    The Effect of Heating on Physicochemical Characteristics of Rice Brand Oil

    Rice bran oil has a potential in lowering blood cholesterol. The oil content extracted from rice bran isa influenced by several factors such as raw material and processing method. This research was conducted to study the effect of heating on the physicochemical characteristics of rice bran oil. The study was carried out at the Food Technology and Nutrition Laboratory and Biotechnological Science Laboratory of the Inter University Center, Bogor Agricultural University, from February to MAy 1997. Bran used was from IR64 rice variety obtained from a local milling rice in Tambak Dahan, Binong-Subang, West Java. Rice bran was subjected to different period of heating (0, 15 and 30 minutes) at 121oC by using an autoclave, then the oil was extraxted by using hexane solvent followed by standard processing at room temperature (28-30oC) and 40oC for two hours. The parameters observed were oil content and its quality such as saponification, iodine, acid, peroxide, tiobarbituric acid, and triglyceric acid values as well as oil color. The results showed that oil extracted from rice bran was high, ranging from 9.65 to 11.02%. Heating (sterilizing) of the rice bran for 15 and 30 minutes at 120oC proir to extraction increased the oil content. The Quality of rice bran oil extracted met the standard of AOAC

    The objective of this experiment was to evaluate the effects of isoflavone-riched soybean flour, zinc (Zn) and vitamin E on quality of rats spermatozoa as animal model. Diet was given as isonitrogen and isocaloric with 15% of dietary protein. Thirty male Sprague Dawley post weaning rats (21 days old) were divided into six groups and treated with isoflavone-riched soybean flour, Zn and vitamin E in different combination. Isoflavone-riched soybean flour (3 mg/day) was given by oral administration, whereas Zn and vitamin E were mixed with the basic diet. The treatments were given for 2 months. Thirty female Sprague Dawley post weaning rats (21 days old) were used to observe male fertility. Results indicated that synergic interaction between isoflavone-riched soybean flour, Zn and vitamin E increased motility and concentration of rats spermatozoa. The treatment was not significant on relative weight of testis, conception rate and foetus size. The best results was shown by the group which was given isoflavone-riched soybean flour with diet containing both Zn and vitamin E, i.e. motility rate of 79.50±1.12%, sperm concentration of 1636.90±87.95 million/ml, and abnormality rate of 9.00±0.70%. Conception rate and the number of foetus on female rats were 100% and 11±0.71, respectively. It is concluded that combination treatment of isoflavone-riched soybean flour, Zn and vitamin E on male rats showed the best fertility in comparison with single treatment and the other combination.     The Tuber Extract and Flour of Dioscorea Alatanormalize the Blood Lipid Profile of Rabbits Treated with High Cholesterol Diets

    Background: Dioscorea alata(DA) tuber has potential to prevent the condition of hyperlipidemia due to the bioactive compound, such as anthocyanins, diosgenin, and dietary fiber that beneficial in normalizing blood lipid profiles. In this research, the effect of water extract and flour of DA tuber administration was examined on rabbits treated with high cholesterol diets.Methods:DA tuber extract and flour were administrated to the rabbits for 60 days using completely randomised design. The ration treatment are as follows: 1) Basal ration as negative control (K0), 2) Basal ration + 0.5% cholesterol, as positive control (K1), 3) Basal ration + 0.5% cholesterol + DA extract 1.8 g/100 g (KE1), 4) Basal ration + cholesterol 0.5% + DA extract 3.6 g/100 g (KE2), 5) Basal ration with 15% DA flour + 0.5% cholesterol (KT1) and 6) Basal ration with 30% DA flour + 0.5% cholesterol (KT2). The Total cholesterol, HDL, LDL cholesterol in serum were analysed at baseline, days 28, days 56 and at the end of study.Results:The administration of high cholesterol (1%) ration increased blood lipid levels by 16 fold compared to that of control. The administration of 15% and 30% of DA flour could maintain blood lipid profile to normal condition, in particular at 30% substitution DA flour. However the water extract of DA can not maintain a normal blood lipids of high cholesterol treated rabbitsConclusion: Dioscorea alata flour has suggested to have anti-hyperlipidemia effect.     Effect of isoflavone-enriched soybean flour, zinc (Zn) and vitamin E in the ration on testosterone level and total permatogenic cell in seminiferous tubules of rat

    The objective of this experiment are to evaluate the effects of isoflavoneen-enriched soybean flour, zinc (Zn) and vitamin E on testosterone level of male rats and total spermatogenic cells in the seminiferous tubules of rat testes as animal model. Diet was given as isonitrogen and isocaloric with 15% of dietary protein. Thirty male Sprague Dawley rats were divided into six groups and treated with isoflavoneen-riched soybean flour, Zn and vitamin E in different combination. Isoflavoneen-riched soybean flour (3mg/day) was given by oral administration, whereas Zn and vitamin E were mixed wih the basic diet. The treatment was done for 2 month. Results indicated that complete treatment of isoflavoneen-riched soybean flour, Zn and vitamin E on male rats increased testosteron level and total spermatogenic cells in comparison with single treatment and the other combination. The best results showed in the group that given isoflavoneen-enriched soybean flour with diet containing both Zn and vitamin E i.e; testosteron level 3.49 ± 0.31 ng/ml; while the number of spermatogonia, spermatocytes, early spermatids, late spermatids, and total spermatogenic cells were 37.56 ± 4.48, 67 ± 4.72, 287.11 ± 31.75, 227.22 ± 29.78, and 618.89 ± 47.38, respectively. It was concluded that synergic interaction between isoflavoneen-enriched soybean flour, Zn and vitamin E increased testosteron level and total spermatogenic cells of rat testes.       Key Words: Isoflavone-Riched Soybean Flour, Zn, Vitamin E, Testosterone, Spermatogenic Cell

    Anthiypercholesterolaemic Activity of Zincophyllin

    Pheophytin was obtained from chlorophyll that was extracted from fresh cassava leaves by use of ethanol. One hundred to 200 ppm of zinc was added to pheophytin to produce a complex of zinc-pheophytin (“zincophyllin”). The antihypercholesterolaemic effect of “zincophyllin” was studied in 20 male New Zealand White rabbits fed on a low-cholesterol (0.1%) diet for 12 weeks. Five animals were given 33.4 mg/d (doses I), 5 animals received 100.2 mg/d (doses II) of “zincophyllin”, 5 animals received standard diet as negative controls, and the remaining 5 animals served as cholesterol-fed controls (positive controls). Animal receiving dose Ii had significantly lower serum total and LDL-cholesterol concentrations. Positive control group and doses I group had significantly higher serum total cholesterol. Cholesterol concentration of group II slightly increased for 4 weeks, but decreased to normal until 12 weeks. Triglyceride concentration of all rabbits groups were in normal level, but LDL-cholesterol concentration of positive control group and doses I group were higher than others. It was suggested that the antihypercholesterolaemic effect of “zincophyllin” may benefit rabbits by inhibiting the increase of serum cholesterol concentration.     (the Effect of Heat Application in Producing VCO and the Efficacy of Resulted VCO in Reducing Blood Glucose of Diabetes Mellitus Rat of Sprague Dawley)

    Virgin Covonut Oil (VCO) used to be produced with various methods and each methods claims ait advantege especially in its lauric acid content. The objective of this research was to study the effect of heat application in producing VCO and the efficacy of result VCO in reducing blood glucose of diabetes mellitus (DM) rat of sprague Dawley. Three types of samples were used in the study, i.e., (1) VCO produced without heat application and (2) VCO producd with controlled heat application and (3) Ordinary coconut cooking oil produced with the application of severe heating as well as typical chemical tretment. Each sample was introduced orally to each group consisting of 5 rats. Other two groups consisting of 5 rats were also used as control, i.e., (1) negative control was of healty rat and (2) positive control was of DM rats. The animal study was conduced for 28 days and the observation was carried out on the amount of feed consumed, body weight and blood glucose. At the end of observation, all the rats were terminated and analysis was carried out on blood chorestrol level. This study revealed no significant different between VCO and Coconut Cooking Oil in body weight gain effect. The content of bioactive conpound of Lauric Acid in two types of VCO and Coconut Cooking Oil were also not significantly different. However, the VCO produced without heating showed the most significant effect in reducing blood glucose (corellation value of-0,99 and significant value at a a=1%). All the VCO samples were also not significant in affecting the level of total cholestrol, HDL, LDL, and triglyceide of blood serum of DM rat
