Using Pupil Diameter to Measure Cognitive Load
In this paper, we will present a method for measuring cognitive load and
online real-time feedback using the Tobii Pro 2 eye-tracking glasses. The
system is envisaged to be capable of estimating high cognitive load states and
situations, and adjust human-machine interfaces to the user's needs. The system
is using well-known metrics such as average pupillary size over time. Our
system can provide cognitive load feedback at 17-18 Hz. We will elaborate on

The Genomics of Environmental Adaptation of a Mediterranean Grass
In a rapidly changing climate, plants are facing constant challenges. As sessile organisms, their best strategy to survive and provide their offspring with a fitness advantage is adaptation to local environments. Adaptation is defined as a phenotype that maximizes the organism’s fitness in its local habitat compared to foreign phenotypes. Studying the genetic basis of past changes in plant adaptation can help us understand how plant populations are going to cope with and adapt to future climatic changes. Nevertheless, adaptation is not the only process leading to allele frequency changes in a population. Gene flow, genetic drift and demographic history, can confound with or hinder environmental adaptation, perplexing the study of its genetic basis. Taking advantage of the recent development of whole-genome sequencing techniques and of the theoretical and practical advances in evolutionary genomics, this task becomes yet at reach. In this thesis, we use the Mediterranean grass Brachypodium distachyon, a recently established model species, with a fully-sequenced and annotated genome. Using the species’ extensive diversity panel, we aimed at: i) understanding the molecular basis of its adaptation to different environments, ii) delving into the effect of structured populations on the discovery of adaptive variants using genome-wide studies and iii) investigating the flowering time differences and genetic architecture of this trait on natural populations. By doing so, we also provide the community with genomic resources that close a conspicuous sampling gap in the Balkan region. Our results show that B. distachyon’s genetic clades (populations) occupy different ecological niches, and we suggest that the species recolonized Europe and the Middle East following the last glacial maximum. Furthermore, we confirm that the level of population subdivision influences the detection of candidate variants and that local populations adapted through different strategies to the same environmental pressure. We also demonstrate that natural accessions across the species’ range display large differences regarding flowering time and vernalization requirements, using data of both an outdoor and a greenhouse experiment. Finally, we reveal that eight well-characterized flowering-time genes contribute significantly to flowering time variation and display signs of polygenic selection in the natural populations of B. distachyon. In summary, this thesis establishes B. distachyon as a model for research in ecology and evolution and pave a new avenue of research in the field of population genomics

Analysis of signals related to the generation process of extreme events: towards a unified approach
This thesis was submitted for the award of Doctor of Philosophy and was awarded by Brunel University London.In the last decades, although the scientific community has attempted to explain a series of complex
phenomena, ranging from natural hazards to physical conditions and economic crises, aspects
of their generation process still escape our full understanding. The present thesis intends
to promote our understanding of the spatiotemporal behavior and the generation mechanisms
that govern large and strong earthquakes, employing a broad multidisciplinary perspective for
the interpretation of catastrophic events. Two main questions are debated. The first question
concentrates on “whether the generation process of an extreme event has more than one
facets prior to its final appearance”. In the scientific study of earthquakes, attention is drawn
to the predictive capability and monitoring of different precursory observations. Among them
preseismic electromagnetic emissions have been also observed indicating that the science of
earthquake prediction should be from the start multidisciplinary. Drawing on recently introduced
models for earthquake dynamics, that address issues such as long-range correlations,
self-affinity, complexity-organization and fractal structures, the present work endeavors to further
penetrate on the analysis of preseismic electromagnetic emissions and elucidate their link
with the generation process of large and strong earthquakes. A second question deals with
“whether there is a unified approach for the study of catastrophic events”. This question implies
the possibility for common statistical behavior of diverse extreme events and the potential for
transferability of methods from the study of earthquake dynamics across other fields. On these
grounds, the present work extends the focus of inquiry to the analysis of electroencephalogram
recordings related to epileptic seizures, in the prospect to identify common mechanisms that
may explain the nature and the generation process of both phenomena, and to open up different
directions for future research. Finally, with a view to consider alternative ways of studying
key theoretical principles associated with the generation process of catastrophic phenomena, a
relevant framework based on proposed algorithms is presented, focusing on parameters such as:
the energy of earthquakes, the mean and maximum magnitude of the sample, the probability
that two samples may come from the same population. Such an attempt aims to contribute
to the knowledge of natural phenomena, by extending the existing theory and models and

Σύνθεση παραγώγων β-D-γλυκοπυρανοζυλο πυριμιδινών ως πιθανών αναστολέων της φωσφορυλάσης του γλυκογόνου και φωτομετρικός προσδιορισμός pKa αναστολέων του καταλυτικού κέντρου
Η φωσφορυλάση του γλυκογόνου (GP), ένα από τα πλέον μελετημένα ένζυμα, αποτελεί μοριακό στόχο για την καταπολέμηση του σακχαρώδη διαβήτη τύπου 2, καθώς εμπλέκεται στη διατήρηση των επιπέδων της γλυκόζης στο αίμα. Σύμφωνα με πρόσφατες μελέτες φαίνεται ότι η φωσφορυλάση του γλυκογόνου παίζει σημαντικό ρόλο στην ανάπτυξη συγκεκριμένων τύπων καρκίνου, όπου τα καρκινικά κύτταρα διαφοροποιούνται μεταβολικά εκφράζοντας την εγκεφαλική ισομορφή της GP. Το εργαστήριο μας διαθέτει πολύχρονη εμπειρία στη σύνθεση αναστολέων της φωσφορυλάσης του γλυκογόνου.
Στα πλαίσια της παρούσας πτυχιακής εργασίας πραγματοποιήθηκε η σύνθεση 2-αρυλο-5-αμινοβενζιμιδαζολίων, απαραίτητων πρόδρομων ενώσεων για τη σύνθεση νέων πιθανών αντιδιαβητικών φαρμάκων. Η συνθετική πορεία η οποία αναπτύχθηκε, περιλαμβάνει αντίδραση συμπύκνωσης της ο-φαινυλενοδιαμίνης με αρωματικά καρβοξυλικά οξέα, η οποία ακολουθείται από αντίδραση νίτρωσης και στη συνέχεια αναγωγή της νιτρο- προς αμινο-ομάδα, προκειμένου να παραληφθούν τα επιθυμητά προϊόντα. Συνολικά, συντέθηκαν δώδεκα βιβλιογραφικά γνωστές ενώσεις, οι οποίες απομονώθηκαν και χαρακτηρίστηκαν πλήρως οι φασματοσκοπικές και φυσικές τους ιδιότητες.Glycogen phosphorylase (GP), one of the most well-studied enzymes, is a molecular target for fighting type 2 diabetes mellitus, since it is involved into maintaining normal blood glucose levels. According to recent studies, glycogen phosphorylase also plays an important role in the development of certain types of cancer, whereas cancer cells differentiate expressing the brain isoform of GP. Our laboratory has many years of experience in the synthesis of glycogen phosphorylase inhibitors.
During this diploma thesis, the synthesis of 2-aryl-5-aminobenzimidazoles, important intermediates in the synthesis of new potential antidiabetic drugs, was undertaken. The synthetic process developed, entailed to a condensation reaction of o-phenylenediamine with aromatic carboxylic acids, followed by nitration and finally reduction of the produced nitro- to the corresponding amino- group. In total, twelve bibliographically reported compounds were synthesized, isolated, purified and characterized by their spectroscopic and physical properties
Σύνθεση και Μελέτη Μοριακών Στροφέων, Ιχνηθετών του Καταλυτικού Κέντρου της Φωσφορυλάσης του Γλυκογόνου
Ο τομέας της μοριακής απεικόνισης ασχολείται με την ανίχνευση συγκεκριμένων μοριακών στόχων και την παρακολούθηση βιολογικών διεργασιών σε κύτταρα, ιστούς και ζωτικά όργανα με μη επεμβατικό τρόπο. Η συλλογή μοριακής φύσης πληροφορίας απαιτεί τη χρήση μορίων ιχνηθετών τα οποία παρουσιάζουν υψηλή εκλεκτικότητα για τις περιοχές ενδιαφέροντος. Οι μοριακοί στροφείς είναι μια οικογένεια συνθετικών φθοριζουσών ενώσεων οι οποίες μετά από φωτοδιέγερση, υφίστανται μια ενδομοριακή διαδικασία στρέψης ως αποτέλεσμα μεταφοράς φορτίου. Αυτή η κατηγορία μορίων έχει ως δομικά χαρακτηριστικά την ύπαρξη μίας ηλεκτρονιοδοτικής και μίας ηλεκτρονιοελκτικής ομάδας οι οποίες συνδέονται με ένα π-συζυγιακό σύστημα και έχουν τη δυνατότητα να περιστραφούν η μία σε σχέση με την άλλη. Η φωσφορυλάση του γλυκογόνου (GP) είναι ένα από τα πιο σημαντικά ρυθμιστικά ένζυμα που ελέγχουν την ομοιόσταση της γλυκόζης και το μεταβολισμό του γλυκογόνου γενικότερα και παίζει καταλυτικό ρόλο στην διάσπαση του αποθηκευμένου στον οργανισμό γλυκογόνου, το οποίο αποτελεί τη βασικότερη αποθήκη ενέργειας μετά τον λιπώδη ιστό. Αναστολή της GP αποτελεί έναν πολλά υποσχόμενο τομέα στη θεραπεία του διαβήτη τύπου ΙΙ.
Στα πλαίσια της παρούσας ερευνητικής εργασίας σχεδιάστηκαν και συντέθηκαν δύο νέοι αναστολείς της GP (RotA και RotB) που ανήκουν στην οικογένεια των μοριακών στροφέων. Αναπτύχθηκε συνθετικό πρωτόκολλο και ειδικά για τον RotA προσδιορίστηκε η σταθερά αναστολής και επιβεβαιώθηκε, με κρυσταλλογραφία ακτίνων-Χ του συμπλόκου ενζύμου : αναστολέα, ότι δεσμεύεται στο καταλυτικό κέντρο της GP. Φασματοσκοπικές μελέτες έδειξαν ότι ο νέος αυτός αναστολέας παρουσιάζει ιδιότητες μοριακού στροφέα οι οποίες εκφράζονται με αύξηση της έντασης φθορισμού μετά από σύμπλεξη στο κλειστό καταλυτικό κέντρο του ενζύμου. Κυτταρικά πειράματα έδειξαν ότι ο RotA μπορεί να χρησιμοποιηθεί ως μοριακός ιχνηθέτης της φωσφορυλάσης του γλυκογόνου σε κυτταρικό περιβάλλον. Συνολικά πρόκειται για ένα νέο εργαλείο που μπορεί να επιτρέψει μια εις βάθος μελέτη της δράσης της φωσφορυλάσης του γλυκογόνου in vitro και in vivo.The field of molecular imaging deals with the detection of specific molecular targets and the monitoring of biological processes in cells, tissues and vital organs in a non-invasive manner. Collection of molecular information requires the use of probes which exhibit high selectivity for the regions of interest. Molecular rotors are a family of synthetic fluorescent compounds which undergo an intramolecular twist process as a result of charge transfer after photoexcitation. This class of molecules bears an electron donating and an electron accepting group linked through a π-conjugate system, that have the ability to rotate relative to each other. Glycogen phosphorylase (GP) is one of the most important regulatory enzymes that control glucose homeostasis and glycogen metabolism, playing a catalytic role in the breakdown of glycogen, a major energy source stored in the body. Inhibition of GP is a promising area in the treatment of type II diabetes.
In the context of this research work, two new GP inhibitors (RotA and RotB), which belong to the molecular rotor family, were designed and synthesized. A synthetic protocol was developed and, for RotA, the inhibition constant was determined and the binding to the catalytic center of GP was confirmed by X-ray crystallography of the enzyme : inhibitor complex. Spectroscopic studies have shown that this new inhibitor exhibits molecular rotor properties which are expressed by an increase in fluorescence intensity after clustering in the closed catalytic center of the enzyme. Cellular experiments have shown that RotA can be used as a molecular marker of glycogen phosphorylase in a cellular environment. Overall, this is a novel tool that can allow an in-depth study of glycogen phosphorylase activity both in vitro and in vivo
