    The CCA Anticodon Specifies Separate Functions Inside and Outside Translation in \u3cem\u3eBacillus cereus\u3c/em\u3e

    Bacillus cereus 14579 encodes two tRNAs with the CCA anticodon, tRNATrp and tRNAOther. tRNATrp was separately aminoacylated by two enzymes, TrpRS1 and TrpRS2, which share only 34% similarity and display different catalytic capacities and specificities. TrpRS1 was 18-fold more proficient at aminoacylating tRNATrp with Trp, while TrpRS2 more efficiently utilizes the Trp analog 5-hydroxy Trp. tRNAOther was not aminoacylated by either TrpRS but instead by the combined activity of LysRS1 and LysRS2, which recognized sequence elements absent from tRNATrp. Polysomes were found to contain tRNATrp, consistent with its role in translation, but not tRNAOther suggesting a function outside protein synthesis. Regulation of the genes encoding TrpRS1 and TrpRS2 (trpS1 and trpS2) is dependent on riboswitch-mediated recognition of the CCA anticodon, and the role of tRNAOther in this process was investigated. Deletion of tRNAOther led to up to a 50 fold drop in trpS1 expression, which resulted in the loss of differential regulation of the trpS1 and trpS2 genes in stationary phase. These findings reveal that sequence-specific interactions with a tRNA anticodon can be confined to processes outside translation, suggesting a means by which such RNAs may evolve non-coding functions

    Robotic partial nephrectomy for posterior tumors through a retroperitoneal approach offers decreased length of stay compared with the transperitoneal approach: A propensity-matched analysis

    INTRODUCTION: We sought to compare surgical outcomes between transperitoneal and retroperitoneal robotic partial nephrectomy (RPN) for posterior tumors. PATIENTS AND METHODS: Using our multi-institutional RPN database, we reviewed 610 consecutive cases for posterior renal masses treated between 2007 and 2015. Primary outcomes were complications, operative time, length of stay (LOS), surgical margin status, and estimated glomerular filtration rate (eGFR) preservation. Secondary outcomes were estimated blood loss, warm ischemia time (WIT), disease recurrence, and disease-specific mortality. Due to significant differences in treatment year and tumor size between approaches, retroperitoneal cases were matched 1:4 to transperitoneal cases based on propensity scores using the greedy algorithm. Outcomes were compared between approaches using the chi-square and Mann-Whitney U tests. RESULTS: After matching, 296 transperitoneal and 74 retroperitoneal cases were available for analysis, and matched groups were well balanced in terms of treatment year, age, gender, race, American Society of Anesthesiologists physical status classification (ASA) score, body mass index, tumor laterality, tumor size, R.E.N.A.L. (radius, exophytic/endophytic properties, nearness of tumor to the collecting system or sinus, anterior/posterior, location relative to polar lines) score, and hilar location. Compared with transperitoneal, the retroperitoneal approach was associated with significantly shorter mean LOS (2.2 vs 2.6 days, p = 0.01), but longer mean WIT (21 vs 19 minutes, p = 0.01). Intraoperative (p = 0.35) and postoperative complications (p = 0.65), operative time (p = 0.93), positive margins (p = 1.0), and latest eGFR preservation (p = 0.25) were not significantly different between approaches. No differences were detected in the other outcomes. CONCLUSIONS: Among high-volume surgeons, transperitoneal and retroperitoneal RPN achieved similar outcomes for posterior renal masses, although with slight differences in LOS and WIT. Retroperitoneal RPN may be an effective option for the treatment of certain small posterior renal masses

    Genic Variation in White-tailed Deer from Arkansas

    Liver and kidney samples of 33 white-tailed deer (Odocoileus virginianus) representing three populations in Arkansas were examined with horizontal starch gel electrophoresis. Of 17 loci examined, only PGM-1 and ES-2 exhibited polymorphism. Average individual heterozygosity, ranging from 2.3% to 4.7% with a mean of 3.1 %, was much lower than that reported for white-tailed deer in other parts of its range. The three populations examined in this study were highly similar based on Rogers\u27 genetic similarity coefficient

    Aquatic Exercise for Better Living on Land: Impact of Shallow-Water Exercise on Older Japanese Women for Performance of Activities of Daily Living (ADL)

    Twenty-six Japanese women (70.5 yr) self-selected water exercise (WEX) (n=13), or control (CON) (n=13) for 12 weeks. WEX was performed 60-minutes/day, 3 days/week with warm-up, cool-down stretch, ADL exercises, and cardiovascular/muscular endurance in 30°C water at a xiphoid level depth. CON continued their current activity/nutrition patterns. Compared to CON, WEX improved (p\u3c.05) functional fitness and balance measures including arm curl (22%), chair stand (21%), 8-feet up & go (13%), chair sit/reach (50%), and 12-min walk (15%). No significant changes in sway velocity (SV) or limits of stability (LOS) were seen for either group. This shallow water exercise improved land-based ADL for older women but not balance. ADL tasks associated with balance did improve which may have indicated enhanced motor control

    Characterization of Space Shuttle Ascent Debris Aerodynamics Using CFD Methods

    An automated Computational Fluid Dynamics process for determining the aerodynamic Characteristics of debris shedding from the Space Shuttle Launch Vehicle during ascent is presented. This process uses Cartesian fully-coupled, six-degree-of-freedom simulations of isolated debris pieces in a Monte Carlo fashion to produce models for the drag and crossrange behavior over a range of debris shapes and shedding scenarios. A validation of the Cartesian methods against ballistic range data for insulating foam debris shapes at flight conditions, as well as validation of the resulting models, are both contained. These models are integrated with the existing shuttle debris transport analysis software to provide an accurate and efficient engineering tool for analyzing debris sources and their potential for damage

    Adapting a Cryogenic Sapphire Oscillator for Very Long Baseline Interferometry

    Extension of very long baseline interferometry (VLBI) to observing wavelengths shorter than 1.3mm provides exceptional angular resolution (~20 micro arcsec) and access to new spectral regimes for the study of astrophysical phenomena. To maintain phase coherence across a global VLBI array at these wavelengths requires that ultrastable frequency references be used for the heterodyne receivers at all participating telescopes. Hydrogen masers have traditionally been used as VLBI references, but atmospheric turbulence typically limits (sub) millimeter VLBI coherence times to ~1-30 s. Cryogenic Sapphire Oscillators (CSO) have better stability than Hydrogen masers on these time scale and are potential alternatives to masers as VLBI references. Here, We describe the design, implementation and tests of a system to produce a 10 MHz VLBI frequency standard from the microwave (11.2 GHz) output of a CSO. To improve long-term stability of the new reference, the CSO was locked to the timing signal from the Global Positioning System satellites and corrected for the oscillator aging. The long-term performance of the CSO was measured by comparison against a hydrogen maser in the same laboratory. The superb short-term performance, along with the improved long-term performance achieved by conditioning, makes the CSO a suitable reference for VLBI at wavelengths less than 1.3mm.Comment: 24 pages, 15 figure

    Ambivalence in Organizations: A Multilevel Approach

    The experience of simultaneously positive and negative orientations toward a person, goal, task, idea, and such appears to be quite common in organizations, but it is poorly understood. We develop a multilevel perspective on ambivalence in organizations that demonstrates how this phenomenon is integral to certain cognitive and emotional processes and important outcomes. Specifically, we discuss the organizational triggers of ambivalence and the cognitive and emotional mechanisms through which ambivalence diffuses between the individual and collective levels of analysis. We offer an integrative framework of major responses to highly intense ambivalence (avoidance, domination, compromise, and holism) that is applicable to actors at the individual and collective levels. The positive and negative outcomes associated with each response, and the conditions under which each is most effective, are explored. Although ambivalence is uncomfortable for actors, it has the potential to foster growth in the actor as well as highly adaptive and effective behavior

    A Competing Values Framework for Analyzing Presentational Communication in Management Contexts

    Communication specialists have long been interested in analyzing messages. More recently, they have stressed the need for evaluative tools that account for situational ex pectations and constraints. Drawing from the literature on organizational and managerial effectiveness, we constructed an empirical model applicable to presenta tional communication. Over 100 communication professors evaluated the relevance of descriptors for six different types of business presentations: three oral and three writ ten. Their judgments were used to create similarity scores, which were submitted to multidimensional scaling. A three-dimensional model emerged. This "competing values model" illustrates the dynamic interplay between the highly contrasting charac teristics of four general types of presentational communication: relational, information al, instructional, and transformational.     Structural and functional effects of acoustic exposure in goldfish: evidence for tonotopy in the teleost saccule

    <p>Abstract</p> <p>Background</p> <p>Mammalian and avian auditory hair cells display tonotopic mapping of frequency along the length of the cochlea and basilar papilla. It is not known whether the auditory hair cells of fishes possess a similar tonotopic organization in the saccule, which is thought to be the primary auditory receptor in teleosts. To investigate this question, we determined the location of hair cell damage in the saccules of goldfish (<it>Carassius auratus</it>) following exposure to specific frequencies. Subjects were divided into six groups of six fish each (five treatment groups plus control). The treatment groups were each exposed to one of five tones: 100, 400, 800, 2000, and 4000 Hz at 176 dB re 1 μPa root mean squared (RMS) for 48 hours. The saccules of each fish were dissected and labeled with phalloidin in order to visualize hair cell bundles. The hair cell bundles were counted at 19 specific locations in each saccule to determine the extent and location of hair cell damage. In addition to quantification of anatomical injury, hearing tests (using auditory evoked potentials) were performed on each fish immediately following sound exposure. Threshold shifts were calculated by subtracting control thresholds from post-sound exposure thresholds.</p> <p>Results</p> <p>All sound-exposed fish exhibited significant hair cell and hearing loss following sound exposure. The location of hair cell loss varied along the length of the saccule in a graded manner with the frequency of sound exposure, with lower and higher frequencies damaging the more caudal and rostral regions of the saccule, respectively. Similarly, fish exposed to lower frequency tones exhibited greater threshold shifts at lower frequencies, while high-frequency tone exposure led to hearing loss at higher frequencies. In general, both hair cell and hearing loss declined as a function of increasing frequency of exposure tone, and there was a significant linear relationship between hair cell loss and hearing loss.</p> <p>Conclusions</p> <p>The pattern of hair cell loss as a function of exposure tone frequency and saccular rostral-caudal location is similar to the pattern of hearing loss as a function of exposure tone frequency and hearing threshold frequency. This data suggest that the frequency analysis ability of goldfish is at least partially driven by peripheral tonotopy in the saccule.</p
