    Chastity is the state in which the love life of Tamils is centred and manifested.  Chastity is to fully attain the state of mind of genuine love (Kalaviyal).  Chastity is conducting the wedding ceremony (Karanam) with the knowledge of the whole world. Marriages are completed by tying the Mangal Sutra. Even after the lover's misconduct, in the state of women being still considered unworthy, a procedure called ‘Karanam’ is conducted publicly. Chastity sets in to illustrate the way the Man and Women conduct their lives and morality that is well established. Tolkappiyam completely embodies the lifestyles of the people and the personal ideas expressed in the Sangam literature in alliance with the cultural traditions of the people. Tolkaapiyar describes chastity in the form of the ritual itself. It is also called chastity when the parents give away their daughter to the groom; the parents of the groom receive her by a ritual confirmation ceremony. The woman remaining chaste when the man is away from home is said to be happening in the forest (Mullai Nilam). In the life of a lover, a ritual that is being chaste as ‘One man; one woman and one woman; one man’ and living together after marrying publicly is accepted universally

    Chain flexibility and configurational dimensions of left handed Z- DNA and right handed B-DNA helices

    The configurational behaviour of flexible helices of right handed B- and left handed Z-types have been analysed using statistical mechanical procedures. The configuration-dependent parameter, most importantly, the persistence length has been computed, using the heminucleotide scheme of treating polynucleotide chains under the approximation that perturbations in the backbone torsions produce sufficient flexibility in these helices. The values of persistence lengths obtained for Z-helices are very much higher than that of B-helices indicating that former is less flexible compared to the latter. These are in accordance with the results obtained recently on B- and Z-forms of poly(dG-dC) · (dG-dC) using light scattering studies. Also the persistence lengths of BII-DNA helices characterised by a skew 3'-hemiucleotide (Δ ≃ 270°), and also when they coexist with B-DNA have been computed and the values lie within the range of experimentally reported values on B-helices. It is argued that the decrease in the persistence length values of B-DNA at higher salt concentration is due to additional small fluctuations in sugar residue torsions induced due to neutralisation of electrostatic repulsions between adjacent phosphates of the nucleotide. Noteworthy is that these are correlated to winding angle variations and the consequent bending of the helix

    A Comparative Study on the Neuro-Protective Effects of DHA, Vitamin E and Memantine in the Neonatal Rat Model of LPS Induced Cerebral Palsy

    In conclusion, the derived results demonstrate that DHA, TOCOPHEROL and MEMANTINE could protect neuronal cells, and thereby preventing brain damage in adverse conditions. The neuroprotective pathway appears to implicate NMDA receptors and inflammatory modulation, leading to a promotion of oligodendrocyte maturation. The present study also delineates the cellular mechanisms of Memantine’s neuroprotective benefits. Furthermore, our data strongly suggest that DHA, TOCOPHEROL and MEMANTINE could be of great interest not only in perinatal white matter damage but also as a potential neuroprotective strategy for myelinopathy diseases observed in adults. The data provides the proof of concept that antioxidant therapy with DHA and TOCOPHEROL may antagonize the effects of free-radicals that may lead to neuronal loss. The results provide new insights into the potential of employing n-3 TG specifically DHA as a unique long chain FA aiding in neonatal H/I brain injury. A number of pathways are likely involved in n-3 TG neuroprotection. For example, chronic administration of DHA resulted in increases of DHA levels in brain mitochondria. My findings suggest a need for further studies to determine if acute injection of DHA could be neuroprotective after stroke injury in humans. I hypothesize that n-3 fatty acids, specifically DHA, will prove to be a “novel” and important therapy to treat various neuro-degenerative disorders and could decrease mortality and increase long-term functional recovery

    FIRST COMPLETE GENOME SEQUENCE OF OCULAR ISOLATE CARRYING BLAVIM-2 MEDIATED MULTI-DRUG-RESISTANT(MDR) PSEUDOMONAS AERUGINOSA VRFPA04 FROM INDIA AND TWO DRAFT GENOME SEQUENCE OF OCULAR ISOLATES PSEUDOMONAS AERUGINOSA VRFPA03 AND VRFPA05 HARBORING NOVEL BLA

    Objective: Ocular infections caused by Multi-Drug resistant (MDR) Pseudomonas aeruginosa are rare but increasingly identified recently. In this context, we have applied Next Generation Sequencing (NGS) based whole genome analysis on three ocular isolates of MDR P. aeruginosa to explore the drug resistance determinants and genomic level variations.Methods: Three ocular isolates namely P. aeruginosa VRFP03, VRFPA04 from two different Keratitis patients (corneal button) and VRFPA05 isolated from the intraocular specimen (Vitreous humor) collected from an endophthalmitis patient was included in this study. Phenotypically VRFPA03, VRFPA04, VRFPA05 showed resistant to a wide group of antibiotics and hence they were taken up for Ion Torrent-PGM based whole genome study.Results: Here, we report the first complete genome sequence of MDR P. aeruginosa VRFPA04 isolated from the Indian keratitis patients clinical specimen (corneal button) submitted to L & T Microbiology Research Centre, Vision Research Foundation, Sankara Nethralaya, Chennai, Tamil Nadu, India. The circular chromosome of P aeruginosa VRFPA04 was published under the NCBI accession number CP008739.2. Two Draft genome sequences of MDR P. aeruginosa VRFPA03 and VRFPA05 were published under the NCBI accession number ATNK01000000.1 and AXZJ01000000.1 respectively.Conclusion: Preliminary genomic analysis on P. aeruginosa VRFPA03, VRFPA04 and VRFPA05 revealed the presence of Metallo Beta-lactamase (MBL) genes blaDim-1, blaVim-2 andblaGes-9 genes respectively. These MBL genes were concordance with a phenotypic pattern of carbapenem resistance among VRFPA03, VRFPA04 and VRFPA05 strains, respectively. We are proud to share that this is the first of its kind to report blaDIM-1 from an Indianisolate especially from an Ocular origin.Â

    Enhanced Dwarf Mongoose Optimization Based Node Localization Scheme for Underwater Wireless Sensor Networks

    Recently, the Underwater Wireless Sensor Network (UWSN) with movable nodes has been launched because it has a lot of potential uses in aquatic science and is getting a lot of attention from researchers. Because there are so many more possible underwater uses, it is important to make sure that signals can get from one underwater point to another. Because of the long lags in signal transmission and the changing speed of sound underwater, it is hard and not possible to use the usual localization method in UWSN. Node localization (NL) in UWSN tries to figure out where new nodes are by using known nodes. The correctness of location can have a big effect on how well a UWSN works. UWSN has a lot of trouble with precise NL. When it comes to WSN, NL problems mean figuring out where unknown sensor nodes (SNs) are. This shows how important it is to have a perfect NL system. The Enhanced Dwarf Mongoose Optimization based Node Localization Scheme (EDMO-NLS) for UWSN is being worked on in this study. The scout group, the alpha group, and the babysitters are the three social groups of dwarf mongooses that were used in the suggested method. The family hunts as a unit, and the main female starts hunting. This sets the sleeping mounds, the road for hunting, and the distance that is covered. The EDMO-NLS method shown here finds nodes whose locations are unknown in UWSN. The modeling results showed that the proposed model worked better than existing methods. Based on the findings and the discussion, it is clear that the suggested method has shown the best localization success in UWSN

    DRAFT GENOME SEQUENCE OF HUMAN HERPES VIRUS-4 VRF_EBV_01, AN EPSTEIN BARR VIRUS OBTAINED FROM A PEDIATRIC POST TRANSPLANT LYMPHOPROLIFERATIVE DISORDER (PTLD) PATIENT

    Objectives: To decode the sequence of Epstein-barr virus (EBV) genome isolated from a pediatric patient with post transplant lymphoproliferative disorder (PTLD).Methods: EBV culture harvested from the blood sample of a 4-year-old patient with post liver transplant lymphoproliferative disorder prior to treatment was subjected to whole genome sequencing using Illumina platform. Generated data were subjected to various quality analysis and the filtered sequences were submitted to NCBI and published under accession number KM269735-KM269744.Results: Annotation results of VRF_ EBV01 genome using Prokka tool and manual blast search infers 48 hypothetical proteins, each 3 genes coding for Epstein-Barr nuclear antigen 3A and 3B and each one gene coding for Epstein-Barr nuclear antigens 1, 4, 4B and DNA primase UL70 protein. Single copy of genes coding for each variant of Tegument protein BSRF1, BLRF2, BGLF2 mir-BART 1,2,3,5,7,12,15,17,20 and two copies of gene coding for primary envelopment factor BFRF1 gene was also found.Conclusion: To date, only eight EBV genome sequences have been reported worldwide and there is no genome sequence reported from India. This study is the first of its kind to report on EBV genome from a postñ€“transplant lymphoproliferative disorder to the scientific community for the welfare of research against EBV diagnostic markers and drug discovery.Â

    ANTIPROLIFERATIVE EFFECTS OF PSIDIUM GUAJAVA LEAF EXTRACT ON ORAL SQUAMOUS CARCINOMA CELLS

    Objective: Cancer is the major cause of mortality affecting population irrespective of age. Oral cancer is one among the various cancers affecting major population in India. To overcome toxicity of chemotherapy and disfiguration by surgical procedures, researchers are targeting phytochemicals for their anticancer properties. This study evaluates the antiproliferative effects of Psidium guajava leaf extract against OSC cells. Methods: KB cells were purchased from NCCS, Pune. Extract from leaves of P. guajava was prepared with ethanol and evaluated with high-performance thin-layer chromatography (HPTLC). Antiproliferative effects of the extract were assessed with 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl--tetrazolium bromide (MTT) assay and flow cytometry. Results: HPTLC revealed the presence of quercetin in the extract. MTT assay showed decreasing pattern in cell viability with the increasing dose of extract. Flow cytometry revealed the seizing of cycle by the extract. Conclusion: The study results conclude the presence of antiproliferative properties in the leaf extract of P. guajava

    COMPARATIVE STUDY OF HAEMATOLOGICAL PARAMETERS IN FISHES FROM THE NATURAL HABITAT, MADURAI REGION

    The heamatological profile of two freshwater fishes Labeo rohita (Rohu) and Channa striatus (Murrel) with a relationship to the sex and the water quality under natural habitat was analyzed. The freshwater fishes were collected from the pond located in Thodaneri, Vadipatti taluk, Madurai district, Tamil Nadu, India. In heamatology studies, no significant variation was observed in Heamoglobin content and found almost the same in both the fish irrespective of the sex and also falls in the range of control value. In differential Leucocyte count (DLC) the following blood components Lymphocyte, Eosinophil, and Monocyte count were significantly higher than the control values and the values are found maximum in Rohu when compared to Murrel.RBC (Erythrocyte) count was almost the same in both the fish and the value falls between the range of control value Packed Cell Volume (PCV) count was almost the same in both the fish and the value falls between the range of control value. The values of Hb and PCV were found higher in male fish as compared to female fish. The leucocyte count is also higher in the males when compared to the females of both Rohu and Murrel. These may be also influenced by physiological activity, feeding habits, infectious agents, water quality parameters, and the environmental stress on the organism

    EVALUATION OF A METHOD FOR DETECTION OF BOTH METALLO BETALACTAMASE (MBL) AND EFFLUX PRODUCTION USING CARBONYL CYANIDE 3-CHLOROPHENYLHYDRAZONE (CCCP)±COMBINATION METHOD AMONG PSEUDOMONAS AERUGINOSA ISOLATED FROM CLINICAL SPECIMENS

    Objective: To standardize a method for the differential detection of Metallo Betalactamase (MBL) production and efflux pump mechanism using Carbonyl Cyanide 3-ChloroPhenylhydrazone (CCCP) among the clinical isolates of Pseudomonas aeruginosa recovered from ocular specimens.Methods: A total of 110 clinical isolates of Pseudomonas aeruginosa were plated onto Muller-Hinton agar (MHA) plate with CCCP and without CCCP. Carbapenems-Ethylenediaminetetraacetic acid (EDTA) combination disk method was performed on CCCP± plates for individual isolates and the results were compared.Results: Out of 110 clinical isolates of P aeruginosa studied, 11(10%) were tested positive for MBL Production by carbapenem-EDTA combination test/CCCP-test. In which, six isolates showed positive for both efflux and MBL production using CCCP+plate and 5 isolates showed positive only for MBL production.Conclusion: The current study standardized a method for detecting both MBL and efflux mechanism in a single plate consists of CCCP± plate containing antibiotic disk with and without EDTA. Implication of this method in routine diagnostics will help the clinicians to treat the patient with appropriate drug choice and dosage.Â
