    Cross-Task and Cross-Lightpath Failure Detection and Localization in Optical Networks Using Transfer Learning

    Practical deployments of Machine-Learning(ML)-based solutions for failure management in optical networks often suffer from limited data availability, due to, especially, scarcity of labelled data describing different failure scenarios. Transfer Learning (TL) is regarded as a promising direction in cases of data scarcity, thanks to its ability to transfer knowledge from a Source Domain (SD) (e.g. SD could be a digital twin or a laboratory testbed) to a Target Domain (TD) (e.g., the infield network). In this paper, we focus on cross-lightpath and cross-task application of TL for failure localization and failure detection in optical networks. We found that, depending on the number of retrained parameters in the ML model, cross-lightpath TL for failure localization provides satisfactory accuracy (higher than 90%, in some cases) with limited amounts of TD data, and is also convenient in terms of TD retraining duration with respect to cases where TL is not used. Moreover, we found that cross-task failure detection/localization reaches up to 12% or 25% improvement in TD accuracy when considering failure localization and detection as TD task, respectively

    Pliocene crustal shortening on the Tyrrhenian side of the northern Apennines: evidence from the Gavorrano antiform (southern Tuscany, Italy)

    The northern Tyrrhenian Sea and the inner northern Apennines are classically regarded as a late Miocene–Pleistocene back-arc system developed as a consequence of slab rollback along active subduction zones. We present new geological and structural data on the Gavorrano antiform, a key sector of the inner northern Apennines. Lying close to the northern Tyrrhenian Sea, it provides clear evidence of Pliocene shortening deformation and magma emplacement. The orientation of 1 (N50°E–N80°E) derived by fault slip data inversion is consistent with a general ENE–WSW shortening direction. Furthermore, this ENE–WSW-trending orientation of 1 is compatible with the compressive deformation recorded in coeval sedimentary basins. On this basis we suggest that the inner northern Apennines were affected by crustal shortening during the Pliocene. This scenario matches well geophysical data suggesting that since the Late Messinian (6–5 Ma) subduction rollback and back-arc extension strongly decreased in the northern Tyrrhenian Sea, whereas they continued as active processes in the southern Tyrrhenian Sea

    PACAP and VIP Mitigate Rotenone-Induced Inflammation in BV-2 Microglial Cells.

    Rotenone is a commercial pesticide commonly used to model Parkinson's disease (PD) due to its ability to induce dopaminergic degeneration. Studies have confirmed that rotenone causes microglial activation, which seems to contribute to the toxic effects seen in rodent models. Pituitary adenylate cyclase-activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP) are two structurally related neuropeptides that have robust neuroprotective and anti-inflammatory properties. However, their ability to regulate microglial activity in response to rotenone is not fully understood. Using rotenone as an inflammatory stimulus, we tested whether PACAP or VIP could mitigate microglial activation in BV2 microglial cells. Rotenone dose-dependently reduced cell viability and the percentage of apoptotic cells. It also increased the release of nitric oxide (NO) in culture media and the expression of microglial activation markers and pro-inflammatory markers, including CD11b, MMP-9 and IL-6, and heightened the endogenous levels of PACAP and its preferring receptor PAC1. Co-treatment with PACAP or VIP prevented rotenone-induced increase of NO, CD11b, MMP-9 and IL-6. These results indicate that both PACAP and VIP are able to prevent the pro-inflammatory effects of rotenone in BV2 cells, supporting the idea that these molecules can have therapeutic value in slowing down PD progression

    Expanded Endoscopic Approach for Anterior Skull Base Tumors: Experience of a Multidisciplinary Skull Base Team

    Abstract: The aim of this study is to describe the experience of a multidisciplinary skull base team with transnasal endoscopic surgery for anterior cranial base tumors. A retrospective chart review was conducted on patients who underwent an exclusive expanded transnasal approach to the anterior skull base in the period from December 2014 to November 2015. Data on patient demographics, tumor characteristics, surgical information, imaging, and postoperative complications were collected and analyzed. From a total of 120 patients with skull base diseases managed by the skull base team, 36 were admitted to this study. The overall complication rate in this series was 16.7%, gross total resection was achieved in 32 cases (88.9%) and postoperative CSF leakage occurred in 5 cases (13.9%).     Physical activity levels and related energy expenditure during COVID-19 quarantine among the sicilian active population: A cross-sectional online survey study

    Background: During the coronavirus disease 2019 (COVID-19) pandemic, the Italian government has adopted containment measures to control the virus's spread, including limitations to the practice of physical activity (PA). The aim of this study was to estimate the levels of PA, expressed as energy expenditure (MET-minute/week), among the physically active Sicilian population before and during the last seven days of the COVID-19 quarantine. Furthermore, the relation between this parameter and specific demographic and anthropometric variables was analyzed. Methods: 802 Sicilian physically active participants (mean age: 32.27 ± 12.81 years; BMI: 23.44 ± 3.33 kg/m2) were included in the study and grouped based on gender, age and BMI. An adapted version of the International Physical Activity Questionnaire-short form (IPAQ-SF) was administered to the participants through an online survey. The Wilcoxon signed-rank test and the Kruskal-Wallis rank-sum test were used for statistical analyses. Results: As expected, we observed a significant decrease of the total weekly energy expenditure during the COVID-19 quarantine (p < 0.001). A significant variation in the MET-min/wk in the before quarantine condition (p = 0.046) and in the difference between before and during quarantine (p = 0.009) was found for males and females. The male group decreased the PA level more than the female one. Moreover, a significant difference in the MET-min/wk was found among groups distributions of BMI (p < 0.001, during quarantine) and of age (p < 0.001, both before and during quarantine). In particular, the highest and the lowest levels of PA were reported by the young and the elderly, respectively, both before and during quarantine. Finally, the overweight group showed the lowest level of PA during quarantine. Conclusion: Based on our outcomes, we can determine that the current quarantine has negatively affected the practice of PA, with greater impacts among males and overweight subjects.     A Systematic Review of Pharmacologic and Rehabilitative Treatment of Small Fiber Neuropathies

    The aim of this systematic review is to guide the physician in defining the pharmacologicand rehabilitative therapeutic approaches for adopting the best strategies described in the currentliterature. The search was conducted in PubMed, EMBASE, Cochrane Library and Web of Scienceto identify the treatment of small fiber neuropathies. Two reviewers independently reviewed andcame to a consensus on which articles met inclusion/exclusion criteria. The authors excluded theduplicates, animal studies and included the English articles in which the treatment of patients withsmall fiber neuropathies was described. The search identified a total of 975 articles with the keywords“small fiber neuropathy” AND “rehabilitation” OR “therapy” OR “treatment”. Seventy-eight selectedfull-text were analyzed by the reviewers. Forty-two publications met the inclusion criteria and wereincluded in the systematic review to describe the rehabilitative and pharmacologic treatment of smallfiber neuropathies. Despite the range of different protocols of treatment for small fiber neuropathy,other robust trials are needed. In addition, always different therapeutic approaches are used; a uniqueprotocol could be important for the clinicians. More research is needed to build evidence for the beststrategy and to delineate a definitive therapeutic protocol

    Identification of dysregulated microRNA networks in schwann cell-like cultures exposed to immune challenge: Potential crosstalk with the protective VIP/PACAP neuropeptide system

    © 2018 by the authors. Licensee MDPI, Basel, Switzerland. Following peripheral nerve injury, dysregulations of certain non-coding microRNAs (miRNAs) occur in Schwann cells. Whether these alterations are the result of local inflammation and/or correlate with perturbations in the expression profile of the protective vasoactive intestinal peptide (VIP)/pituitary adenylate cyclase-activating polypeptide (PACAP) system is currently unknown. To address these issues, we aimed at profiling the expression of selected miRNAs in the rat RT4 Schwann cell line. Cells exposed to lipopolysaccharide (LPS), to mimic the local inflammatory milieu, were appraised by real-time qPCR, Western blot and ELISAs. We found that upon LPS treatment, levels of pro-inflammatory cytokines (IL-1ÎČ, -6, -18, -17A, MCP-1 and TNFα) increased in a time-dependent manner. Unexpectedly, the expression levels of VIP and PACAP were also increased. Conversely, levels of VPAC1 and VPAC2 receptors were reduced. Downregulated miRNAs included miR-181b, -145, -27a, -340 and -132 whereas upregulated ones were miR-21, -206, -146a, -34a, -155, -204 and -29a, respectively. Regression analyses revealed that a subset of the identified miRNAs inversely correlated with the expression of VPAC1 and VPAC2 receptors. In conclusion, these findings identified a novel subset of miRNAs that are dysregulated by immune challenge whose activities might elicit a regulatory function on the VIP/PACAP system

    Tackling dipeptidyl peptidase IV in neurological disorders

Dipeptidyl peptidase IV (DPP-IV) is a serine protease best known for its role in inactivating glucagon-like peptide-1 (GLP-1), pituitary adenylate cyclase-activating polypeptide (PACAP) and glucose-dependent insulinotropic peptide (GIP), three stimulators of pancreatic insulin secretion with beneficial effects on glucose disposal. Owing to the relationship between DPP-IV and these peptides, inhibition of DPP-IV enzyme activity is considered as an attractive treatment option for diabetic patients. Nonetheless, increasing studies support the idea that DPP-IV might also be involved in the development of neurological disorders with a neuroinflammatory component, potentially through its non-incretin activities on immune cells. In this review article, we aim at highlighting recent literature describing the therapeutic value of DPP-IV inhibitors for the treatment of such neurological conditions. Finally, we will illustrate some of the promising results obtained using berberine, a plant extract with potent inhibitory activity on DPP-IV
