransplanted hatchlings exhibited donor-derived PGCs as detected by Southern hybridization and PCR. Exposure of chicken embryos to similar to 7.2 Gy of x-radiation at stage 13 with the application of a lead shield to the embryo proper is thus a feasible approach to depletion of endogenous germ cells and the production of chicken-quail germline chimeras.ArticleJOURNAL OF POULTRY SCIENCE. 46(2): 136-143(2009)journal articl

    Gamma Band Neural Synchronization Deficits for Auditory Steady State Responses in Bipolar Disorder Patients

    Periodic auditory click stimulation has been reported to elicit an auditory steady state response (ASSR). The ASSR has been suggested to reflect the efficiency of γ-amino butyric acid (GABA) inhibitory interneuronal activity. Although a potential role for GABAergic dysfunction has been previously proposed, the role of neural synchronization in the ASSR in people with bipolar disorder (BD) has received little attention. In the current study, we investigated ASSRs to 20 Hz, 30 Hz, 40 Hz and 80 Hz click trains in BD patients. A total of 14 (4 males) BD patients and 25 (10 males) healthy controls participated in this study. ASSRs were obtained using whole-head 306-channel magnetoencephalography to calculate, ASSR power values and phase locking factors (PLF). BD patients exhibited significantly reduced mean ASSR power and PLF values bilaterally at frequencies of 30, 40, and 80 Hz (p<0.05 for these frequencies). At 20 Hz, bipolar patients showed no significant reduction in mean ASSR power and PLF values. There was a significant negative correlation between 80 Hz-ASSR-power values obtained from the right hemisphere and scores on the Hamilton Depression Rating Scale (rho = −0.86, p = 0.0003). The current study showed reduced low and high gamma band ASSR power and PLF bilaterally with no significant beta band ASSR reduction in BD patients. BD patients are characterized by deficits in gamma band oscillations, which may be associated with GABA inhibitory interneuronal activity dysfunction

    Distribution of Faraday Rotation Measure in Jets from Active Galactic Nuclei II. Prediction from our Sweeping Magnetic Twist Model for the Wiggled Parts of AGN Jets and Tails

    Distributions of Faraday rotation measure (FRM) and the projected magnetic field derived by a 3-dimensional simulation of MHD jets are investigated based on our "sweeping magnetic twist model". FRM and Stokes parameters were calculated to be compared with radio observations of large scale wiggled AGN jets on kpc scales. We propose that the FRM distribution can be used to discuss the 3-dimensional structure of magnetic field around jets and the validity of existing theoretical models, together with the projected magnetic field derived from Stokes parameters. In the previous paper, we investigated the basic straight part of AGN jets by using the result of a 2-dimensional axisymmetric simulation. The derived FRM distribution has a general tendency to have a gradient across the jet axis, which is due to the toroidal component of the magnetic field generated by the rotation of the accretion disk. In this paper, we consider the wiggled structure of the AGN jets by using the result of a 3-dimensional simulation. Our numerical results show that the distributions of FRM and the projected magnetic field have a clear correlation with the large scale structure of the jet itself, namely, 3-dimensional helix. Distributions, seeing the jet from a certain direction, show a good matching with those in a part of 3C449 jet. This suggests that the jet has a helical structure and that the magnetic field (especially the toroidal component) plays an important role in the dynamics of the wiggle formation because it is due to a current-driven helical kink instability in our model.Comment: Accepted for publication in Ap

    Acute Polyradiculoneuropathy Associated With Salmonella Gastroenteritis

    We reported a case of acute polyradiculoneuropathy associated with Salmonella gastroenteritis. A68-year-oldman developed progressive motor weakness and areflexia following the febrile illness and diarrhea caused by a strain of Salmonella species O8 group. He showed a rapid and complete recovery from the illness. This is the first report in which Salmonella gastroenteritis might be etiologically related to an acute polyradiculoneuropathy

    Extraction of the pectic substances from a marine alga, Porphyra tenera, was attempted. The pectic substances were separated into three fractions (water-soluble, sodium hexametaphosphate-soluble, and hydrochloric acid-soluble). In the course of the separation, it was found that there are two kinds of water-soluble pectic substances one is easy to extract with water, the other is not so easily extractable. The extraction of water-soluble fraction and sodium hexametaphosphate-soluble fraction from the sample took very long period. Extraction of the pectic substances from laver was not so easy as vegetables

    Abnormal Liver Function in Patients with Sjogren\u27s Syndrome

    We measured the liver function tests of 145 patients with Sjogren\u27s syndrome (SjS) (75 patients with primary SjS, 70 patients with secondary SjS), and characterized the SjS patients with abnormal liver function tests from several points of view : 1, the incidence of them in the primary SjS comparing with that in secondary SjS. 2, the staining pattern of anti-nuclear antibodies, and 3, the existence of antihepatitis C virus (HCV) antibody, hepatitis B surface (HBs) antigen, and antibody against human T-lymphotropic virus type I (HTLV-I). Abnormal liver function tests were detected in 38 out of 145 patients (26.2%) with SjS. Fifteen of the 38 patients (20.0%) had primary SjS while the remaining patients (32.9%) had secondary SjS. Histopathological examination identified primary biliary cirrhosis (PBC) in 2 patients, autoimmune hepatitis in 4 patients, and autoimmune cholangitis in a single patient with SjS. No significant difference in the presence of antinuclear antibody (ANA) was found between SjS patients with and without abnormal liver function tests. However, the incidence of discrete speckled pattern was significantly higher in SjS patients with abnormal liver function than in the patients with normal liver function. Two sera showing cytoplasmic pattern of ANA were also positive for anti-mitochondrial M2 antibody, allowing the diagnosis of PBC. All 11 sera exhibiting discrete speckled pattern contained significant amounts of anti-centromere antibody. Abnormal liver function tests were detected in 8 of 11 sera with these antibodies, 2 patients with PBC, 2 patients with autoimmune hepatitis, one patient with autoimmune cholangitis, one patient with chronic hepatitis B and 2 other patients with unconfirmed diagnosis. The percentages of anti-HCV antibody-positive, HBs-Agpositive and anti-HTLV-I antibody-positive in sera of patients were higher than those of blood donors from the same geographical area. However, no significant difference was seen of these percentages in sera between the patients with and without abnormal liver function. Taken together, present study indicated that SjS patients with anti-centromere antibody may have some susceptibility for acquiring autoimmune liver disease

    Using the numerical data of MHD simulation for AGN jets based on our ``sweeping magnetic twist model'', we calculated the Faraday rotation measure (FRM) and the Stokes parameters to compare with observations. We propose that the FRM distribution can be used to discuss the 3-dimensional structure of magnetic field around jets, together with the projected magnetic field derived from the Stokes parameters. In the present paper, we supposed the basic straight part of AGN jet, and used the data of axisymmetric simulation. The FRM distribution we derived has a general tendency to have gradient across the jet axis, which is due to the toroidal component of the helical magnetic field generated by the rotation of the accretion disk. This kind of gradient in the FRM distribution is actually observed in some AGN jets (e.g. Asada et al. 2002), which suggests helical magnetic field around the jets and thus supports our MHD model. Following this success, we are now extending our numerical observation to the wiggled part of the jets using the data of 3-dimensional simulation based on our model in the following paper.Comment: Accepted for publication in Ap
