    Isolation of anaerobic, extremely thermophilic, sulphur metabolising archaebacteria from New Zealand hot springs

    Enrichments of New Zealand geo-thermal samples, initiated in anaerobic sulphur-containing media and incubated at temperatures above 85°C, yielded rod and coccal shaped organisms which possessed archaebacterial characteristics. Pure cultures were isolated and characterised. Five of the seven isolates, which were rod-shaped organisms and did not have an obligate requirement for sulphur respiration, were similar to Ther-moproteus sp. but had more neutral pH optima for growth. Three of these five Thermoproteus sp. were obligate heterotrophs, which has not previously been reported. The two coccal isolates had an obligate requirement for sulphur as an electron acceptor and were similar to Desulfurococcus sp. but again with more neutral pH optima for growth

    Identification of a mutation in the para-sodium channel gene of the cattle tick Rhipicephalus (Boophilus) microplus associated with resistance to synthetic pyrethroid acaricides

    Resistance against synthetic pyrethroid (SP) products for the control of cattle ticks in Australia was detected in the field in 1984, within a very short time of commercial introduction. We have identified a mutation in the domain II S4-5 linker of the para-sodium channel that is associated with resistance to SPs in the cattle tick Rhipicephalus (Boophilus) microplus from Australia. The cytosine to adenine mutation at position 190 in the R. microplus sequence AF134216, results in an amino acid substitution from leucine in the susceptible strain to isoleucine in the resistant strain. A similar mutation has been shown to confer SP resistance in the whitefly, Bemisia tabaci, but has not been described previously in ticks. A diagnostic quantitative PCR assay has been developed using allele-specific Taqman® minor groove-binding (MGB) probes. Using the assay to screen field and laboratory populations of ticks showed that homozygote allelic frequencies correlated highly with the survival percentage at the discriminating concentration of cypermethrin

    Development of the Multi-Level Seismic Receiver (MLSR)

    The Advanced Geophysical Technology Department (6114) and the Telemetry Technology Development Department (2664) have, in conjunction with the Oil Recovery Technology Partnership, developed a Multi-Level Seismic Receiver (MLSR) for use in crosswell seismic surveys. The MLSR was designed and evaluated with the significant support of many industry partners in the oil exploration industry. The unit was designed to record and process superior quality seismic data operating in severe borehole environments, including high temperature (up to 200{degrees}C) and static pressure (10,000 psi). This development has utilized state-of-the-art technology in transducers, data acquisition, and real-time data communication and data processing. The mechanical design of the receiver has been carefully modeled and evaluated to insure excellent signal coupling into the receiver

    Strength Reduction in Electrical and Elastic Networks

    Particular aspects of problems ranging from dielectric breakdown to metal insu- lator transition can be studied using electrical o elastic networks. We present an expression for the mean breakdown strength of such networks.First, we intro- duce a method to evaluate the redistribution of current due to the removal of a finite number of elements from a hyper-cubic network of conducatances.It is used to determine the reduction of breakdown strength due to a fracture of size κ\kappa.Numerical analysis is used to show that the analogous reduction due to random removal of elements from electrical and elastic networks follow a similar form.One possible application, namely the use of bone density as a diagnostic tools for osteorosporosis,is discussed.Comment: one compressed file includes: 9 PostScrpt figures and a text fil

    Compression ultrasonography for false femoral artery aneurysms: Hypocoagulability is a cause of failure

    Objectives:false femoral artery aneurysm is an occasional complication of percutaneous cardiovascular radiological procedures. Compression ultrasonography causes thrombosis non-invasively, reducing need for operative intervention. The technique fails in a proportion of cases. Analysis was undertaken to identify causes of failure.Design:prospective open study.Materials and Methods:patients presenting with false femoral artery aneurysm since 1984 were identified from a computerised database (BIPAS). Since 1993 compression ultrasonography has been performed as first line treatment according to a standard protocol. Prospectively collected ultrasonographic data and case notes were reviewed to identify causes of failed compression.Results:false femoral artery aneurysm occurred as a complication in 32/26 687 (0.12%) cardiovascular radiological procedures. Eighteen aneurysms were treated by compression. The technique was successful in 11/18 (61%) cases but primary failure occurred in seven cases. Six out of seven had bleeding abnormalities (Chi-squared analysis with Yates correction 10.55, p=0.0012), four were anticoagulated and compression was subsequently successful following reversal of warfarin therapy in three of these patients. In 4/18 cases surgical repair was necessary.Conclusions:compression ultrasonography is an effective treatment of false femoral aneurysms, however, hypocoagulability is a significant cause of failure. For patients in whom anticoagulation cannot be reversed, primary surgical repair should be considered

    Applications of Urban Tree Canopy Assessment and Prioritization Tools: Supporting Collaborative Decision Making to Achieve Urban Sustainability Goals

    Urban Tree Canopy (UTC) Prioritizations can be both a set of geographic analysis tools and a planning process for collaborative decision-making. In this paper, we describe how UTC Prioritizations can be used as a planning process to provide decision support to multiple government agencies, civic groups and private businesses to aid in reaching a canopy target. Linkages to broader City-scale sustainability plans are explored. This article represents an extension and update to the UTC Canopy Goal Setting Guide by Raciti et al (2006). We conclude with recommendations for a market-like analysis of neighborhoods to better match planting initiatives to particular neighborhoods’ motivations, capacities and interests in order to improve the adoption of improved urban forestry practices

    Prioritizing Preferable Locations for Increasing Urban Tree Canopy in New York City

    This paper presents a set of Geographic Information System (GIS) methods for identifying and prioritizing tree planting sites in urban environments. It uses an analytical approach created by a University of Vermont service-learning class called “GIS Analysis of New York City\u27s Ecology” that was designed to provide research support to the MillionTreesNYC tree planting campaign. These methods prioritize tree planting sites based on need (whether or not trees can help address specific issues in the community) and suitability (biophysical constraints and planting partners’ existing programmatic goals). Criteria for suitability and need were based on input from three New York City tree-planting organizations. Customized spatial analysis tools and maps were created to show where each organization may contribute to increasing urban tree canopy (UTC) while also achieving their own programmatic goals. These methods and associated custom tools can help decision-makers optimize urban forestry investments with respect to biophysical and socioeconomic outcomes in a clear and accountable manner. Additionally, the framework described here may be used in other cities, can track spatial characteristics of urban ecosystems over time, and may enable further tool development for collaborative decision-making in urban natural resource management

    Identifying sulphate-reducing and magnetotactic bacteria in a hyperalkaline cave system

    Sulphate-reducing bacteria (SRB) and magnetotactic bacteria (MTB) are two special groups of prokaryotes that emerged early in Earth’s history. These extremotolerant bacterial groups have rarely been identified or studied within cave and karst environments, especially hyperalkaline cave systems (pH > 9), such as Poole’s Cavern in Derbyshire, England. In this study, we identify the sulphate-reducing MTB Desulfovibrio magneticus, within this hyperalkaline cave system. It appears to survive in a diverse range of environments including soil, stalactites and cave sediments. Additionally, we identify various extremotolerant SRB in similar Poole’s Cavern environments. We show that these SRB and MTB can move successfully into subsurface environments and adapt concomitantly to the anomalous pH, saline, and relatively nutrient-poor conditions found in Poole’s Cavern. These findings are significant to our understanding of microorganisms on early Earth because it is believed, but not proven, that underground environments might have been hot spots for early microbial life. We expect that these early bacteria would have been able to adopt similar adaptation strategies, transferring and acclimatizing to underground environments, in ways comparable to the SRB and MTB identified in this study

    Using Nonlinear Response to Estimate the Strength of an Elastic Network

    Disordered networks of fragile elastic elements have been proposed as a model of inner porous regions of large bones [Gunaratne et.al., cond-mat/0009221, http://xyz.lanl.gov]. It is shown that the ratio Γ\Gamma of responses of such a network to static and periodic strain can be used to estimate its ultimate (or breaking) stress. Since bone fracture in older adults results from the weakening of porous bone, we discuss the possibility of using Γ\Gamma as a non-invasive diagnostic of osteoporotic bone.Comment: 4 pages, 4 figure
