    No full text

A Novel PHOX/CD38/MCOLN1/TFEB Axis Important For Macrophage Activation During Bacterial Phagocytosis [preprint]

Macrophages are a key and heterogenous class of phagocytic cells of the innate immune system, which act as sentinels in peripheral tissues and are mobilized during infection. Macrophage activation in the presence of bacterial cells and molecules entails specific and complex programs of gene expression. How such triggers elicit the gene expression programs is incompletely understood. We previously discovered that transcription factor TFEB is a key contributor to macrophage activation during bacterial phagocytosis. However, the mechanism linking phagocytosis of bacterial cells to TFEB activation remained unknown. In this article, we describe a previously unknown pathway that links phagocytosis with the activation of TFEB and related transcription factor TFE3 in macrophages. We find that phagocytosis of bacterial cells causes an NADPH oxidase (PHOX)-dependent oxidative burst, which activates enzyme CD38 and generates NAADP in the maturing phagosome. Phago-lysosome fusion brings Ca2+ channel TRPML1/MCOLN1 in contact with NAADP, causing Ca2+ efflux from the lysosome, calcineurin activation, and TFEB nuclear import. This drives TFEB-dependent expression of important pro-inflammatory cytokines, such as IL-1α, IL-1β, and IL-6. Thus, our findings reveal that TFEB activation is a key regulatory event for the activation of macrophages. These findings have important implications for infections, cancer, obesity, and atherosclerosis

Magnetic Behavior Of Rare-earth Iron-rich Intermetallic Compounds

    Get PDF
The thermal variation of lattice parameters of rare earth-transition metal intermetallic compounds rich in Fe, Co, or Ni in the temperature range of 25-900oK is studied. For the Fe compounds, negative thermal expansion is observed below their magnetic ordering temperatures regardless of the nature of substitutional ordering. For Co and Ni compounds, the thermal expansion behavior is normal. It is then concluded that for the Fe compounds, the magnetic properties are mainly determined by the Fe-Fe interatomic distances and the number of Fe nearest neighbors, whereas for Co and Ni compounds the magnetic properties are determined by the conduction electron transfer from the rare earth to the 3 d band of Co or Ni, The anomalous thermal expansion of Fe compounds and the metamagnetic transition of the Lu compound is explained in terms of the distance dependence. of the interaction energy as proposed by Neel. © 1971, IEEE.

CADDA: Class-wise Automatic Differentiable Data Augmentation for EEG Signals

    Get PDF
International audienceData augmentation is a key element of deep learning pipelines, as it informs the network during training about transformations of the input data that keep the label unchanged. Manually finding adequate augmentation methods and parameters for a given pipeline is however rapidly cumbersome. In particular, while intuition can guide this decision for images, the design and choice of augmentation policies remains unclear for more complex types of data, such as neuroscience signals. Besides, class-dependent augmentation strategies have been surprisingly unexplored in the literature, although it is quite intuitive: changing the color of a car image does not change the object class to be predicted, but doing the same to the picture of an orange does. This paper investigates gradient-based automatic data augmentation algorithms amenable to class-wise policies with exponentially larger search spaces. Motivated by supervised learning applications using EEG signals for which good augmentation policies are mostly unknown, we propose a new differentiable relaxation of the problem. In the class-agnostic setting, results show that our new relaxation leads to optimal performance with faster training than competing gradient-based methods, while also outperforming gradient-free methods in the class-wise setting. This work proposes also novel differentiable augmentation operations relevant for sleep stage classification
