    The direction of the breeding process in a herd of Ayrshire breed cows in the conditions of the Vologda region

    The direction of the breeding process in herds and populations of dairy cattle breeds based on the actual indicators and variability of the trait is one of the main factors that allows to determine the effectiveness of breeding measures. As the initial material for the research there were taken the data of breeding records (n=681) of Ayrshire breed cows of the agricultural complex "Maysky" of the Vologda region. The number of cows was distributed by generations (195 cows with the first incomplete lactation, 177 cows with the first completed lactation, 130 cows for the second lactation, 179 cows for the third lactation and older). According to the research results, the superiority of the actual indicators for the live weight of heifers in animals of modern generation in early age periods was from 28 to 39 kg. Consequently, with each new generation, animals have better development indicators, as evidenced by the indicators of variation (variability) of the live weight of heifers in the early age periods of weak and moderate Cv = 5.3 to 10.6 %, depending on generation. The reduction of the age of the first insemination of heifers to 13.4 months (-2.2 months) and the first fruitful insemination to 14.0 months (-2.1 months) in animals from full-age to the first incomplete lactation was revealed. A targeted selection of cows was carried out in the herd according to the level of milk productivity of mothers after the first lactation - the selection efficiency was 977 kg of milk, according to the highest - 456 kg. The selection of breeding bulls was carried out according to the level of productivity of the breeding stock, however, it should be noted that the intensity of selection in cows for the incomplete first (10968 kg of milk) and first lactation (10635 kg of milk) is reduced compared to cows for the second lactation (11417 kg of milk). Consequently, in herds and populations of dairy cattle breeds, in order to achieve genetic progress in productivity in the next generation, it is necessary to determine the direction of the breeding process

    Uakitite, VN, a new mononitride mineral from uakit iron meteorite (IIAB)

    Uakitite was observed in small troilite–daubréelite (±schreibersite) inclusions (up to 100 µm) and in large troilite–daubréelite nodules (up to 1 cm) in Fe-Ni-metal (kamacite) of the Uakit iron meteorite (IIAB), Republic of Buryatia, Russia. Such associations in the Uakit meteorite seemed to form due to high-temperature (>1000 °C) separation of Fe-Cr-rich sulfide liquid from Fe-metal melt. Most inclusions represent alternation of layers of troilite and daubréelite, which may be a result of solid decay of an initial Fe-Cr-sulfide. These inclusions are partially resorbed and mainly located in fissures of the meteorite, which is now filled with magnetite, and rarely other secondary minerals. Phase relations indicate that uakitite is one of the early minerals in these associations. It forms isometric (cubic) crystals (in daubréelite) or rounded grains (in schreibersite). The size of uakitite grains is usually less than 5 µm. It is associated with sulfides (daubréelite, troilite, grokhovskyite), schreibersite and magnetite. Carlsbergite CrN, a more abundant nitride in the Uakit meteorite, was not found in any assemblages with uakitite. Physical and optical properties of uakitite are quite similar to synthetic VN: yellow and transparent phase with metallic luster; Mohs hardness: 9–10; light gray color with a pinky tint in reflected light; density (calc.) = 6.128 g/cm3. Uakitite is structurally related to the osbornite group minerals: carlsbergite CrN and osbornite TiN. Structural data were obtained for three uakitite crystals using the electron backscatter diffraction (EBSD) technique. Fitting of the EBSD patterns for a synthetic VN model (cubic, Fm-3m, a = 4.1328(3) Å; V = 70.588(9) Å3; Z = 4) resulted in the parameter MAD = 0.14–0.37° (best-good fit). Analytical data for uakitite (n = 54, in wt. %) are: V, 71.33; Cr, 5.58; Fe, 1.56; N, 21.41; Ti, below detection limit (<0.005). The empirical formula (V0.91Cr0.07Fe0.02)1.00N1.00 indicates that chromium incorporates in the structure according to the scheme V3+ → Cr3+ (up to 7 mol. % of the carlsbergite end-member). © 2020 by the authors.     The paper presents the materials of the complex epidemiological evaluation of tuberculosis process by its main indicators (incident cases, prevalence and mortality) in the Siberian Federal district over the period of 2002-2014. The ranking of the territories included in the Siberian Federal district was performed by the integral index. That allowed to identify three groups of territories by the level of intensity of TB epidemiological process (favorable, typical, unfavorable). The most favorable epidemiological situation was registered in Tomsk region, the most unfavorable one - in the Republic of Tuva

    Molecular epidemiology of multidrug resistant tuberculosis in Mongolia and Eastern Siberia: two independent dissemination processes for dominant strains

    Mongolia and Russia are among the countries with the high tuberculosis (TB) burden. The prevalence of tuberculosis, including multidrug-resistant tuberculosis (MDR), in Eastern Siberia bordering Mongolia is significantly higher than in the European part of Russia. In addition, unlike Mongolia, Eastern Siberia is characterized by a high prevalence of HIV infection. The cross-border spread of socially significant infections in these countries seems to occur due to their wide-range cooperation and cultural exchange. Whereas the HIV infection has no epidemiological significance for Mongolia at the moment, tuberculosis, however, has a similar prevalence on both sides of the border. The aim was to evaluate the cross-border MDR M. tuberculosis distribution in Mongolia and Eastern Siberia by using molecular genetic data. Materials and methods. A total of 1045 M. tuberculosis strains isolated in Mongolia (291) and the three regions of Eastern Siberia (754) were studied by using the MIRU-VNTR-24 loci genotyping. The CC2/W148 and CC1 subtypes were identified by the specific deletion in the kdpD gene and SNP in the pks17 gene at position 1887060, respectively. Phylogenetic analysis of MIRU-VNTR patterns was carried out by generating UPGMA tree and maximum likelihood tree. Results. The Beijing genotype was found in 75.3% (219/291) and 69.0% (520/754) from Mongolian and East Siberian collection, respectively. Common minor genotypes were LAM (11.0% and 15.1%), T (10.3% and 4.5%), and Haarlem (1.4% and 2.4%) found in Mongolia and Eastern Siberia, respectively. The genotypes S (1.3%) and Ural (5.0%) were found solely in the Russia-derived samples. The main epidemic Beijing subtypes in each country belonged to different clonal complexes (CC): the majority of Mongolian Beijing strains displayed profiles 342-32, 3819-32, 1773-32 MLVA types and belonged to the CC4 subtype; Russian Beijing strains mainly belonged to the CC1 (43.7% — 227/520) and CC2/W148 (34.8% — 181/520) subtypes. The MDR level and distribution patterns differed significantly between Mongolia and Eastern Siberia. Modeling of Beijing strain expansion evidences about extremely subtle contribution of the M. tuberculosis cross-border transmission between Mongolia and Russia. The phylogenetic reconstruction of Beijing CC4 subtype evolution in Mongolia suggests that its distribution is primarily associated with China and other countries of the Western Pacific Region. Three main phylogenetic branches of CC4 subtype were traced, which probably spread throughout Mongolia in the 11—12th centuries. It may be assumed that spreading of the epidemic Beijing CC4 subtype might occur in two stages: early period — emergence of ancestral CC4 variants in Mongolia or their introduction from China (they are homologous to the strains preserved in the Chinese population), later period — dissemination due to the active exchange of M. tuberculosis with countries of Southeast Asia, but not Russia. Conclusion. Using MIRU-VNTR-24 genotyping as well as classification according to specific single nucleotide polymorphisms specific to certain Beijing subtypes, it allowed to describe separate patterns of the epidemic variants spread in Mongolia and Russia. It has been demonstrated that emergence and spread of MDR-TB in Mongolia are entirely iatrogenic in nature, while the epidemic subtypes of the Beijing genotype (subtypes CC1 and CC2/W148) contribute markedly into the MDR-TB spreading in Eastern Siberia

    Genotypic and phenotypic characteristics of <i>Mycobacterium tuberculosis</i> drug resistance in TB children

    Background. Russian Federation is included in the list of 30 countries with the highest burden of tuberculosis, including MDR tuberculosis. The most important part of this problem is the primary MDR/XDR TB in children.The aim: a comparative analysis of the phenotypic and genotypic profile of drug resistance to anti-tuberculosis drugs (ATP) according to whole genome sequencing of M. tuberculosis strains from children.Materials and methods. Whole genome sequencing (WGS) results of 61 M. tuberculosis isolates from children with tuberculosis in 2006–2020 in the Russian Federation were analyzed for anti-TB drug resistance mutations, according to the WHO catalog and were compared with the results of phenotypic drug sensitivity.Results. The M. tuberculosis belonged to two genetic groups: Beijing genotype – 82 % (50/61) dominant Central Asian Russian (31/50) and B0/W148 (16/50) subtypes, and non-Beijing (Ural, S, LAM) – 18 % (11/61). Three isolates belonged to Asian Ancestral subtype (3/50). Of the 61 isolates, only 14.7 % (9/61) were sensitive to antiTB drugs, 49.2 % (30/61) were MDR and 14.7 % (9/61) were pre-XDR. Comparison of the resistance profile (MDR/pre-XDR) with genotype revealed an upward shift for Beijing isolates, in particular Beijing B0/W148 (15/16) subline compared to other Beijing (19/34) (Chi-square with Yates correction = 5.535; p &lt; 0.05) and nonBeijing (5/12) (Chi-square with Yates correction = 6.741; p &lt; 0.05) subtypes. Discrepancies between genotypic and phenotypic drug resistance profiles were found in 11.5 % (7/61) of cases.Conclusions. Based on the analysis of WGS data, the genotypic characteristics of M. tuberculosis and the most complete set of drug resistance mutations were obtained, indicating a significant prevalence in MDR and pre-XDR TB of cases caused by epidemic subtypes of Beijing (B0/W148 and Central Asian Russian). The molecular mechanisms of adaptation of M. tuberculosis to the treatment of anti-TB drugs are not unique for the child population but reflect the general processes of the spread of MDR/XDR in Russia

    The cytoplasmic actins (β and γ) play crucial roles during key cellular processes like adhesion, migration, polarization and cytokinesis. The understanding of their specific underlying mechanisms would be of major relevance not only for fundamental research but also for clinical applications, since modulations of actin isoforms are directly or indirectly correlated with severe pathologies. The major goal of the research was to elucidate the function of the actin isoforms during motile activities, adhesions and cell division and to investigate whether their expression and/or structural organization is related to pathological function. Selective depletion of β- and γ-cytoplasmic actins allowed attributing functional diversities of β- and γ-сytoplasmic actins. β-Сytoplasmic actin plays a preferential role in contractile activities, whereas γ-cytoplasmic actin mainly participates in the formation of a submembranous network necessary for cell shape flexibility and motile activity. The roles of isoforms in regulating the integrity of adherens and tight junctions respectively were demonstrated. Unique roles of β- and γ-cytoplasmic actins in normal cells were shown. Similar results were obtained in cancer cells compared with normal epithelial cells in culture and in human pathological tissue sections of mammary gland, colon, lung and cervix. Malignant cell transformation requires changes in the ability of cells to migrate. The disruption of actin cytoskeleton and intercellular adhesions is an important component of the acquisition of invasive properties in epithelial malignancies.Цитоплазматические изоформы актина (β и γ) играют важную роль в ключевых клеточных процессах, таких как адгезия, миграция, поляризация и цитокинез. Понимание специфических механизмов, лежащих в основе этих процессов, является связующим звеном между фундаментальными и клиническими исследованиями, так как модуляции актиновых изоформ прямо или косвенно связаны с различными патологиями. Исследованию функций цитоплазматических изоформ актина, связанных с подвижностью и делением нормальных и опухолевых клеток, адгезионными структурами, а также изучению соответствия их экспрессии и / или структурной организации нормальным и патологическим функциям клеток посвящен данный обзор. Селективная редукция β- или γ-цитоплазматических актинов позволила определить функциональные различия между этими изоформами. Преимущественную роль в сократительных и адгезионных активностях играет β-актин, тогда как цитоплазматический γ-актин участвует в образовании подмембранной сети, необходимой для клеточной пластичности и подвижности. Определяющую роль в установлении и поддержании нормальной архитектуры и динамики эпителиальных плотных и адгезионных межклеточных контактов играет связь с актиновым цитоскелетом. Продемонстрирована уникальная роль β- и γ-актинов в регуляции и поддержании целостности адгезионных и плотных межклеточных контактов соответственно. Похожие результаты были получены при сравнении опухолевых клеток с нормальными эпителиальными клетками в культуре и на срезах патологических тканей молочной железы, кишечника, легких и шейки матки человека. Изоформ-специфичная перестройка актинового цитоскелета и адгезионных межклеточных контактов является важным шагом в приобретении инвазивности эпителиальными опухолями

    Viral Load in COVID-19: Underestimated Clinical and Epidemiological Marker

    Background. The viral load of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of the new coronavirus infection, is becoming increasingly important in clinical and epidemiological contexts. Despite this, there are significant complexities in the implementation of viral load quantitative measurement into clinical practice due to the limited approaches to its assessment.The aim of this work was to develop an approach for SARS-CoV-2 viral load analysis by the value of sample threshold cycles (Ct) relative to the Ct of the internal control sample obtained in routine PCR diagnostics of the COVID-19, and to use this approach for quantitative monitoring of viral load in patients with first positive SARS-CoV-2 test from the Irkutsk region.Materials and methods. Using regression models based on the least squares method, an approach to determine the number of copies of SARS-CoV-2 RNA in 1 ml of nasopharyngeal secretion was developed. The viral load of SARS-CoV-2 was assessed in nasopharyngeal and pharyngeal samples obtained from 1370 patients from Irkutsk and Angarsk with primary diagnosed positive PCR result in the period from July 1 to November 10, 2020.Results. A tenfold increase in the average monthly viral load among patients in September-October 2020 was revealed. We assume that the change in the epidemiological pattern of the spread of the new coronavirus infection during this period is associated with an increase in the number of contacts in the population due to the school year beginning. Higher viral loads are observed in populations at risk for COVID-19 – among healthcare workers and adults/elderly patients. Conclusion. The development of a standardized quantification of SARS-CoV-2 viral load in the nasopharyngeal samples can be a predictive clinical marker and a reliable tool for improving COVID-19 surveillance using the proposed approach to assess average viral load in a local population

    Vasoprotective effect of effective lipid-lowering therapy in patients with <i>ST</i>-segment elevation myocardial infarction

    Aim. To study the vasoprotective effects of atorvastatin depending on the achievement of the target level of low-density lipoprotein cholesterol (LDL-C) in patients with ST-segment elevation myocardial infarction (STEMI) within 48 weeks of follow-up. Materials and methods. Included were 112 STEMI patients who received atorvastatin 204080 mg. On days 79 from the onset of the disease, after 24 and 48 weeks, ultrasound examination of the carotid arteries with RF technology and applanation tonometry were performed, the lipid profile was determined. The patients were divided into groups: group 1 (n=41) of highly effective therapy (HET) who achieved the target LDL-C after 24 and 48 weeks; group 2 (n=29) in relatively effective therapy (RET) achieving target values at 24th or 48th week; group 3 (n=42) insufficiently effective therapy (IET) did not reach the target LDL-C. Results. When examining the carotid arteries in the HET group, the intima-media thickness (IMT) decreased by 10.713.1%, the b index by 14.926.3% after 2448 weeks. In the RET group, the IMT regression was 10.413.3%; b index 23.9% by the 48th week. In the IET group, the b index decreased by the 48th week by 14.3%. According to applanation tonometry in the HET group, the central pressure did not change. In the RET group, systolic pressure in the aorta increased by 1015.7% after 2448 weeks, pulse pressure by 33.9% by the end of observation. With IET, the increase was 8.66.8 and 19.825.9%, respectively. The odds ratio of developing endpoints in the RET group was 4.7 (95% CI 1.226.4; p=0.02), in the IET group 3.9 (95% CI 1.124.8; p=0.03) compared with HET. Conclusion. The most pronounced vasoprotective effect and a decrease in cardiovascular risk are associated with the achievement of the target LDL-C throughout the entire treatment period

    Online service for interpretation of the resistance prediction results to bedaquiline by the molecular data

    Background. Bedaquiline is a new and promising anti-tuberculosis drug, but longterm use requires resistance. This is due to mutations in the atpE and mmpR genes in M. tuberculosis (MBT).The aim of the research was to test a system for automated interpretation of results for predicting resistance to bedaquiline by the molecular data.Materials and methods. DNA was isolated from strains of M. tuberculosis in the Irkutsk region and Yakutia. The total quantity of DNA samples was 27 strains from Yakutia and 21 strains from the Irkutsk region. The study of MBT genomes was carried out on the DNA previously obtained by the authors in the territories of the Irkutsk region (n = 5), Yakutia (n = 4), Buryatia (n = 3), Zabaykalskiy kray (n = 4) and the Far East (n = 8). We used the BSATool program to detect bedaquiline resistance based on  Sanger and genomic data. Sanger sequencing analyzed the atpE and  mmpR genes, and whole genome sequencing examined mutations in the same sequences, as well as additionally in mmpL5, mmpS5, Rv0678, Rv1979c, and pepQ.Results. Complete agreement between the phenotypic and genotypic analysis of resistance to bedaquiline was found for three strains from Yakutia. One genome with significant mutations to bedaquiline was identified. A conclusion was made about the importance of molecular analysis of target genes with subsequent detection of resistance to bedaquiline in silico
