    Identification of a novel ß-adrenergic octopamine receptor-like gene (ßAOR-like) and increased ATP-binding cassette B10 (ABCB10) expression in a Rhipicephalus microplus cell line derived from acaricide-resistant ticks

    Background: The cattle tick Rhipicephalus (Boophilus) microplus is an economically important parasite of livestock. Effective control of ticks using acaricides is threatened by the emergence of resistance to many existing compounds. Several continuous R. microplus cell lines have been established and provide an under-utilised resource for studies into acaricide targets and potential genetic mutations associated with resistance. As a first step to genetic studies using these resources, this study aimed to determine the presence or absence of two genes and their transcripts that have been linked with acaricide function in cattle ticks: β-adrenergic octopamine receptor (βAOR, associated with amitraz resistance) and ATP-binding cassette B10 (ABCB10, associated with macrocyclic lactone resistance) in six R. microplus cell lines, five other Rhipicephalus spp. cell lines and three cell lines representing other tick genera (Amblyomma variegatum, Ixodes ricinus and Hyalomma anatolicum). Methods: End-point polymerase chain reaction (PCR) was used for detection of the βAOR gene and transcripts in DNA and RNA extracted from the tick cell lines, followed by capillary sequencing of amplicons. Quantitative real-time PCR (qPCR) was performed to determine the levels of expression of ABCB10. Results: βAOR gene expression was detected in all Rhipicephalus spp. cell lines. We observed a second amplicon of approximately 220 bp for the βAOR gene in the R. microplus cell line BME/CTVM6, derived from acaricide-resistant ticks. Sequencing of this transcript variant identified a 36 bp insertion in the βAOR gene, leading to a 12-amino acid insertion (LLKTLALVTIIS) in the first transmembrane domain of the protein. In addition, nine synonymous SNPs were also discovered in R. appendiculatus, R. evertsi and R. sanguineus cell lines. Some of these SNPs appear to be unique to each species, providing potential tools for differentiating the tick species. The BME/CTVM6 cell line had significantly higher ABCB10 (P = 0.002) expression than the other R. micropluscell lines. Conclusions: The present study has identified a new βAOR gene and demonstrated a higher ABCB10 expression level in the BME/CTVM6 cell line, indicating that tick cell lines provide a useful experimental tool for acaricide resistance studies and further elucidation of tick genetics

    Road mode of transport dominates African and Cameroonian cities but unequally developed to portray sturdy spatio-temporal variation trends.This assessment of the negative development ramifications of traffic congestion along road axis of Bamendais carried out through a sample survey of road users from September to October 2014. Field observation and sampling on traffic trend variables indicate that technical factors play over infrastructural variables to trigger mobility flow bottlenecks and trends. Statistical treatment of primary and secondary data depictunequal daily/seasonal variation with distance from city centre being higher in the rainy season and school periods than dry seasons and holidays. Traffic flow and congestion is higher along the NE than NW road axis with significant socio-economic consequences road users such as weakening of income, lateness, emotional and psychological stress, accidents, material damageas well aspollution. This study opts for a holistic approach to mitigate traffic problems using structural and non-structuralmeasures

    Accurate segregation of genetic material into two daughter cells is essential for organism reproduction, development, and survival. The cell assembles a macromolecular structure called the mitotic spindle, which is composed of dynamic microtubules (MTs) and many associated proteins that assemble the spindle and drive the segregation of the chromosomes. Members of the kinesin superfamily of MT associated proteins use the energy of ATP hydrolysis to help organize the spindle, to transport cargo within the spindle, and to regulate spindle MT dynamics. The Kinesin-8 and Kinesin-13 families are involved in controlling mitotic spindle morphology, spindle positioning, and chromosome movement. While both kinesin families are MT destabilizing enzymes, it is unclear whether their mechanisms of MT destabilization are mechanistically similar or how they act to destabilize MTs. Recently, three groups identified an additional MT binding domain within the tail of Kinesin-8s that is essential for their roles in regulating MT dynamics and chromosome positioning

    Space-Based Observations for Understanding Changes in the Arctic-Boreal Zone

    Observations taken over the last few decades indicate that dramatic changes are occurring in the ArcticBoreal Zone (ABZ), which are having significant impacts on ABZ inhabitants, infrastructure, flora and fauna, and economies. While suitable for detecting overall change, the current capability is inadequate for systematic monitoring and for improving processbased and largescale understanding of the integrated components of the ABZ, which includes the cryosphere, biosphere, hydrosphere, and atmosphere. Such knowledge will lead to improvements in Earth system models, enabling more accurate prediction of future changes and development of informed adaptation and mitigation strategies. In Duncan et al. (2020), we review the strengths and limitations of current spacebased observational capabilities for several important ABZ components and make recommendations for improving upon these current capabilities. We recommend an interdisciplinary and stepwise approach to develop a comprehensive ABZ Observing Network (ABZON), beginning with an initial focus on observing networks designed to gain processbased understanding for individual ABZ components and systems that can then serve as the building blocks for a comprehensive ABZON

    Nursing students doing gender: Implications for higher education and the nursing profession

    The average age of women nursing students in Australia is rising. With this comes the likelihood that more now begin university with family responsibilities, and with their lives structured by the roles of mother and partner. Women with more traditionally gendered ideas of these roles, such as nurturing others and self-sacrifice, are known to be attracted to nursing as a profession; once at university, however, these students can be vulnerable to gender role stress from the competing demands of study. A qualitative research design, guided by Gadamer\u27s hermeneutic philosophy, explored the gendered behaviours and experiences of 22 women nursing students, all of whom had children and began university in a heterosexual intimate relationship. The findings reveal traditional ideas of gender were almost universal among participants, and these ideas had a significant influence on the nursing degree experience. Participants commonly prioritised family over the university and practiced maternal gatekeeping (prevention of male partner involvement in domestic work). These traditionally gendered behaviours, coupled with experiences of gender role stress, had a detrimental impact on participants\u27 capacity to study and their personal wellbeing. The importance of these findings to the burgeoning nursing workforce shortage nursing is considered in terms of student retention and the supply of graduates into the profession. The implications to the nursing profession are also explored against the evidence that nursing students with traditional gender beliefs are less likely to develop as autonomous, critical thinking nurses compared to their gender-egalitarian peers. The introduction of gender theory via critical pedagogy in the undergraduate nursing degree curriculum is recommended to enlighten and empower women nursing students and promote the competence, agility, and sustainability of the nursing profession
