    A prospective study of vaginal trichomoniasis and HIV-1 shedding in women on antiretroviral therapy

    <p>Abstract</p> <p>Background</p> <p><it>Trichomonas vaginalis </it>has been associated with increased vaginal HIV-1 RNA shedding in antiretroviral therapy (ART)-naïve women. The effect of trichomoniasis on vaginal HIV-1 shedding in ART-treated women has not been characterized. We tested the hypothesis that <it>T. vaginalis </it>infection would increase vaginal HIV-1 RNA shedding in women on ART, and that successful treatment would reduce vaginal HIV-1 RNA levels.</p> <p>Methods</p> <p>We conducted a prospective cohort study including monthly follow-up of 147 women receiving ART in Mombasa, Kenya. Those with <it>T. vaginalis </it>infection, defined by the presence of motile trichomonads on vaginal saline wet mount, received treatment with single dose metronidazole (2 g). Test of cure was performed at the next monthly visit. Using the pre-infection visit as the reference category, we compared detection of vaginal HIV-1 RNA before versus during and after infection using generalized estimating equations. A cut-off of 100 HIV-1 RNA copies/swab was used as the lower limit for linear quantitation.</p> <p>Results</p> <p>Among 31 women treated for trichomoniasis, the concentration of vaginal HIV-1 RNA was above the limit for quantitation before, during, and after <it>T. vaginalis </it>infection in 4 (13% [95% CI 4% - 30%]), 4 (13% [95% CI 4% - 30%]), and 5 (16% [95% confidence interval {CI} 5% - 34%]) women respectively. After adjusting for potential confounding factors, we could detect no difference in the likelihood of detecting vaginal HIV-1 RNA before versus during infection (odds ratio [OR] 1.41, 95% CI 0.23 - 8.79, p = 0.7). In addition, detection of HIV-1 RNA was similar before infection versus after successful treatment (OR 0.68, 95% CI (0.13 - 3.45), p = 0.6).</p> <p>Conclusion</p> <p>Detection of vaginal HIV-1 RNA during ART was uncommon at visits before, during and after <it>T. vaginalis </it>infection.</p

    Predicting environmental chemical factors associated with disease-related gene expression data

    <p>Abstract</p> <p>Background</p> <p>Many common diseases arise from an interaction between environmental and genetic factors. Our knowledge regarding environment and gene interactions is growing, but frameworks to build an association between gene-environment interactions and disease using preexisting, publicly available data has been lacking. Integrating freely-available environment-gene interaction and disease phenotype data would allow hypothesis generation for potential environmental associations to disease.</p> <p>Methods</p> <p>We integrated publicly available disease-specific gene expression microarray data and curated chemical-gene interaction data to systematically predict environmental chemicals associated with disease. We derived chemical-gene signatures for 1,338 chemical/environmental chemicals from the Comparative Toxicogenomics Database (CTD). We associated these chemical-gene signatures with differentially expressed genes from datasets found in the Gene Expression Omnibus (GEO) through an enrichment test.</p> <p>Results</p> <p>We were able to verify our analytic method by accurately identifying chemicals applied to samples and cell lines. Furthermore, we were able to predict known and novel environmental associations with prostate, lung, and breast cancers, such as estradiol and bisphenol A.</p> <p>Conclusions</p> <p>We have developed a scalable and statistical method to identify possible environmental associations with disease using publicly available data and have validated some of the associations in the literature.</p

    The Role of Ethnic Directors in Corporate Social Responsibility: Does Culture matter? The Cultural Trait Theory Perspectives

    This paper investigates the effect of cultural differences between ethnic directors on corporate social responsibility (CSR) of Public Liability Companies (PLCs) in Nigeria. Using the cultural trait theory, the study focuses on how the ethnic directors are influenced when making decisions concerning CSR. Adopting multiple regression analysis of data, the study investigates the three major ethnic groups (Yoruba, Igbo and Hausa) and finds cultural differences between the ethnic directors affect the adoption of CSR. Empirical results indicate that ethnic directors (Yoruba, Igbo and Hausa) were positively and significantly related to CSR. The paper contributes to the corporate governance and CSR debate concerning how ethnic directors’ decisions impact on CSR activities, particularly on the directors who are individualistic and collectivists towards CSR

    Assessing the Impacts of Land Use and Climate Changes on River Discharge towards Lake Victoria

    The Lake Victoria basin’s expanding population is heavily reliant on rainfall and river flow to meet their water needs, making them extremely vulnerable to changes in climate and land use. To develop adaptation and mitigation strategies to climate changes it is urgently necessary to evaluate the impacts of climate change on the quantity of water in the rivers that drain into Lake Victoria. In this study, the semi-distributed hydrological SWAT model was used to evaluate the impact of current land use and climate changes for the period of 1990–2019 and assess the probable future impacts of climate changes in the near future (2030–2060) on the Simiyu river discharge draining into Lake Victoria, Northern Tanzania. The General Circulation Model under RCPs 4.5, 6.0 and 8.5 predicted an increase in the annual average temperature of 1.4 °C in 2030 to 2 °C in 2060 and an average of 7.8% reduction in rainfall in the catchment. The simulated river discharge from the hydrological model under RCPs 4.5, 6.0 and 8.5 revealed a decreasing trend in annual average discharge by 1.6 m3/s from 5.66 m3/s in 2019 to 4.0 m3/s in 2060. The increase in evapotranspiration caused by the temperature increase is primarily responsible for the decrease in river discharge. The model also forecasts an increase in extreme discharge events, from a range between 32.1 and 232.8 m3/s in 1990–2019 to a range between 10.9 and 451.3 m3/s in the 2030–2060 period. The present combined impacts of climate and land use changes showed higher effects on peak discharge at different return periods (Q5 to Q100) with values of 213.7 m3/s (Q5), 310.2 m3/s (Q25) and 400.4 m3/s (Q100) compared to the contributions of climate-change-only scenario with peak discharges of 212.1 m3/s (Q5), 300.2 m3/s (Q25) and 390.2 m3/s (Q100), and land use change only with peak discharges of 295.5 m3/s (Q5), 207.1 m3/s Q25) and 367.3 m3/s (Q100). However, the contribution ratio of climate change was larger than for land use change. The SWAT model proved to be a useful tool for forecasting river discharge in complex semi-arid catchments draining towards Lake Victoria. These findings highlight the need for catchment-wide water management plans in the Lake Victoria Basin

    Transformation textures in Ni rich NiTi shape memory alloy

    In-situ deformation of Ni rich NiTi shape memory alloy is characterized using synchrotron diffraction. Thechanges in the texture of austenite and martensite phases as a function of applied stress were systematicallyinvestigated. Development of large elastic stresses with a concomitant plastic deformation have been found torestrict the stress induced martensitic transformation. A strong lattice correspondence between the austeniteand martensite phases along with a strong variant selection is reported. Further, based on the inclination ofthe austenite orientations to the applied loading axis, stress induced martensitic transformation or dislocationslip is activated at higher stress. This leads to the strengthening of texture components of the austenite phaseand a strong variant selection in the martensite phase

    A Fluid Pulse on the Hikurangi Subduction Margin: Evidence From a Heat Flux Transect Across the Upper Limit of Gas Hydrate Stability

    A transect of seafloor heat probe measurements on the Hikurangi Margin shows a significant increase of thermal gradients upslope of the updip limit of gas hydrate stability at the seafloor. We interpret these anomalously high thermal gradients as evidence for a fluid pulse leading to advective heat flux, while endothermic cooling from gas hydrate dissociation depresses temperatures in the hydrate stability field. Previous studies predict a seamount on the subducting Pacific Plate to cause significant overpressure beneath our study area, which may be the source of the fluid pulse. Double-bottom simulating reflections are present in our study area and likely caused by uplift based on gas hydrate phase boundary considerations, although we cannot exclude a thermogenic origin. We suggest that uplift may be associated with the leading edge of the subducting seamount. Our results provide further evidence for the transient nature of fluid expulsion in subduction zones

    Effectiveness of low-cost air quality monitors for identifying volcanic SO2 and PM downwind from Masaya volcano, Nicaragua

    Gas and particulate matter (PM) emissions from Masaya volcano, Nicaragua, cause substantial regional volcanic air pollution (VAP). We evaluate the suitability of low-cost SO2 and PM sensors for a continuous air-quality network. The network was deployed for six months in five populated areas (4-16 km from crater). The SO2 sensors failed and recorded erroneous values on multiple occasions, likely due to corrosion, requiring significant maintenance commitment. The PM sensors were found to be robust but data required correction for humidity. SO2 measurements could not be used as stand-alone tools to detect occurrence of VAP episodes (VAPE), but SO2/PM correlation reliably achieved this at near-field stations, as confirmed by meteorological forecasts and satellite imagery. Above-background PM concentrations reliably identified VAPE at both near-field and far-field stations. We suggest that a continuous network can be built from a combination of low-cost PM and SO2 sensors with a greater number of PM-only sensors

    BACKGROUND: The interleukin-2-mediated immune response is critical for host defense against infectious pathogens. Cytokine-inducible SRC homology 2 (SH2) domain protein (CISH), a suppressor of cytokine signaling, controls interleukin-2 signaling. METHODS: Using a case-control design, we tested for an association between CISH polymorphisms and susceptibility to major infectious diseases (bacteremia, tuberculosis, and severe malaria) in blood samples from 8402 persons in Gambia, Hong Kong, Kenya, Malawi, and Vietnam. We had previously tested 20 other immune-related genes in one or more of these sample collections. RESULTS: We observed associations between variant alleles of multiple CISH polymorphisms and increased susceptibility to each infectious disease in each of the study populations. When all five single-nucleotide polymorphisms (SNPs) (at positions -639, -292, -163, +1320, and +3415 [all relative to CISH]) within the CISH-associated locus were considered together in a multiple-SNP score, we found an association between CISH genetic variants and susceptibility to bacteremia, malaria, and tuberculosis (P=3.8x10(-11) for all comparisons), with -292 accounting for most of the association signal (P=4.58x10(-7)). Peripheral-blood mononuclear cells obtained from adult subjects carrying the -292 variant, as compared with wild-type cells, showed a muted response to the stimulation of interleukin-2 production--that is, 25 to 40% less CISH expression. CONCLUSIONS: Variants of CISH are associated with susceptibility to diseases caused by diverse infectious pathogens, suggesting that negative regulators of cytokine signaling have a role in immunity against various infectious diseases. The overall risk of one of these infectious diseases was increased by at least 18% among persons carrying the variant CISH alleles
