Circadian ritmusos biológiai folyamatok mechanizmusának vizsgálata madár tobozmirigy modellen. = Mechanisms of circadian rhythmic biological processes - studies on avian pineal gland.
Kimutattuk, hogy a csirke tobozmirigyben a filogenetikailag ismertebb ĂłragĂ©nek többsĂ©ge, a Clock, Bmal1, Bmal2, Per1, Per2, Per3, Cry1 Ă©s Cry2 expresszálĂłdik. A Cry1, Cry2, Bmal2 Ă©s a Clock ĂłragĂ©nek mRNS-eit sikerĂĽlt kimutatni 15 napos csirke embriĂłk tobozmirigyeibĹ‘l is, valamint a kikelt állatokban mĂ©rt mRNS ritmushoz fázisban hasonlĂł mintázatot mĂ©rtĂĽnk 18 napos csirke embriĂłkban is. Az ĂłragĂ©nek expressziĂłs mintázatát mind az Ă©jszaka alkalmazott megvilágĂtás, mind az in vitro alkalmazott PACAP kezelĂ©s megváltoztatta. Kimutattuk, hogy a MT Ă©s cAMP koncentráciĂłjának napszakos ritmusa a 16-18. embrionális napokon jelenik meg in vitro körĂĽlmĂ©nyek között is. A ritmikus szekrĂ©ciĂł kialakulása azonban periodikus ingerek nĂ©lkĂĽl kĂ©sik. Igazoltuk, hogy bár a tobozmirigy fĂ©ny-receptorai már a 14. embrionális napon is működĹ‘kĂ©pesek, a fĂ©nyingernek mĂ©g nincs direkt, in vitro a corpus pinealĂ©ra gyakorolt szinkronizálĂł hatása. Ugyanakkor a 18. embrionális naptĂłl kezdve a csirke tobozmirigy már a rendkĂvĂĽl alacsony intenzitásĂş (10 lux) megvilágĂtással is vezĂ©relhetĹ‘. EredmĂ©nyeink szerint mind a PACAP, mind a VIP már a 13. embrionális napon fokozza a cAMP Ă©s a melatonin termelĂ©st, de a ritmus fejlĹ‘dĂ©sĂ©t, illetve annak fázisát nem változtatja meg. Kimutattuk, hogy a tobozmirigy in vitro MT szekrĂ©ciĂłját a környezeti hĹ‘mĂ©rsĂ©klet periodikusan alkalmazott, mindössze három Ăłrás megváltoztatása is mĂłdosĂtja, a hĹ‘mĂ©rsĂ©klet változására adott válasz azonban az állat Ă©letkorának fĂĽggvĂ©nye. | We have demonstrated that most of the known, philogenetically conserved clock genes (Clock, Bmal1, Bmal2, Per1, Per2, Per3, Cry1 and Cry2) are expressed in the chicken pineal gland. Expression of Cry1, Cry2, Bmal2 and Clock mRNA-s were shown also in the pineal glands of chicken embryos. Similar 24-h pattern of clock gene expression were detected in 18 day old chicken embryos and in post hatched chickens (6 weeks old). The expression patterns of clock genes were altered by light exposure at subjective night and also by in vitro PACAP administration. It was shown, that the circadian rhythm of both MT secretion and cAMP efflux appears on the 16-18th embryonic days, even in vitro. Without periodic environmental stimuli, the development of the MT rhythm was delayed. Although the light receptors of the pineal gland were found to be responsive to light from the 14th embryonic day, the illumination did not have a synchronizing effect on the in vitro MT secretion at this age. However, from the 18th day, the pineal MT secretion could be controlled by illumination with even the very low intensity (10 lux). Our results showed that both PACAP and VIP induced an increase in both cAMP and MT secretion already from the 13th day of embryonic life, but none of these drugs influenced the development or the phase of the MT rhythm. It was presented that the in vitro MT secretion could be modified age-dependently by only 3 hour-long daily alterations of environmental temperature
Az óragének és a környezeti fényviszonyok szerepe az N-acetiltranszferáz génexpressziójának szabályozásában csirke retinában = The role of clock-genes and environmental lighting conditions in the regulation of N-acetyltransferase gene expression in the chicken retina
Csirke retina Ă©s tobozmirigy sejtekben a melatonin (MT) szintĂ©zis kulcsenzimĂ©nek, az arilalkilamin N-acetiltranszferáznak gĂ©nje (cAanat) co-expresszálĂłdik az ĂłragĂ©nekkel. Emellett a dopamin (DA) a fĹ‘ katekolamin a retinában, mely neuroregulatorkĂ©nt hat. Tanulmányoztuk: 1. a cBmal1 Ă©s cAanat mRNS expressziĂłk dinamikáját retinális/pineális sejtekben in vivo Ă©s in vitro RT-PCR-ral. A cBmal1 mRNS csĂşcsa a fĂ©nyes fázisban volt Zeitgeber idĹ‘ (ZT) 8 Ă©s 10 között, megelĹ‘zve az a cAanat expressziĂł Ă©jszakai emelkedĂ©sĂ©t ZT 16-17-nál. A retinális/pineális cBmal1 Ă©s cAanat mRNS ritmusainak fázisai arra utalnak, hogy kapcsolat van az expressziĂłk között; 2. locked nucleinsav tartalmĂş cBmal1 antisense oligonukleotidok (AON) hatását a cAanat transzkriptumokra Ă©s az MT termelĂ©sre pinealocita kultĂşrában, melyet szuperfĂşziĂłs rendszerben transzfektáltunk. Az AON, mely vagy az RNáz H-t aktiválta, vagy a transzláciĂłs masinĂ©riához valĂł kötĹ‘dĂ©st blokkolta szignifikánsan csökkentette a cAanat transzkripciĂłját Ă©s az MT szekrĂ©ciĂłját jelezvĂ©n a cBmal1 kulcsszerepĂ©t az indol metabolizmus szabályozásában; 3. kĂ©t fĹ‘ DA-receptor család (D1 Ă©s D2) szerepĂ©t a circadián retinális MT ritmus szabályozásában eye-cup modellen. A D2 agonista Quinpirol szignifikánsan csökkentette az MT szekrĂ©ciĂłját sötĂ©t fázisban, mĂg a D1 agonista SKF hatástalan volt, amely arra utal, hogy a DA gátlĂł hatást fejt ki a retinális MT szekrĂ©ciĂłra (reciprok negativ feed-back), melyet elsĹ‘sorban a D2 receptorok mediálnak. | In chicken retinal and pineal cells gene of arylalkylamine N-acetyltransferase (cAanat), the key enzyme of melatonin (MT) synthesis is co-expressed with clock genes. In addition, dopamine (DA) is the major catecholamine in the retina serving as a neuroregulator. We studied: 1. the temporal profile of cBmal1 and cAanat mRNA expressions in retinal/pineal cells in vivo and in vitro using RT-PCR. cBmal1 mRNA had a peak in the light phase between Zeitgeber time (ZT) 8 and 10, preceding the nocturnal increase in cAanat expression at ZT 16 to 17. The phase of the rhythms of retinal/pineal cBmal1 and cAanat mRNAs suggests a link between their expressions; 2. the effect of cBmal1 antisense oligonucleotides (AONs) containing locked nucleic acid on cAanat transcripts and MT production in cultured pinealocytes transfected in superfusion system. These AONs synthesized for activating RNase H or blocking the binding of the translation machinery were able to reduce significantly cAanat transcription and MT secretion indicating the key role of cBmal1 in the regulation of indole metabolism; 3. the role of the two major DA-receptor families (D1 and D2) in the regulation of circadian retinal MT rhythm on an eye-cup model. Quinpirole (D2 agonist) reduced significantly MT secretion in dark phase, whereas SKF (D1 agonist) was ineffective suggesting that DA exerts an inhibitory effect on retinal MT secretion (reciprocal negative feed-back), which is mediated primarily via D2 receptors
