    Experimental Nonlinear Control for Flutter Suppression in a Nonlinear Aeroelastic System

    Experimental implementation of input–output feedback linearization in controlling the dynamics of a nonlinear pitch–plunge aeroelastic system is presented. The control objective is to linearize the system dynamics and assign the poles of the pitch mode of the resulting linear system. The implementation 1) addresses experimentally the general case where feedback linearization-based control is applied using as the output a degree of freedom other than that where the physical nonlinearity is located, using a single trailing-edge control surface, to stabilize the entire system; 2) includes the unsteady effects of the airfoil’s aerodynamic behavior; 3) includes the embedding of a tuned numerical model of the aeroelastic system into the control scheme in real time; and 4) uses pole placement as the linear control objective, providing the user with flexibility in determining the nature of the controlled response. When implemented experimentally, the controller is capable of not only delaying the onset of limit-cycle oscillation but also successfully eliminating a previously established limit-cycle oscillation. The assignment of higher levels of damping results in notable reductions in limit-cycle oscillation decay times in the closed-loop response, indicating good controllability of the aeroelastic system and effectiveness of the pole-placement objective. The closed-loop response is further improved by incorporating adaptation so that assumed system parameters are updated with time. The use of an optimum adaptation parameter results in reduced response decay times

    Strongly aligned coastal boulders on Ko Larn island (Thailand): a proxy for past typhoon-driven high-energy wave events in the Bay of Bangkok

    © 2019 Institute of Australian Geographers Recent studies of elevated carbonate boulder deposits on several rock islands near Bangkok have indicated that Thailand\u27s capital city may not be as protected from typhoon strikes as previously thought. Here, new evidence is presented for past high-energy wave (HEW) events in the form of statistically significant patterns of boulder alignment on exposed rocky shorelines of Ko Larn island. The long-axis orientations of 193 coastal sandstone boulders were analysed across four study sites. Several scenarios for the unimodal, bimodal, and polymodal patterns found can be envisaged. Either the most recent HEW event was the strongest—in which case most clasts were rearranged unimodally (one observation site), or the strongest HEW event was earlier and subsequent weaker ones realigned only smaller boulders to produce bimodal or polymodal patterns (three observation sites). Inferred northeastward or eastward onshore flow directions are consistent with palaeo-typhoons penetrating into the Bay of Bangkok on northwestward curving tracks. The calculated minimum flow velocities required to transport all sampled boulders are 5.5–7.8 m s−1, similar to other findings throughout the Asia-Pacific region. It was observed that the absence of a fitted boulder geomorphology lends credence to the earlier proposed time frame of 150–200 years between typhoon phases in the upper Gulf of Thailand. The current work has provided additional insights into the characteristics of past HEW events that have a possibility of reoccurring again at some time in the future. Our findings continue to raise awareness for a reassessment of the risks of coastal hazards for the Chao Phraya River delta and densely populated Bangkok, for which storm surge modelling should be an urgent priority, so as to give better perceptions on how typhoon-driven marine incursion would impact the city

    Effects of differing oocyte-secreted factors during mouse in vitro maturation on subsequent embryo and fetal development

    Purpose We hypothesised that varying native oocyte-secreted factor (OSF) exposure or using different recombinant OSF peptides would have differential effects on post-in vitro maturation (IVM) embryo and fetal development. Methods Mouse cumulus oocyte complexes (COCs) were treated with the purified mature domain of GDF9 and/or BMP15 or were co-cultured with denuded oocytes (DOs) from 0 h or 3 h of IVM. DOs were matured for 3 h as either intact COCs+/-FSH before denuding, or as DOs + FSH. COCs were fertilised and blastocyst development was assessed on days 5 and 6, and either differentially stained for ICM numbers or vitrified/warmed embryos were transferred to recipients to assess implantation and fetal rates. Results No improvement in embryo development was observed with the addition of GDF9 and/or BMP15 to IVM. In contrast, embryos derived from COCs co-cultured with DOs had significantly improved blastocyst rates and ICM numbers compared to controls (P <0.05). The highest response was obtained when DOs were first added to COCs at 3 h of IVM, after being pre-treated (0–3 h) as COCs + FSH. Compared to control, co-culture with DOs from 3 h did not affect implantation rates but more than doubled fetal yield (21 % vs 48 %; P <0.05). GDF9 Western blot analysis was unable to detect any differences in quantity or form of GDF9 (17 and 65 kDa) in extracts of DO at 0 h or 3 h. Conclusions This study provides new knowledge on means to improve oocyte quality in vitro which has the potential to significantly aid human infertility treatment and animal embryo production technologies.J.     Blood-Brain Barrier Permeability of Normal Appearing White Matter in Relapsing-Remitting Multiple Sclerosis

    BACKGROUND: Multiple sclerosis (MS) affects the integrity of the blood-brain barrier (BBB). Contrast-enhanced T1 weighted magnetic resonance imaging (MRI) is widely used to characterize location and extent of BBB disruptions in focal MS lesions. We employed quantitative T1 measurements before and after the intravenous injection of a paramagnetic contrast agent to assess BBB permeability in the normal appearing white matter (NAWM) in patients with relapsing-remitting MS (RR-MS). METHODOLOGY/PRINCIPAL FINDINGS: Fifty-nine patients (38 females) with RR-MS undergoing immunomodulatory treatment and nine healthy controls (4 females) underwent quantitative T1 measurements at 3 tesla before and after injection of a paramagnetic contrast agent (0.2 mmol/kg Gd-DTPA). Mean T1 values were calculated for NAWM in patients and total cerebral white matter in healthy subjects for the T1 measurements before and after injection of Gd-DTPA. The pre-injection baseline T1 of NAWM (945±55 [SD] ms) was prolonged in RR-MS relative to healthy controls (903±23 ms, p = 0.028). Gd-DTPA injection shortened T1 to a similar extent in both groups. Mean T1 of NAWM was 866±47 ms in the NAWM of RR-MS patients and 824±13 ms in the white matter of healthy controls. The regional variability of T1 values expressed as the coefficient of variation (CV) was comparable between the two groups at baseline, but not after injection of the contrast agent. After intravenous Gd-DTPA injection, T1 values in NAWM were more variable in RR-MS patients (CV = 0.198±0.046) compared to cerebral white matter of healthy controls (CV = 0.166±0.018, p = 0.046). CONCLUSIONS/SIGNIFICANCE: We found no evidence of a global BBB disruption within the NAWM of RR-MS patients undergoing immunomodulatory treatment. However, the increased variation of T1 values in NAWM after intravenous Gd-DTPA injection points to an increased regional inhomogeneity of BBB function in NAWM in relapsing-remitting MS

    Baculovirus Capsid Display Potentiates OVA Cytotoxic and Innate Immune Responses

    Baculoviruses (BV) are DNA viruses that are pathogenic for insects. Although BV infect a range of mammalian cell types, they do not replicate in these cells. Indeed, the potential effects of these insect viruses on the immune responses of mammals are only just beginning to be studied. We show in this paper that a recombinant Autographa californica multiple nuclear polyhedrosis virus carrying a fragment of ovalbumin (OVA) on the VP39 capsid protein (BV-OVA) has the capacity to act as an adjuvant and vector of antigens in mice, thereby promoting specific CD4 and cytotoxic T cell responses against OVA. BV also induced in vivo maturation of dendritic cells and the production of inflammatory cytokines, thus promoting innate and adaptive immune responses. The OVA-specific response induced by BV-OVA was strong enough to reject a challenge with OVA-expressing melanoma cells (MO5 cells) and effectively prolonged survival of MO5 bearing mice. All these findings, together with the absence of pre-existing immunity to BV in humans and the lack of viral gene expression in mammalian cells, make BV a candidate for vaccination

    Gonadal Transcriptome Alterations in Response to Dietary Energy Intake: Sensing the Reproductive Environment

    Reproductive capacity and nutritional input are tightly linked and animals' specific responses to alterations in their physical environment and food availability are crucial to ensuring sustainability of that species. We have assessed how alterations in dietary energy intake (both reductions and excess), as well as in food availability, via intermittent fasting (IF), affect the gonadal transcriptome of both male and female rats. Starting at four months of age, male and female rats were subjected to a 20% or 40% caloric restriction (CR) dietary regime, every other day feeding (IF) or a high fat-high glucose (HFG) diet for six months. The transcriptional activity of the gonadal response to these variations in dietary energy intake was assessed at the individual gene level as well as at the parametric functional level. At the individual gene level, the females showed a higher degree of coherency in gonadal gene alterations to CR than the males. The gonadal transcriptional and hormonal response to IF was also significantly different between the male and female rats. The number of genes significantly regulated by IF in male animals was almost 5 times greater than in the females. These IF males also showed the highest testosterone to estrogen ratio in their plasma. Our data show that at the level of gonadal gene responses, the male rats on the IF regime adapt to their environment in a manner that is expected to increase the probability of eventual fertilization of females that the males predict are likely to be sub-fertile due to their perception of a food deficient environment
