Az iskolai lemorzsolódás elleni küzdelem és a hátrányos helyzetű fiatalok sikeres társadalmi és munkaerőpiaci integrációja napjaink egyik legnagyobb társadalmi kihívása. Kiemelt kutatási programok keresik a jelenség okait, következményeit és a lehetséges kezelési módokat. Az eddig rendelkezésre álló elemzések, statisztikai adatok azonban nagyon keveset árulnak el a fiatalok élethelyzetéről, családjáról, barátairól, anyagi helyzetéről, gondolkodásáról, céljairól, minden-napjaikról. Kutatásunk során erről szerettünk volna többet megtudni az iskolából lemorzsoló-dott, vagy lemorzsolódással veszélyeztetett 15–19 éves kor közötti fiatalok körében egy második esélyt biztosító középiskolában készített interjúk során

A common tool used to determine genetic variation in different species, breeds and populations is mitochondrial DNA analysis, which can be used to estimate phylogenetic relationships among animal taxa and for molecular phylogenetic evolution analysis. With the development of sequencing technology, more and more mitochondrial sequences have been made available in public databases, including whole mitochondrial DNA sequences. These data have been used for phylogenetic analysis of animal species, and for studies of evolutionary processes

Identifying Suitable Reference Gene Candidates for Quantification of DNA Damage-Induced Cellular Responses in Human U2OS Cell Culture System
DNA repair pathways trigger robust downstream responses, making it challenging to select suitable reference genes for comparative studies. In this study, our goal was to identify the most suitable housekeeping genes to perform comparable molecular analyses for DNA damage-related studies. Choosing the most applicable reference genes is important in any kind of target gene expression-related quantitative study, since using the housekeeping genes improperly may result in false data interpretation and inaccurate conclusions. We evaluated the expressional changes of eight well-known housekeeping genes (i.e., 18S rRNA, B2M, eEF1α1, GAPDH, GUSB, HPRT1, PPIA, and TBP) following treatment with the DNA-damaging agents that are most frequently used: ultraviolet B (UVB) non-ionizing irradiation, neocarzinostatin (NCS), and actinomycin D (ActD). To reveal the significant changes in the expression of each gene and to determine which appear to be the most acceptable ones for normalization of real-time quantitative polymerase chain reaction (RT-qPCR) data, comparative and statistical algorithms (such as absolute quantification, Wilcoxon Rank Sum Test, and independent samples T-test) were conducted. Our findings clearly demonstrate that the genes commonly employed as reference candidates exhibit substantial expression variability, and therefore, careful consideration must be taken when designing the experimental setup for an accurate and reproducible normalization of RT-qPCR data. We used the U2OS cell line since it is generally accepted and used in the field of DNA repair to study DNA damage-induced cellular responses. Based on our current data in U2OS cells, we suggest using 18S rRNA, eEF1α1, GAPDH, GUSB, and HPRT1 genes for UVB-induced DNA damage-related studies. B2M, HPRT1, and TBP genes are recommended for NCS treatment, while 18S rRNA, B2M, and PPIA genes can be used as suitable internal controls in RT-qPCR experiments for ActD treatment. In summary, this is the first systematic study using a U2OS cell culture system that offers convincing evidence for housekeeping gene selection following treatment with various DNA-damaging agents. Here, we unravel an indispensable issue for performing and assessing trustworthy DNA damage-related differential gene expressional analyses, and we create a “zero set” of potential reference gene candidates
A kiégés megjelenése a szociális szakmában és elkerülésének lehetőségei
Az emberekkel foglalkozó szakmák, így a szociális munkás pálya is magában hordozza az úgynevezett kiégési szindróma veszélyét, azt az állapotot, amikor az egyébként motivált, érzelem- gazdag személyiség telítődik azokkal a problémákkal, amelyek nap, mint nap jelentkeznek tevékenységében. A szociális munkásoknak csakúgy, mint minden embernek meg kell küzdeniük saját belső feszültségeikkel, személyes életük során felmerülő problémákkal, különböző súlyú és tartalmú konfliktusokkal.
A munkahelyen megélt feszültség, stressz hatással van az életünk egyéb területeire is. Előfordulhat, hogy a munkával kapcsolatos stresszel nem tudunk mindig hatékonyan megküzdeni, ami problémákat hagy megoldatlanul saját magunkban, kiábrándultság érzéséhez és kiégéshez vezethet. A szakirodalomban egyre többet találkozhatunk a kiégés jelenségével. Szinte minden „emberközpontú” foglalkozásban megjelenhet, de egyáltalán nem mindegy, hogy emberekkel vagy tárgyakkal dolgozik-e az illető.
A szociális munka gyakorlása során rendkívül nagy jelentősége van annak, hogy miként boldogul az illető önnön problémáival, hogyan alakul mentálhigiénés állapota, mennyire képes megőrizni belső egyensúlyát, fékezni, kezelni indulatait az egyes helyzetekben.
Az egyetemen töltött évek alatt, több szociális intézményben jártam a gyakorlataim során és találkoztam sok szakmabelivel. Órákon is többször találkoztunk a kiégés fogalmával melyet igen fenyegetőnek találtam a magamra, csoporttársaimra, és a már szociális munkásként dolgozók számára. Mindig azt hallottuk, hogy a szociális munkás a személyiségével dolgozik, tehát igencsak figyelnünk kell magunkra, ne vonódjunk bele mások életébe érzelmileg túlságosan, de ne is tárgyilagosan kezeljük az eseteket. Láttam példát arra, milyen kihívásokkal kerülnek szembe munkájuk során a különböző élethelyzetben élő embereket segítő szakemberek. Nagyon nehéz még pozitív hozzáállással is mindig megtalálni a szépet és a jót ebben a szakmában, mivel sokszor apró változásokat tudunk elérni hosszú munka után is, amire gyakran nem érkezik köszönet. Egyre több a kliens, egyre több az elesett, reménytelennek tűnő élethelyzetű család.
Elgondolkodtató, hogy az a szakember, akinek kevés sikerélménye van, mindennapi munkájában vagy érzelmi megterhelésnek van kitéve, hogyan tud folyamatosan megújulni. Ezt a problémát szeretném körüljárni, és a bevezető után, interjúkon keresztül bizonyítani, hogy ez a probléma fennáll, illetve azt is, hogy mennyire komoly ez a gond. Végül szeretnék különböző megoldási javaslatokat kínálni minden szociális munkás és szociális területen dolgozó számára.BSc/BASzociális munk
A korai iskolaelhagyás trendjei ózdi és környékbeli általános iskolákban
Szakdolgozatomban a korai iskolaelhagyás témájával foglalkozom. Hogyan
alakult az elmúlt években a korai iskolaelhagyás, Ózdon és környékén, milyen
lehetőségekkel élhetnek a veszélyeztetett helyzetben lévő tanulók. Különösen azért,
mert Borsod-Abaúj-Zemplén megyében sok veszélyeztető tényező közül itt szinte
minden jelen van: kiemelkedően magas a munkanélküliség és a roma lakosság
aránya, valamint alacsony az iskolázottság szintje is.
Dolgozatom első felében szeretném tisztázni az alapvető fogalmakat a témával
kapcsolatosan, bemutatok néhány Magyarországon végzett kutatási eredményt,
ismertem az Európai Unió milyen szakpolitikai ajánlásokat tesz a jövőre nézve.
Továbbá, hogy Magyarországon a területi különbségek milyen oktatásbeli
hátrányokat idéznek elő, Európában és a környező országokon belül hogyan
alakulnak a tanulmányi eredmények a PISA felmérés alapján, és ennek milyen okok
állnak a hátterében. Valamint milyen okok és események vezetnek a
lemorzsolódáshoz és korai iskolaelhagyáshoz.
Interjús kutatásban ózdi és Ózd környéki iskolák igazgatóival, tanáraival,
illetve gyermekvédelmi felelőseivel készített interjúk során keresem kérdéseimre a
választ. Hogyan alakult 5 év távlatában a korai iskolaelhagyás, milyen személyi vagy
intézményi okok állnak ennek a hátterében, a helyi oktatók és szakemberek szerint,
milyen új kihívások jelentkeznek. Ugyan még általános iskola korai elhagyása esetén
is vannak visszavezető programok, de kevesen beszélnek azokról, akik végzettség
megszerzése nélkül is boldogulni szeretnének.MSc/MAszociálpolitikaB
Mining the red deer genome (CerEla1.0) to develop X-and Y-chromosome-linked STR markers.
Microsatellites are widely applied in population and forensic genetics, wildlife studies and parentage testing in animal breeding, among others, and recently, high-throughput sequencing technologies have greatly facilitated the identification of microsatellite markers. In this study the genomic data of Cervus elaphus (CerEla1.0) was exploited, in order to identify microsatellite loci along the red deer genome and for designing the cognate primers. The bioinformatics pipeline identified 982,433 microsatellite motifs genome-wide, assorted along the chromosomes, from which 45,711 loci mapped to the X- and 1096 to the Y-chromosome. Primers were successfully designed for 170,873 loci, and validated with an independently developed autosomal tetranucleotide STR set. Ten X- and five Y-chromosome-linked microsatellites were selected and tested by two multiplex PCR setups on genomic DNA samples of 123 red deer stags. The average number of alleles per locus was 3.3, and the average gene diversity value of the markers was 0.270. The overall observed and expected heterozygosities were 0.755 and 0.832, respectively. Polymorphic Information Content (PIC) ranged between 0.469 and 0.909 per locus with a mean value of 0.813. Using the X- and Y-chromosome linked markers 19 different Y-chromosome and 72 X-chromosome lines were identified. Both the X- and the Y-haplotypes split to two distinct clades each. The Y-chromosome clades correlated strongly with the geographic origin of the haplotypes of the samples. Segregation and admixture of subpopulations were demonstrated by the use of the combination of nine autosomal and 16 sex chromosomal STRs concerning southwestern and northeastern Hungary. In conclusion, the approach demonstrated here is a very efficient method for developing microsatellite markers for species with available genomic sequence data, as well as for their use in individual identifications and in population genetics studies
Novel insights into the athlete’s heart: is myocardial work the new champion of systolic function?
AIMS: We sought to investigate the correlation between speckle-tracking echocardiography (STE)-derived myocardial work (MW) and invasively measured contractility in a rat model of athlete's heart. We also assessed MW in elite athletes and explored its association with cardiopulmonary exercise test (CPET)-derived aerobic capacity. METHODS AND RESULTS: Sixteen rats underwent a 12-week swim training program and were compared to controls (n = 16). STE was performed to assess global longitudinal strain (GLS), which was followed by invasive pressure-volume analysis to measure contractility [slope of end-systolic pressure–volume relationship (ESPVR)]. Global MW index (GMWI) was calculated from GLS curves and left ventricular (LV) pressure recordings. In the human investigations, 20 elite swimmers and 20 healthy sedentary controls were enrolled. GMWI was calculated through the simultaneous evaluation of GLS and non-invasively approximated LV pressure curves at rest. All subjects underwent CPET to determine peak oxygen uptake (VO(2)/kg). Exercised rats exhibited higher values of GLS, GMWI, and ESPVR than controls (−20.9 ± 1.7 vs. −17.6 ± 1.9%, 2745 ± 280 vs. 2119 ± 272 mmHg·%, 3.72 ± 0.72 vs. 2.61 ± 0.40 mmHg/μL, all P(Exercise) < 0.001). GMWI correlated robustly with ESPVR (r = 0.764, P < 0.001). In humans, regular exercise training was associated with decreased GLS (−17.6 ± 1.5 vs. −18.8 ± 0.9%, P(Exercise) = 0.002) but increased values of GMWI at rest (1899 ± 136 vs. 1755 ± 234 mmHg·%, P(Exercise) = 0.025). GMWI exhibited a positive correlation with VO(2)/kg (r = 0.527, P < 0.001). CONCLUSIONS: GMWI precisely reflected LV contractility in a rat model of exercise-induced LV hypertrophy and captured the supernormal systolic performance in human athletes even at rest. Our findings endorse the utilization of MW analysis in the evaluation of the athlete’s heart

The red deer Cervus elaphus genome CerEla1.0: sequencing, annotating, genes, and chromosomes
We present here the de novo genome assembly CerEla1.0 for the
red deer, Cervus elaphus, an emblematic member of the natural
megafauna of the Northern Hemisphere. Humans spread the species
in the South. Today, the red deer is also a farm-bred animal
and is becoming a model animal in biomedical and population
studies. Stag DNA was sequenced at 74× coverage by Illumina
technology. The ALLPATHS-LG assembly of the reads resulted in
34.7 × 103 scaffolds, 26.1 × 103 of which were utilized in
Cer.Ela1.0. The assembly spans 3.4 Gbp. For building the red
deer pseudochromosomes, a pre-established genetic map was used
for main anchor points. A nearly complete co-linearity was
found between the mapmarker sequences of the deer genetic map
and the order and orientation of the orthologous sequences in
the syntenic bovine regions. Syntenies were also conserved at
the in-scaffold level. The cM distances corresponded to 1.34
Mbp uniformly along the deer genome. Chromosomal rearrangements
between deer and cattle were demonstrated. 2.8 × 106 SNPs, 365
× 103 indels and 19368 protein-coding genes were identified in
CerEla1.0, along with positions for centromerons. CerEla1.0
demonstrates the utilization of dual references, i.e., when a
target genome (here C. elaphus) already has a pre-established
genetic map, and is combined with the well-established whole
genome sequence of a closely related species (here Bos taurus).
