    ON THE DIAGNOSTICS OF SAGITTAL OCCLUSION ANOMALIES

    In the known classifications, there is no terminological wording as asymmetrical sagittal occlusion of segments of dentition in lateral areas. The work complemented classification of sagittal occlusion anomalies in the lateral areas of dentition of following types of asymmetric occlusion: Type I of asymmetrical sagittal occlusion: correct (neutral - class 1] ratio of lateral teeth on one side and distal (class 2) ratio on the other side. Type II of asymmetrical sagittal occlusion: correct (neutral - class 1] ratio of lateral teeth on one side and mesial (class 3) ratio on the other side. Type III of asymmetrical sagittal occlusion: distal (class 2] ratio of lateral teeth on one side and mesial (class 3) ratio on the other side. Type of asymmetrical sagittal occlusion is not the basis for the establishment of gravity anomalies

    Clinical and roentgenological aspects of diagnosis and treatment of temporomandibular joint pathology

    The article presents the occurrence of temporomandibular joint pathology (20,9 %) in children of prepubertal age. It was revealed that habitual dislocation that appears on the background of asymmetric occlusion with close position of teeth in frontal and lateral areas of dentition is typical for this age group. It was determined that clinical signs of TM] pathology in children and adults are different: pain syndrome and noise effect at the lower jaw movements are not typical for children in contrast to the adults. According to the data of outpatient's cards the structure of nosologic forms of pathology during different periods of occlusion formation is defined. It was determined that TM] pathology begins to form in preschool age and later on its frequency increases. The analysis of morbidity by sex showed that women have TM] pathology more often and also they have age tendency to the aggravation of the pathology that develops in future in appearance of dislocations of intraarticular disks. The article suggests the differentiated approach tactics for the treatment of TM] diseases that was rather effective

    Socio-demographic factors of social development in the conditions of post-global reality

    The article considers the system-wide impact of socio-demographic factors in the social development of countries and regions in the context of post-global reality. It is emphasized that modern demographic changes are the result of global transformation processes. The permanent relationship of demographic processes with climate change on the planet, the emergence and spread of new infectious diseases, the development of digital technologies, etc. is shown. The meaning of the concept of “post-global reality” is clarified in terms of the uniqueness of modern conditions of social development, in which the desire of a person and society for “openness” is traced in the conditions of growing “social closeness” and “social tension”, which indicates the limited possibilities of society in regulating sociodemographic processes. It is concluded that the spread of new infectious diseasesin the 21st century, especially the new coronavirus infection COVID-19, has contributed to the emergence of new groups of problems associated with a deterioration in health status, an increase in mortality, a decrease in the reproductive and labor potential of the population. The authors substantiate the formation of the concept of “global determinism” in terms of the role of socio-demographic factors in the social development of countries and regions in the context of post-global reality. Among the most influential concepts that contribute to its development are “geographical determinism”, “economic determinism” and actually “demographic determinism”. Based on the materials of a sociological study, it is shown that new external and internal challenges and new crises of a systemic order in the 21st century contribute to social upheaval and profound transformations. It was revealed that the sustainability of social development in the focus of public opinion is directly related to effective public administration and regulation, the effectiveness of national security strategies and strategies for the socio-economic and socio-demographic development of countries and regions

    Comparative evaluation of organoleptic properties of toothpaste containing NovaMin by young people who have had COVID-19

    The article presents an assessment of the organoleptic properties of the toothpaste with regenerating and desensetic effects of Sensodyne "Recovery and Protection" containing NovaMin (GlaxoSmithKline Helsker, UK) by young people who have suffered from COVID-19. The analysis of the questionnaire revealed changes in taste and smell sensations, which are the delayed effects of Sars-CoV-2 and recorded among student volunteers during the use of Sensodyne "Restore and Protect" toothpaste containing NovaMin. The paper explains the features of organoleptic perception of toothpaste in COVID-19 over-exposed students, during and after the disease. These studies and comparative evaluation can be applied in the future to study coronavirus infection on the body condition of young peopleВ статье представлена оценка органолептических свойств зубной пасты с восстанавливающим и десенсетивным эффектами – Sensodyne «Восстановление и Защита» с NovaMin, (ГлаксоСмитКляйн Хелскер, Великобритания) молодыми людьми, перенесшими COVID-19.     Mieap, a p53-Inducible Protein, Controls Mitochondrial Quality by Repairing or Eliminating Unhealthy Mitochondria

    Maintenance of healthy mitochondria prevents aging, cancer, and a variety of degenerative diseases that are due to the result of defective mitochondrial quality control (MQC). Recently, we discovered a novel mechanism for MQC, in which Mieap induces intramitochondrial lysosome-like organella that plays a critical role in the elimination of oxidized mitochondrial proteins (designated MALM for Mieap-induced accumulation of lysosome-like organelles within mitochondria). However, a large part of the mechanisms for MQC remains unknown. Here, we report additional mechanisms for Mieap-regulated MQC. Reactive oxygen species (ROS) scavengers completely inhibited MALM. A mitochondrial outer membrane protein NIX interacted with Mieap in a ROS-dependent manner via the BH3 domain of NIX and the coiled-coil domain of Mieap. Deficiency of NIX also completely impaired MALM. When MALM was inhibited, Mieap induced vacuole-like structures (designated as MIV for Mieap-induced vacuole), which engulfed and degraded the unhealthy mitochondria by accumulating lysosomes. The inactivation of p53 severely impaired both MALM and MIV generation, leading to accumulation of unhealthy mitochondria. These results suggest that (1) mitochondrial ROS and NIX are essential factors for MALM, (2) MIV is a novel mechanism for lysosomal degradation of mitochondria, and (3) the p53-Mieap pathway plays a pivotal role in MQC by repairing or eliminating unhealthy mitochondria via MALM or MIV generation, respectively

    Sestrin2 Modulates AMPK Subunit Expression and Its Response to Ionizing Radiation in Breast Cancer Cells

    Background: The sestrin family of stress-responsive genes (SESN1-3) are suggested to be involved in regulation of metabolism and aging through modulation of the AMPK-mTOR pathway. AMP-activated protein kinase (AMPK) is an effector of the tumour suppressor LKB1, which regulates energy homeostasis, cell polarity, and the cell cycle. SESN1/2 can interact directly with AMPK in response to stress to maintain genomic integrity and suppress tumorigenesis. Ionizing radiation (IR), a widely used cancer therapy, is known to increase sestrin expression, and acutely activate AMPK. However, the regulation of AMPK expression by sestrins in response to IR has not been studied in depth. Methods and Findings: Through immunoprecipitation we observed that SESN2 directly interacted with the AMPKa1b1c1 trimer and its upstream regulator LKB1 in MCF7 breast cancer cells. SESN2 overexpression was achieved using a Flag-tagged SESN2 expression vector or a stably-integrated tetracycline-inducible system, which also increased AMPKa1 and AMPKb1 subunit phosphorylation, and co-localized with phosphorylated AMPKa-Thr127 in the cytoplasm. Furthermore, enhanced SESN2 expression increased protein levels of LKB1 and AMPKa1b1c1, as well as mRNA levels of LKB1, AMPKa1, and AMPKb1. Treatment of MCF7 cells with IR elevated AMPK expression and activity, but this effect was attenuated in the presence of SESN2 siRNA. In addition, elevated SESN2 inhibited IR-induced mTOR signalling and sensitized MCF7 cells to IR through an AMPK-dependent mechanism

    The sixth international RASopathies symposium: Precision medicine—From promise to practice

    The RASopathies are a group of genetic disorders that result from germline pathogenic variants affecting RAS‐mitogen activated protein kinase (MAPK) pathway genes. RASopathies share RAS/MAPK pathway dysregulation and share phenotypic manifestations affecting numerous organ systems, causing lifelong and at times life‐limiting medical complications. RASopathies may benefit from precision medicine approaches. For this reason, the Sixth International RASopathies Symposium focused on exploring precision medicine. This meeting brought together basic science researchers, clinicians, clinician scientists, patient advocates, and representatives from pharmaceutical companies and the National Institutes of Health. Novel RASopathy genes, variants, and animal models were discussed in the context of medication trials and drug development. Attempts to define and measure meaningful endpoints for treatment trials were discussed, as was drug availability to patients after trial completion

    Differential Regulation of Cutaneous Oncoprotein HPVE6 by wtp53, Mutant p53R248W and ΔNp63α is HPV Type Dependent

    UV exposure and p53 mutations are major factors in non-melanoma skin cancer, whereas a role for HPV infections has not been defined. Previous data demonstrated the wtp53-mediated degradation of cutaneous HPV20E6 by caspase-3. ΔNp63α and hot-spot mutant p53R248W conveyed a protective effect on HPV20E6 under these conditions. We demonstrate a differential regulation by wtp53 of the E6 genes of cutaneous types HPV4, HPV5, HPV7, HPV27, HPV38, HPV48, HPV60 and HPV77. Caspase- or proteasome-mediated down-regulation was HPV type dependent. Mutant p53R248W up-regulated expression of all these E6 proteins as did ΔNp63α except for HPV38E6 which was down-regulated by the latter. None of these cellular proteins affected HPV41E6 expression. Ectopic expression of both mutp53R248W and ΔNp63α in the normal NIKS keratinocyte cell line harbouring endogenous p53 and p63however led to a down-regulation of HPV20E6. We demonstrate that HPV20E6 expression in these cells is modulated by additional, yet unidentified, cellular protein(s), which are not necessarily involved in apoptosis or autophagy. We further demonstrate proliferation of HPV20E6-expressing keratinocytes. Levels of proteins involved in cell cycle control, cyclin-D1, cdk6 and p16INK4a, phosphorylated pRB, as well as c-Jun and p-c-Jun, were all increased in these cells. HPV20E6 did not compete for the interaction between p16INK4a with cyclin-D1 or cdk6. Phosphorylation of pRB in the HPV20E6 expressing cells seems to be sufficient to override the cytokenetic block induced by the p16INK4a/pRB pathway. The present study demonstrates the diverse influence of p53 family members on individual cutaneous HPVE6 proteins. HPV20E6 expression also resulted in varying protein levels of factors involved in proliferation and differentiation
