    Rapamycin attenuates acute seizure-induced astrocyte injury in mice in vivo

    AbstractAstrocytes have been implicated in epileptogenesis and seizure-induced brain injury. Pathological studies reveal a variety of structural abnormalities in astrocytes, such as vacuolization and astrogliosis. While in vivo imaging methods have demonstrated rapid changes in astrocytes under a variety of physiological and pathological conditions, the acute effects of seizures on astrocyte morphology in vivo and corresponding mechanisms of seizure-induced astrocytic injury have not been documented. In this study, we utilized in vivo two-photon imaging to directly monitor the acute structural effects of kainate-induced seizures on cortical astrocytes. Kainate seizures cause an immediate, but transient, vacuolization of astrocytes, followed over several days by astrogliosis. These effects are prevented by pre- or post-treatment with rapamycin, indicating the mTOR pathway is involved in mediating seizure-induced astrocyte injury. These finding have clinical implications for mechanisms of seizure-induced astrocyte injury and potential therapeutic applications with mTOR inhibitors.</jats:p

    Complex Network Approach to the Statistical Features of the Sunspot Series

    Complex network approaches have been recently developed as an alternative framework to study the statistical features of time-series data. We perform a visibility-graph analysis on both the daily and monthly sunspot series. Based on the data, we propose two ways to construct the network: one is from the original observable measurements and the other is from a negative-inverse-transformed series. The degree distribution of the derived networks for the strong maxima has clear non-Gaussian properties, while the degree distribution for minima is bimodal. The long-term variation of the cycles is reflected by hubs in the network which span relatively large time intervals. Based on standard network structural measures, we propose to characterize the long-term correlations by waiting times between two subsequent events. The persistence range of the solar cycles has been identified over 15\,--\,1000 days by a power-law regime with scaling exponent γ=2.04\gamma = 2.04 of the occurrence time of the two subsequent and successive strong minima. In contrast, a persistent trend is not present in the maximal numbers, although maxima do have significant deviations from an exponential form. Our results suggest some new insights for evaluating existing models.     W-Cr-C-N Nanocomposite Thin-Film Coatings via Reactive Magnetron Sputtering

    While binary tungsten carbide can form smooth, hard films, these suffer from low fracture toughness. Tungsten nitride films are frequently harder, but are more brittle. Chromium nitride has excellent wear and oxidation resistance, but films often form with low hardness. Composites of these binary compounds offer a possibility to tailor the material for a desired combination of properties. To this end, we have used reactive RF-magnetron sputtering with Cr and WC targets to form quaternary composites, with nitrogen as the reactive gas. The coatings were deposited on Si, Ti, and steel substrates. The nitrogen partial pressure was varied to investigate the relationship between the film properties and the deposition conditions. Energy dispersive spectroscopy showed changes in the chemical composition as a result of the change in nitrogen partial pressure. X-ray diffraction illuminated the structure as either a solid solution with a B1 NaCl structure, or a nanocomposite with the average crystallite size under 11 nm. Optical interferometer revealed low compressive stresses.
