    Eksplorasi Marka SSR Terpaut Sifat Toleransi Padi Gogo Terhadap Alumunium

    The objective of this research was to study molecular markers were linked to upland rice tolerance of Situ Patenggang x B11930F-TB2 against Al using SSR method. The plant materials were 36 lines of Situ Patenggang x B11930F-TB2 with categories tolerant, moderate, and susceptible to acid soil based on selection in East Lampung. IR6008032 line and ITA variety used as comparative control of tolerant and susceptible. The results showed that 40 primer pairs were applied to 36 lines and allegedly 3 primer pairs that are: RM205, RM257 and RM247 linked to Al tolerance. The markers location analysis on rice chromosome listed as follows: RM205 and RM257 were located on chromosome 9 and RM247 located on chromosome 12 with genetic distance 93,12 cM, 772.62 cM and 13,05 cM, respectively

    Tingginya keragaman varietas rambutan menjadi salah satu potensi untuk mengembangkan varietas unggul baru. Akan tetapi, ketersediaan informasi keragaman genetik rambutan masih terbatas. Penggunaan teknik marka RAPD (Random Amplified Polymorphic DNA) menjadi pilihan untuk analisis keragaman genetik tanaman karena cepat, mudah dan efisien. Tujuan penelitian ini adalah untuk mempelajari keragaman rambutan koleksi KPN (Kebun Plasma Nutfah) LIPI secara genotipik. Sembilan varietas rambutan digunakan dalam penelitian ini. Hasil analisis dengan menggunakan enam marka RAPD menghasilkan total pita 45. Hasil analisis polimorfisme menunjukkan keragaman antar varietas sebesar 56,6% dan nilai PIC (Polymorphic Information Content) sebesar 0,2-0,5. Dendogram UPGMA (Unweighted Pair Group Methode Arithmetic) membagi sembilan varietas rambutan dalam dua kelompok dengan indeks kesamaan sebesar 85-95%. Kelompok satu merupakan jenis varietas Si Nyonya sedangkan kelompok kedua terdiri dari varietas Rapiah, Aceh Lebak, Unidentified, Binjai, Parakan, Si Macan, Aceh Pagar dan Lebak Bulus. Perbedaan kelompok ini diduga berkaitan dengan sifat kelekatan daging buah terhadap biji. Varietas Aceh Lebak memiliki kekerabatan paling dekat dengan varietas unidentified sebesar 95%

    The aim of this research was to get upland rice lines with high yield, aluminum tolerance and resistance to blast. Genetic material used were 380 lines (F6) from six hybridization combinations evaluated under Al stress condition in environment endemic blast using augmented design with five randomized blocks. Performance of lines in the field and level of tolerance was tested. The results showed five genetic clusters formed from 380 lines of upland rice in a test based on quantitative characters. Cluster number 2 was the best cluster with characters: uniform growth vigor of plants, early flowering days, early maturing, medium plant height, high productive tillers, medium panicle length, high number of filled grain, the lowest of empty grains, high weight of 5 panicles and high yield potential. Aluminum tolerance and blast resistance were observed in almost all clusters. The lines in cluster 2 had the highest resistance to blast (98%) and tolerance to Al (94%)

    Water deficiency. Genetic engineering at the level of transcription factors (TF) is particulary a promising strategy in developing drought tolerant rice cultivar. HD-Zip genes are TF that function in plant adaptation to some environmental stresses including water deficit. The recombinant plasmid pC1301H Oshox6 which contained HD-Zip Oshox6 gene was placed under a drought inducible promoter called LEA promoter, gusA and hpt genes were driven with CaMV promoter. The aim of research was to obtain indica rice transgenic plants of Batutegi and Kasalath cultivars using pC1301H Oshox6 plasmid. Recombinant plasmid was transformed into immature rice embryos using Agrobacterium tumefaciens. Kasalath cultivar showed a better capacity to form embryogenic calli compared to Batutegi. Transformation efficiency of Batutegi is lower (1.5 - 0.3%) than Kasalath (2.2-28.3%). Regeneration efficiency is 25-83.3% and 7.7-100% for Batutegi and Kasalath, respectively. Number of putative transformant plantlets of Batutegi and Kasalath are 63 and 48 plantlets, respectively.     Response of Upland Rice towards Biological Fertilizer on the Dry Land in South Konawe District, South East Sulawesi. As the productivity of upland rice is still low, it requires technology improvement such as high yield variety and biological fertilizer. The purpose of this research was to determine the adaptability of upland rice varieties combined with biological fertilizer application. The experiment was conducted in the dry land of South Konawe District, South East Sulawesi using a factorial randomized block design with four replications of two factors. The first factor was three upland rice varieties namely Inpago LIPI Go1, Inpago LIPI Go2 and local varieties Kolono; the second factor was the combination of inorganic fertilizers and biological fertilizers. The experiment was carried out from December 2013 to March 2014. The results showed that the highest productivity was achieved by Inpago LIPI Go2 (4.5 to 5.2 t/ha) combined with the fertilizer combination as follow; Biofertilizer; Biofertilizer + 25% of recommended inorganic fertilizer; and Biofertilizer + 50% of recommended inorganic fertilizer without loosing the yield

    Tingginya keragaman Syzygium, khususnya jambu air disebabkan oleh penyerbukan silang, baik yang terjadi secara alami maupun akibat perbuatan manusia. Ketersediaan informasi keragaman jambu air koleksi Kebun Plasma Nutfah (KPN) Tanaman Cibinong sampai saat ini belum ada dan belum pernah dilakukan. Tujuan penelitian ini ialah mempelajari keragaman morfologi dan genetik tanaman jambu air koleksi KPN Cibinong.Lima jambu air koleksi KPN diamati keragaman morfologinya dan tujuh jambu air koleksi KPN diamati genetiknya.Analisis morfologi dilakukan dengan pengamatan pada karakter daun dan bunga, sedangkan analisis genetik menggunakan primerRandom Amplified Polymorphic DNA (RAPD). Data diolah menggunakan perangkat lunak SPSS, XLSTAT, NTSYS-pc versi 2.2 dan Power Marker. Hasil pengamatan morfologi pada lima jambu air diperoleh tiga kelompok yaitu Cincalo merah, Semarang merah, Semarang hijau (I), Jambu air lokal (II) dan Sigondrong (III). Berdasarkan analisis komponen utama, warna batang dekat daun, susunan tulang daun dan warna pangkal tangkai sari menjadi karakter yang paling berkontribusi dalam keragaman. Sementara hasil analisis genetik pada tujuh jambu air menggunakan 14 primer RAPD dibagi menjadi 3 kelompok yaitu Citra (I), Jambu air lokal (II), Semarang hijau, Merah delima, Sigondrong, Semarang merah dan Cincalo merah (III). Rerata jumlah pita sebanyak 5,79 dan rerata polimorfis pita 4,36 dengan rerata persentase polimorfisme sebesar 72,76%. Empat primer RAPD yaitu OPA-5, OPA-9, OPA-17 dan OPA-18 memberikan nilai Polymorphic Information Content (PIC) yang tinggi sebesar 0,7. Analisis morfologi dan genetik dalam penelitian ini diharapkan dapat menjadi dasar dalam perakitan varietas unggul dan konservasi tanaman jambu air di KPN Cibinong
