Single-molecule force spectroscopy of the add adenine riboswitch relates folding to regulatory mechanism
Riboswitches regulate gene expression via ligand binding to an aptamer domain which induces conformational changes in a regulatory expression platform. By unfolding and refolding single add adenine riboswitch molecules in an optical trap, an integrated picture of the folding was developed and related to the regulatory mechanism. Force-extension curves (FECs) and constant-force folding trajectories measured on the aptamer alone revealed multiple partially-folded states, including several misfolded states not on the native folding pathway. All states were correlated to key structural components and interactions within hierarchical folding pathways. FECs of the full-length riboswitch revealed that the thermodynamically stable conformation switches upon ligand binding from a structure repressing translation to one permitting it. Along with rapid equilibration of the two structures in the absence of adenine, these results support a thermodynamically-controlled regulatory mechanism, in contrast with the kinetic control of the closely-related pbuE adenine riboswitch. Comparison of the folding of these riboswitches revealed many similarities arising from shared structural features but also essential differences related to their different regulatory mechanisms
Imaging Inter-Edge State Scattering Centers in the Quantum Hall Regime
We use an atomic force microscope tip as a local gate to study the scattering
between edge channels in a 2D electron gas in the quantum Hall regime. The
scattering is dominated by individual, microscopic scattering centers, which we
directly image here for the first time. The tip voltage dependence of the
scattering indicates that tunneling occurs through weak links and localized

Scanned Potential Microscopy of Edge and Bulk Currents in the Quantum Hall Regime
Using an atomic force microscope as a local voltmeter, we measure the Hall
voltage profile in a 2D electron gas in the quantum Hall (QH) regime. We
observe a linear profile in the bulk of the sample in the transition regions
between QH plateaus and a distinctly nonlinear profile on the plateaus. In
addition, localized voltage drops are observed at the sample edges in the
transition regions. We interpret these results in terms of theories of edge and
bulk currents in the QH regime.Comment: 4 pages, 5 figure
Scanned Potential Microscopy of Edge States in a Quantum Hall Liquid
Using a low-temperature atomic force microscope as a local voltmeter, we
measure the Hall voltage profile in a quantum Hall conductor in the presence of
a gate-induced non-equilibrium edge state population at n=3. We observe sharp
voltage drops at the sample edges which are suppressed by re-equilibrating the

Pediatric urolithiasis: the current surgical management
Children represent about 1% of all patients with urolithiasis, but 100% of these children are considered high risk for recurrent stone formation, and it is crucial for them to receive a therapy that will render them stone free.
