    Linking drought indices to impacts in the Liaoning province of China

    Drought is an inherent meteorological characteristic of any given region, but is particularly important in China due to its monsoon climate and the “three ladder” landform system. The Chinese government has constructed large-scale water conservation projects since 1949, and developed drought and water scarcity relief frameworks. However, drought still causes huge impacts on water supply, environment and agriculture. China has, therefore, created specialized agencies for drought management called Flood Control and Drought Relief Headquarters, which include four different levels: state, provincial, municipal and county. The impact datasets they collect provide an effective resource for drought vulnerability assessment, and provide validation options for hydro-meteorological indices used in risk assessment and drought monitoring. In this study, we use the statistical drought impact data collected by the Liaoning province Drought Relief Headquarter and meteorological drought indices (Standardized Precipitation Index, SPI and Standard Precipitation Evaporation Index, SPEI) to explore a potential relationship between drought impacts and these indices. The results show that SPI-24 and SPEI-24 are highly correlated to the populations that have difficulties in obtaining drinking water in four out of the six cities studied. Three impacts related to reservoirs and the availability of drinking water for humans and livestock exhibit strong correlations with SPI and SPEI of different accumulated periods. Results reveal that meteorological indices used for drought monitoring and early warning in China can be effectively linked to drought impacts. Further work is exploring how this information can be used to optimize drought monitoring and risk assessment in the whole Liaoning province and elsewhere in China

    Effects of treatment with Astragalus Membranaceus on function of rat leydig cells

    Background Astragalus membranaceus (AM) is a Chinese traditional herb which has been reported to have broad positive effects on many diseases, including hepatitis, heart disease, diabetes and skin disease. AM can promote cell proliferation, increase the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx), and inhibit apoptosis by regulating the transcription of proto-oncogenes controlling cell death. While AM is included in some commercially available “testosterone boosting supplements”, studies directly testing ability of AM to modulate testosterone production are lacking. In the present study, we examined the effects of AM on Leydig cell function in vitro. Methods Rat Leydig cells were purified and treated with AM at different concentrations (0 μg/mL, 10 μg/mL, 20 μg/mL, 50 μg/mL, 100 μg/mL and 150 μg/mL) and cell counting-8 (CCK-8) assay, Enzyme-linked immunosorbent assay, quantitative real time PCR and analysis of activities of SOD and GPx were done respectively. Results Treatment with 100 μg/mL (P \u3c 0.05) and 150 μg/mL AM (P \u3c 0.01) significantly increased Leydig cell numbers. Treatment with AM (20 μg/mL, 50 μg/mL and 100 μg/mL) significantly increased testosterone production (P \u3c 0.01). In addition, increased Leydig cell SOD and GPx activities were observed in response to 20 μg/mL and 50 μg/mL AM treatment (P \u3c 0.01). Furthermore, expression of Bax mRNA was significantly decreased (P \u3c 0.01), and the ratio of Bcl-2/Bax mRNA was significantly increased in response to 20 μg/mL AM in the culture medium (P \u3c 0.05). Conclusions Results supported a beneficial effect of AM on multiple aspects of rat Leydig cell function in vitro including testosterone production

    Hepatitis B virus–induced imbalance of inflammatory and antiviral signaling by differential phosphorylation of STAT1 in human monocytes

    It is not clear how hepatitis B virus (HBV) modulates host immunity during chronic infection. In addition to the key mediators of inflammatory response in viral infection, monocytes also express a high-level IFN-stimulated gene, CH25H, upon response to IFN-a exerting an antiviral effect. In this study, the mechanism by which HBV manipulates IFN signaling in human monocytes was investigated. We observed that monocytes from chronic hepatitis B patients express lower levels of IFN signaling/stimulated genes and higher levels of inflammatory cytokines compared with healthy donors. HBV induces monocyte production of inflammatory cytokines via TLR2/MyD88/NF-kB signaling and STAT1-Ser727 phosphorylation and inhibits IFN-a–induced stat1, stat2, and ch25h expression through the inhibition of STAT1-Tyr701 phosphorylation and in an IL-10–dependent, partially autocrine manner. Further, we found that enhancement of STAT1 activity with a small molecule (2-NP) rescued HBV-mediated inhibition of IFN signaling and counteracted the induction of inflammatory cytokines. In conclusion, HBV contributes to the monocyte inflammatory response but inhibits their IFN-a/b responsiveness to impair antiviral innate immunity. These effects are mediated via differential phosphorylation of Tyr701 and Ser727 of STAT1

    Linking drought indices to impacts to support drought risk assessment in Liaoning province, China

    Drought is a ubiquitous and recurring hazard that has wide-ranging impacts on society, agriculture and the environment. Drought indices are vital for characterising the nature and severity of drought hazards, and there have been extensive efforts to identify the most suitable drought indices for drought monitoring and risk assessment. However, to date, little effort has been made to explore which index (or indices) best represents drought impacts for various sectors in China. This is a critical knowledge gap, as impacts provide important ground truth information for indices used in monitoring activities. The aim of this study is to explore the link between drought indices and drought impacts, using Liaoning province (northeast China) as a case study due to its history of drought occurrence. To achieve this we use independent, but complementary, methods (correlation and random forest analysis) to identify which indices link best to drought impacts for prefectural-level cities in Liaoning province, using a comprehensive database of reported drought impacts in which impacts are classified into a range of categories. The results show that the standardised precipitation evapotranspiration index with a 6-month accumulation (SPEI6) had a strong correlation with all categories of drought impacts, while the standardised precipitation index with a 12-month accumulation (SPI12) had a weak correlation with drought impacts. Of the impact datasets, “drought-suffering area” and “drought impact area” had a strong relationship with all drought indices in Liaoning province, while “population and number of livestock with difficulty in accessing drinking water” had weak correlations with the indices. The results of this study can support drought planning efforts in the region and provide context for the indices used in drought-monitoring applications, so enabling improved preparedness for drought impacts. The study also demonstrates the potential benefits of routine collection of drought impact information on a local scale

    Gut Microbes Reveal Pseudomonas Medicates Ingestion Preference via Protein Utilization and Cellular Homeostasis Under Feed Domestication in Freshwater Drum, Aplodinotus grunniens

    With strong demand for aquatic products, as well as a rapid decrease in global fishery resources and capture fisheries, domesticating animals to provide more high-quality proteins is meaningful for humans. Freshwater drum (Aplodinotus grunniens) is widely distributed in the wild habitats of North America. However, the research on A. grunniens and the feed domestication with diets composed of artificial compounds remains unclear. In this study, a 4-month feeding domestication experiment was conducted with A. grunniens larvae to evaluate the underlying mechanism and molecular targets responsible for alternations in the ingestion performance. The results indicated that a significant increase in the final body weight was exhibited by the feed domesticated group (DOM, 114.8 g) when compared to the group that did not ingest the feed (WT, 5.3 g) as the latest version we raised From the result, the final body weight exhibited significant increase between unfavorable with the feed (WT, 5.3 g) and feed domesticated group (DOM, 114.8 g). In addition, the enzyme activity of digestive enzymes like amylase, lipase, and trypsin was increased in DOM. Genes related to appetite and perception, such as NPY4R, PYY, and LEPR, were activated in DOM. 16s rRNA gene sequencing analysis revealed that Pseudomonas sp. increased from 58.74% to 89.77% in DOM, which accounts for the dominant upregulated microbial community at the genus level, followed by Plesiomonas. Analogously, Mycobacterium, Methylocystis, and Romboutsia also accounted for the down-regulated microbes in the diversity. Transcriptome and RT-PCR analysis revealed that feed domestication significantly improved protein digestion and absorption, inhibited apoptosis by AGE-RAGE signaling, and activated extracellular matrix remodeling by relaxin signaling. Integrated analysis of the microbiome and host transcriptome revealed that Pseudomonas-mediated ingestion capacity, protein utilization, and cellular homeostasis might be the underlying mechanism under feed domestication. These results indicate Pseudomonas and its key genes relating to food ingestion and digestion could serve as the molecular targets for feed domestication and sustainable development in A. grunniens

    Interactive Genetic Algorithm Oriented toward the Novel Design of Traditional Patterns

    To create alternative complex patterns, a novel design method is introduced in this study based on the error back propagation (BP) neural network user cognitive surrogate model of an interactive genetic algorithm with individual fuzzy interval fitness (IGA-BPFIF). First, the quantitative rules of aesthetic evaluation and the user’s hesitation are used to construct the Gaussian blur tool to form the individual’s fuzzy interval fitness. Then, the user’s cognitive surrogate model based on the BP neural network is constructed, and a new fitness estimation strategy is presented. By measuring the mean squared error, the surrogate model is well managed during the evolution of the population. According to the users’ demands and preferences, the features are extracted for the interactive evolutionary computation. The experiments show that IGA-BPFIF can effectively design innovative patterns matching users’ preferences and can contribute to the heritage of traditional national patterns
