    1001–24 Local Delivery of Urokinase to Porcine Coronary Arteries Using the Localmed Infusion Sleeve

    Local Delivery of thrombolytic agents may reduce thrombus formation after balloon angioplasty. The Localmed Infusion Sleeve enables localized infusion of urokinase to be performed at the time of balloon angioplasty without the need for catheter exchange.MethodsBalloon angioplasty was performed on 13 coronary arteries of 5 pigs at 4 atmospheres with a balloon to artery ratio of approximately 1.1 to 1. After angioplasty the Infusion Sleeve was advanced over the dilatation balloon and the balloon was reinflated to 2 atmospheres to appose the sleeve to the vessel wall. 50,000 Units (8 cc) of 123l-urokinase was infused through the microperforations in the sleeve for 10 seconds by a computer controlled pump. The coronaries were then excised and counted in a gamma counter.ResultsConclusions(1) The Localmed Infusion Sleeve enables drug infusion to be uncoupled from balloon dilatation. (2) Successful delivery of Urokinase to the vessel wall may be achieved using this device without the need for catheter exchange. (3) Persistence of urokinase within the vessel wall occurs after local delivery

    Pigeons received food for key pecking according to a fixed-ratio schedule, while, at the same time, food also was available for not pecking for a specified time. With a fixed ratio of 60, responding was not affected by not-pecking times of 80 or 40 seconds, and was eliminated completely at 10 seconds. With ratios of 180, pecking stopped with not-pecking times of 80 seconds or less; with ratios of 300, it stopped at 120 seconds or less. Not-responding schedules produced steady-state performance immediately following contact with the schedule. With return to the fixed-ratio schedule alone, response rate sometimes was elevated temporarily. When response-independent food presentations replaced the not-pecking schedule, response rate often was enhanced, and the ratio pattern was lost. Only the highest densities of food delivery eliminated responding, even with a fixed ratio of 300. In general, the effects corresponded to those of punishment, except that contrast had appeared both during and after punishment, and now appeared only after the response elimination procedure was suspended

    Absence of shock-elicited aggression in pigeons

    Two pigeons that attacked a taxidermically prepared target pigeon during a schedule of positive reinforcement for key pecking, and two that did not, were shocked through implanted electrodes in the presence of the target. Shock intensities of 2 and 4 mA, durations of 0.1 and 1.3 sec, and frequencies of 2, 6, 20, and 35 per minute were delivered across 16 sessions with 180 shocks per session. No pigeon attacked the target; one pecked the shockplug on its back. The two pigeons that had not attacked during the positive reinforcement schedules were conditioned to peck the target for food reinforcement before another 16 sessions of shock. No attack was observed in these shock sessions. During subsequent positive reinforcement of key pecking, the target was attacked by the two pigeons that had originally attacked and by one that had not. Absence of shock-elicited attack in these pigeons may be related to the parameters of the experiment or may be yet another instance of the absence of shock-elicited attack in the class Aves. At least under the present conditions, it was not possible to predict the level of attack during electric shock from the level of attack during schedules of positive reinforcement for key pecking

    Positive reinforcement and the elimination of reinforced responses

    Key pecking was maintained on a fixed-interval schedule while either a differential-reinforcement-of-not-responding or a fixed-time schedule was imposed simultaneously. The lower the time parameter of the not-responding schedule, the lower was the response rate. Similar effects occurred with the fixed-time schedule, if the pigeons had experience with reinforcement for not responding. Otherwise the effects were less orderly, to the extent that rate could reach maximum with the lowest-valued fixed-time schedule. The not-responding and the response-independent schedules had similar effects on rate in experienced pigeons only when the time parameter or nominal frequency of food presentation was considered. When considered in terms of obtained frequency of food presentation, reinforcement of not responding produced larger decrements in rate than did the fixed-time schedule

    Lever attacking by rats during free-operant avoidance

    Rats pressed a lever to avoid shock on a free-operant avoidance schedule. Some subjects were also exposed to extinction in which the response-shock contingency was eliminated while the shock-shock contingency remained in effect. A specially constructed lever was used that registered not only presses, but also biting attacks on the lever. Throughout various phases of the study, shocks often elicited lever biting as well as post-shock responding. The results suggested that shock-elicited attacks that are forceful enough to activate the operandum might account for some of the responding that occurs in experiments on free-operant avoidance behavior. In particular, shock-elicited operandum attacking might account for post-shock response bursting during free-operant avoidance and the extreme persistence of responding sometimes noted when shocks are delivered during the extinction of avoidance behavior. To the extent that this is true, these phenomena should not be characterized as operant behavior in interpreting the results of experiments on free operant avoidance

    Elimination of reinforced behavior: intermittent schedules of not-responding

    Pigeons' key pecking resulted in food according to either a variable-ratio or a variable-interval schedule. At the same time, food was available for not pecking for a specified time. The required time of not-pecking was segmented into not-responding units, and these units were followed by food according to a fixed-ratio schedule. Both unit duration and the number required were varied. In general, the shorter the time unit or the smaller the ratio, the lower was response rate. When total required not-responding time was constant, but changes in unit duration and the number required altered how the total was achieved, shorter units produced lower rates. Other conditions involved substitution of food delivered independent of responding for the not-responding schedule. With low and moderate total times to food presentation, the not-responding schedule produced lower rates; with the longest times, the response-independent schedule generated less responding. When considered in terms of relative frequency of food presentation available from a source other than pecking, the not-responding schedule reduced rate more effectively than did the response-independent schedule. Comparisons with other research suggested that food presented dependent on not responding compared favorably with punishment as a procedure for reducing response rate. Transient effects differed. Although punishment temporarily depresses rate when first imposed and temporarily enhances it when first removed, food given for not responding quickly generated steady-state rates

    Reward supports flexible orienting of attention to category information and influences subsequent memory

    Preparatory control of attention facilitates the efficient processing and encoding of an expected stimulus. However, this can occur at the expense of increasing the processing cost of unexpected stimuli. Preparatory control can be influenced by motivational factors, such as the expectation of a reward. Interestingly, expectation of a high reward can increase target processing, as well as reduce the cost associated with reorienting. Using a semantic cueing paradigm, we examined the interaction of reward expectation and cue-validity on semantic judgment performance and subsequent memory. Preparatory attention was assessed with pupillometry. Valid category cueing, was associated with better semantic judgment performance and better subsequent memory compared to invalidly cued items. Higher reward also resulted in larger pre-target pupil diameter, which could be indicative of increased preparatory task engagement or arousal. Critically, higher reward also reduced reorienting cost in both semantic judgment and subsequent memory performance. Our findings suggest that reward expectation can facilitate the effective control of preparatory attention for semantic information, and can support optimal goal-directed behavior based on changing task demands
