    Assessment of atmospheric aerosols from two reanalysis products over Australia

    Assessments of atmospheric aerosols from reanalysis are important for understanding uncertainty in model simulations, and ultimately predictions, such as for solar power or air quality forecasts and assessments. This study intercompares total aerosol optical depth (AOD) and dust AOD (DAOD) from two global reanalyses datasets, the European Centre for Medium-Range Weather Forecasts (ECMWF) Monitoring Atmospheric Composition and Climate (MACC) and the NASA Modern-Era Retrospective Analysis for Research-2 (MERRA-2). These are evaluated against AeroSpan (Aerosol characterisation via Sun photometry: Australian Network) ground observations which forms part of the Aerosol Robotic Network (AERONET) over the Australian continent for the 2002–2012 period. During dust storms, AeroSpan/AERONET AOD measurements were missing due to cloud screening. To overcome validation limitations in sun photometry for dust events, a nephelometer's scattering coefficient is qualitatively compared against reanalysis of DAOD at a key dust storm activation site, Tinga Tingana in South Australia (~200 km east of Lake Eyre). A specific extreme event that occurred in 2009 originating from the Lake Eyre basin, a major dust source covering one-sixth of Australia, was studied. The results show that MERRA-2 reanalysis overestimates monthly total AOD twice as much compared to AeroSpan/AERONET ground observations but seems better correlated against AeroSpan/AERONET than ECMWF/MACC. Mean data of MERRA-2 time series over 10 years provide lower DAOD values and lower dust aerosol estimates than ECMWF/MACC reanalysis (over the Lake Eyre basin with spatial averaging). Specifically at Tinga Tingana, the correlation from MERRA-2 (0.45 correlation) and ECMWF/MACC (0.43 correlation) against AeroSpan/AERONET's AOD were similar. Between MERRA-2 and ECMWF/MACC decade long daily gridded DAOD, the correlation coefficient was high at 0.73, again indicating similarity between the datasets. MERRA-2 total AOD correlation is significantly higher (by 0.26) against AeroSpan/AERONET than ECMWF/MACC. MERRA-2 also provides higher AOD values in extreme cases which may correspond to dust storms. During dust storms, a hybrid strategy using nephelometers and hourly reanalysis from MERRA-2 is able to identify dust storms better than AeroSpan/AERONET. Overall, this work can enable and inform better aerosol data assimilation into forecast models such as for solar energy, agriculture or air quality over Australia

    Modulation of Cytochrome P450 Metabolism and Transport across Intestinal Epithelial Barrier by Ginger Biophenolics

    Natural and complementary therapies in conjunction with mainstream cancer care are steadily gaining popularity. Ginger extract (GE) confers significant health-promoting benefits owing to complex additive and/or synergistic interactions between its bioactive constituents. Recently, we showed that preservation of natural ‘‘milieu’’ confers superior anticancer activity on GE over its constituent phytochemicals, 6-gingerol (6G), 8-gingerol (8G), 10-gingerol (10G) and 6-shogaol (6S), through enterohepatic recirculation. Here we further evaluate and compare the effects of GE and its major bioactive constituents on cytochrome P450 (CYP) enzyme activity in human liver microsomes by monitoring metabolites of CYPspecific substrates using LC/MS/MS detection methods. Our data demonstrate that individual gingerols are potent inhibitors of CYP isozymes, whereas GE exhibits a much higher half-maximal inhibition value, indicating no possible herb-drug interactions. However, GE’s inhibition of CYP1A2 and CYP2C8 reflects additive interactions among the constituents. In addition, studies performed to evaluate transporter-mediated intestinal efflux using Caco-2 cells revealed that GE and its phenolics are not substrates of P-glycoprotein (Pgp). Intriguingly, however, 10G and 6S were not detected in the receiver compartment, indicating possible biotransformation across the Caco-2 monolayer. These data strengthen the notion that an interplay of complex interactions among ginger phytochemicals when fed as whole extract dictates its bioactivity highlighting the importance of consuming whole foods over single agents. Our study substantiates the need for an indepth analysis of hepatic biotransformation events and distribution profiles of GE and its active phenolics for the design of safe regimens

    Quantification of experimental venous thrombus resolution by longitudinal nanogold-enhanced micro-computed tomography

    AbstractIntroductionThe assessment of thrombus size following treatments directed at preventing thrombosis or enhancing its resolution has generally relied on physical or histological methods. This cross-sectional design imposes the need for increased numbers of animals for experiments. Micro-computed tomography (microCT) has been used to detect the presence of venous thrombus in experimental models but has yet to be used in a quantitative manner. In this study, we investigate the use of contrast-enhanced microCT for the longitudinal assessment of experimental venous thrombus resolution.Materials and methodsThrombi induced by stenosis of the inferior vena cava in mice were imaged by contrast-enhanced microCT at 1, 7 and 14days post-induction (n=18). Thrombus volumes were determined longitudinally by segmentation and 3D volume reconstruction of microCT scans and by standard end-point histological analysis at day 14. An additional group of thrombi were analysed solely by histology at 1, 7 and 14days post-induction (n=15).ResultsIVC resident thrombus was readily detectable by contrast-enhanced microCT. MicroCT-derived measurements of thrombus volume correlated well with time-matched histological analyses (ICC=0.75, P<0.01). Thrombus volumes measured by microCT were significantly greater than those derived from histological analysis (P<0.001). Intra- and inter-observer analyses were highly correlated (ICC=0.99 and 0.91 respectively, P<0.0001). Further histological analysis revealed noticeable levels of contrast agent extravasation into the thrombus that was associated with the presence of neovascular channels, macrophages and intracellular iron deposits.ConclusionContrast-enhanced microCT represents a reliable and reproducible method for the longitudinal assessment of venous thrombus resolution providing powerful paired data

    Strain Invariant Failure Theory – Part 1: An extensible framework for predicting the mechanical performance of fibre reinforced polymer composites

    Failure prediction for composite materials is still a major area of interest within both the academic literature and the composite industry. This paper presents a modelling and experimental framework which has been developed for the application of Strain Invariant Failure Theory (SIFT), but which can be adapted to other failure prediction methods if required. SIFT is set of physics-based failure criteria developed to predict the onset of irreversible deformation of glassy polymers. The paper provides an overview of the available evidence for SIFT and provides a complete methodology for applying SIFT to predict the onset of failure in a composite component.Fundamental to the method presented is a modular and hierarchical dehomogenisation procedure; allowing for the evaluation of stress and strain state variables within the constituent components of the composite. Due to the constant evolution of composite characterisation techniques, the method presented in this paper is designed to be extensible (i.e. accommodating of new materials and product forms) and modular (i.e. subsections of the method can be easily substituted or refined). Known limitations of the current method are clearly presented and discussed. A companion paper will present validation of the approach for simple uniaxial and biaxial coupon experiments and will discuss how the method can be applied to a range of existing composite characterisation testing standards
