GM-CSF-Producing Th Cells in Rats Sensitive and Resistant to Experimental Autoimmune Encephalomyelitis
Given that granulocyte macrophage colony-stimulating factor (GM-CSF) is identified as the key factor to endow auto-reactive Th cells with the potential to induce neuroinflammation in experimental autoimmune encephalomyelitis (EAE) models, the frequency and phenotype of GM-CSF-producing (GM-CSF+) Th cells in draining lymph nodes (dLNs) and spinal cord (SC) of Albino Oxford (AO) and Dark Agouti (DA) rats immunized for EAE were examined. The generation of neuroantigen-specific GM-CSF+ Th lymphocytes was impaired in dLNs of AO rats (relatively resistant to EAE induction) compared with their DA counterparts (susceptible to EAE) reflecting impaired CD4+ lymphocyte proliferation and less supportive of GM-CSF+ Th cell differentiation dLN cytokine microenvironment. Immunophenotyping of GM-CSF+ Th cells showed their phenotypic heterogeneity in both strains and revealed lower frequency of IL-17+ IFN-gamma+, IL-17+ IFN-gamma-, and IL-17-IFN-gamma+ cells accompanied by higher frequency of IL-17-IFN-gamma- cells among them in AO than in DA rats. Compared with DA, in AO rats was also found (i) slightly lower surface density of CCR2 (drives accumulation of highly pathogenic GM-CSF+ IFN-gamma+ Th17 cells in SC) on GM-CSF+ IFN-gamma+ Th17 lymphocytes from dLNs, and (ii) diminished CCL2 mRNA expression in SC tissue, suggesting their impaired migration into the SC. Moreover, dLN and SC cytokine environments in AO rats were shown to be less supportive of GM-CSF+ IFN-gamma+ Th17 cell differentiation (judging by lower expression of mRNAs for IL-1 beta, IL-6 and IL-23/p19). In accordance with the (i) lower frequency of GM-CSF+ Th cells in dLNs and SC of AO rats and their lower GM-CSF production, and (ii) impaired CCL2 expression in the SC tissue, the proportion of proinflammatory monocytes among peripheral blood cells and their progeny (CD45(hi) cells) among the SC CD11b+ cells were reduced in AO compared with DA rats. Collectively, the results indicate that the strain specificities in efficacy of several mechanisms controlling (auto) reactive CD4+ lymphocyte expansion/differentiation into the cells with pathogenic phenotype and migration of the latter to the SC contribute to AO rat resistance to EAE
Pemphigus herpetiformis - a case report of a rare form of pemphigus and review of the literature
Introduction. Pemphigus herpetiformis is the rare variant of pemphigus with
characteristic clinical features, histopathological findings different from
the convectional pemphigus, and immunological findings consistent with
pemphigus. Case report. We presented a 65-year-old woman with initial
pruritus followed by pruritic urticarial papules and plaques, some with
annular rings of tense vesicles on the periphery, on the trunk and
extremities, with no mucous lesions. Histopathological examination
demonstrated spongiosis and intraepidermal vesicles in the mid or subcorneal
epidermis in some biopsy specimen, with neutrophil and eosinophil
infiltrate. Direct immunoflorescent microscopy revealed intercellular IgG
deposition, most prominent in the upper layers of epidermis. Indirect
immunoflorescent microscopy showed intercellular binding of IgG
autoantibodies in the patientās sera. Initially the patient was threated
with systemic corticosteroids and azathioprine, but dapson provided complete
clinical remission. Conclusion. This entity was established 40 years ago,
and around 100 patients have been reported worldwide. It is important to be
aware of this particular form of pemphigus because clinical presentation,
course of the disease and therapeutic approach are different from
conventional forms of pemphigus

Comparative analysis of the first permanent molars caries prevalence in younger and older school children
Introduction. The data on the health-condition of the first permanent molars among children in our region is scarce. Therefore, the need for more thorough analysis of the state of health of these teeth in the children from our region imposes itself, along with the need to determine the most critical period in which a significant increase in caries prevalence of these teeth takes place. Aim. The aim of the research was to perform a comparative analysis of the prevalence, average rate and structure of decay, missed and filled first permanent molars in the children of the younger school age in relation to those of the older school age. Material and methods. The research included 1.119 examinees, aged from 7 to 14 years. Examinees were divided into two groups according to their age: examinees of the younger school age and older school age. Dental check-up of the students was performed by means of a dental probe and mirror and the health condition of the examined teeth was registered by means of Klein-Palmer's DMF system. Results. The research determined high values of the examined parameters of the first permanent molars caries prevalence which increases with the age of the examinees. In the examinees of the age of 8, 9, and 10, statistically more significant values of the examined teeth caries prevalence parameters were registered, in relation to the examinees of seven years of age. In the examinees of the older school age, statistically significantly higher values were registered only for TCI and ACI and only in the examinees aged 14 in relation to the examinees aged 11. With the age, the percent of decay is decreases, while the percent of filled and extracted first permanent molars increases. Conclusion. Results show that the period between 7 and 10 years of age is the most critical and the most important period for the implementation of preventive-prophylactic measures aimed to preserve the health of the first permanent molars
Ribavirin reduces clinical signs and pathological changes of experimental autoimmune encephalomyelitis in dark Agouti rats
The effect of ribavirin on development of experimental autoimmune encephalomyelitis (EAE) was investigated. The disease was induced in genetically susceptible Dark Agouti rats with syngeneic spinal cord homogenate in complete Freund's adjuvant (SCH-CFA). Depending on the amount of mycobacteria in CFA, the animals developed either moderate or severe EAE. Ribavirin (1-beta-Dribofuranosyl-1,2,4-triazole-3-carboxamide) was applied i.p. at a daily dosage of 30 mg/kg in two treatment protocols: from the start of immunization (preventive treatment) or from the onset of the first EAE signs after the induction (therapeutic treatment). Signs of EAE began between 7 and 9 days after induction and peaked at days 11-13. In moderate EAE (mean maximal severity score 3.33 +/- 0.21), the recovery was completed by days 23-26, whereas, in severe EAE (mean maximal severity score 4.5 +/- 0.23), obvious recovery was not detected. Preventive ribavirin treatment significantly decreased clinical signs after both moderate (score 1.75 +/- 0.25, P < 0.05) and severe (score 3.62 +/- 0.31, P < 0.015) immunization. Also, disease manifestations were reduced by therapeutic treatment of ribavirin (mean maximal severity score 2.5 +/- 0.2 vs. 3.33 +/- 0.21 in controls, P < 0.005) but less so in comparison with preventive treatment. Analysis of the effects of ribavirin on histopathologic changes in the spinal cord tissue revealed a reduction of mononuclear cell infiltrates, composed of T cells and macrophages/microglia, and the absence of demyelination, which were pronounced in control EAE animals. Beneficial effects of preventive and therapeutic treatment with ribavirin on development of EAE suggest this nucleoside analogue as a useful candidate for therapy in multiple sclerosis.
