    CLEC5A Regulates Japanese Encephalitis Virus-Induced Neuroinflammation and Lethality

    CLEC5A/MDL-1, a member of the myeloid C-type lectin family expressed on macrophages and neutrophils, is critical for dengue virus (DV)-induced hemorrhagic fever and shock syndrome in Stat1−/− mice and ConA-treated wild type mice. However, whether CLEC5A is involved in the pathogenesis of viral encephalitis has not yet been investigated. To investigate the role of CLEC5A to regulate JEV-induced neuroinflammation, antagonistic anti-CLEC5A mAb and CLEC5A-deficient mice were generated. We find that Japanese encephalitis virus (JEV) directly interacts with CLEC5A and induces DAP12 phosphorylation in macrophages. In addition, JEV activates macrophages to secrete proinflammatory cytokines and chemokines, which are dramatically reduced in JEV-infected Clec5a−/− macrophages. Although blockade of CLEC5A cannot inhibit JEV infection of neurons and astrocytes, anti-CLEC5A mAb inhibits JEV-induced proinflammatory cytokine release from microglia and prevents bystander damage to neuronal cells. Moreover, JEV causes blood-brain barrier (BBB) disintegrity and lethality in STAT1-deficient (Stat1−/−) mice, whereas peripheral administration of anti-CLEC5A mAb reduces infiltration of virus-harboring leukocytes into the central nervous system (CNS), restores BBB integrity, attenuates neuroinflammation, and protects mice from JEV-induced lethality. Moreover, all surviving mice develop protective humoral and cellular immunity against JEV infection. These observations demonstrate the critical role of CLEC5A in the pathogenesis of Japanese encephalitis, and identify CLEC5A as a target for the development of new treatments to reduce virus-induced brain damage

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    After Total Knee Arthroplasty, Monitored Active Ankle Pumping Improves Lower Leg Circulation More Than Unmonitored Pumping: A Pilot Study

    [[abstract]]1) Background: deep venous thrombosis (DVT) has long been recognized as the most devastating complication after total knee replacement (TKR). To prevent DVT, intermittent pneumatic compression to improve venous return in the lower leg has been advocated by surgeons. Physical activities such as active ankle pumping and early mobilization have been recommended as auxiliary measures to increase venous return in the lower leg and help in ambulation after TKR. In this study, in order to remind patients to exercise their ankle actively and efficiently after TKR, a foot band with motion sensor and reminder alarm was used. (2) Methods: The patients were randomly allocated into three groups according to the therapeutic protocols. The patients in group 1 conducted active ankle pumping without any reminders, those in group 2 underwent intermittent pneumatic compression, and those in group 3 conducted active ankle pumping with ankle motion sensor/reminder. The parameters of blood flow, namely, peak flow velocity and flow volume, in the bilateral common femoral vein and popliteal vein on the 1st, 3rd, and 14th days after surgery were measured using the echo technique, an index to evaluate the effect on promotion of venous return, among the three groups. (3) Results: The peak flow velocity and flow volume of the operative limb in group 3 (with motion sensor/reminder) were significantly higher than those in other groups. The peak flow velocity and flow volume in the popliteal vein in group 3 increased by 112% and 93.8%, respectively, compared to group 1 on the 14th day. No significant difference in peak flow velocity or flow volume was found in the nonoperative limb between the groups. (4) Conclusions: According to the results, a motion sensor/reminder with vibration alarms can improve the performance of active ankle pumping exercises in improving lower leg circulation, and hence may reduce the risk of DVT

    Efficacy of a Smart Antisnore Pillow in Patients with Obstructive Sleep Apnea Syndrome

    Objective. Untreated obstructive sleep apnea syndrome (OSAS) increases the risk of cardiovascular, dementia, and motor vehicle accident events. However, continuous positive airway pressure (CPAP) which is the gold standard treatment is not acceptable for many patients with OSAS. Development of devices for the patients of nonadherence to CPAP is necessary. Materials and Methods. We evaluated the effect of the smart antisnore pillow (SAP) in patients with OSAS in a prospective, noncontrolled, nonrandomized, pilot study. According to the apnea-hypopnea index (AHI), they were divided into two groups: mild-to-moderate OSAS group and severe OSAS group. Single-night polysomnography (PSG) with application of a SAP was performed. Thirty patients, 15 males and 15 females, 33–82 years old (mean age, 59.3±12.9 years), completed the smart antisnore pillow therapy test. Among them, 23 patients had mild-to-moderate OSAS. Results. The SAP significantly improved the snore number (p=0.018), snore index (p=0.013), oxygen denaturation index (p=0.001), total AHI (p=0.002), and supine AHI (p=0.002) in the mild-to-moderate OSAS group, but there was no significant improvement in the severe OSAS group. Conclusions. We concluded that the SAP is an effective positional therapy device for patients with OSAS of mild-to-moderate severity
